Chemical Composition and in-vitro antimicrobial screening of leaf essential oil of Plectranthus gerardianus Benth., and isolation of the major constituent of oil

Background: The Lamiaceae (Labiatae) is one of the most diverse and widespread plant families’ in terms of ethno medicine and its medicinal value is based on the volatile oils concentration. This family is important for flavour, fragrance and medicinal properties. Manyplants belonging to this family have indigenous value. Method: The essential oil of Plectranthus gerardianusBenth. (Lamiaceae), was analysed by GC and GC-MS analysis, while the major component was isolated and conformed by NMR spectroscopy. Result: The oil was found to be rich in oxygenated monoterpenes, which contribute around 62% of the total oil. The major components identified were fenchone (22.90%) and carvenone oxide (16.75%), besides other mono and sesquiterpenoids. The in-vitro antimicrobial activity of essential oil was tested against three gram negative bacteria viz. Pasteurellamultocida, Escherichia coli, and Salmonella enterica, two gram positive bacteria viz. Staphylococcus aureus and Bacillus subtilis and two fungi viz. Candida albicans and Aspergillusflavus. The antimicrobial activity of the oil was also compared to the antimicrobial activity of leaf essential oil of another Himalayan plant viz. Nepetacoerulescens. Conclusion: The oil showed in-vitro antimicrobial activity against all the microbial strains and can lessen the ever-growing demand of potentially hazardous antibiotics for treatment.

The genus Dorema (Apiaceae) is represented in the flora of Iran with seven species of which two, D. ammoniacum D. Don. and D. aucheri Boiss. are endemic. Ripe fruits of D. ammoniacum collected just in the deciduous time were subjected to hydrodistillation to yield the essential oil, which was subsequently analyzed by GC and GC-MS. Twenty-nine compounds were identified and quantified, representing 95.1 % of the total oil. (Z)-Ocimenone (22.3 %) and (E)-ocimenone (18.1 %) were the main components of the oil. In vitro antimicrobial activity of the oil was evaluated against seven Gram-positive and Gram-negative bacteria (Bacillus subtilis, Enterococcus faecalis, Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Klebsiella pneumoniae) and three fungi (Candida albicans, Saccharomyces cerevisiae and Aspergillus niger). The results of the antimicrobial assay of the oil by the disc diffusion method and the MIC values indicated that the oil exhibited moderate to high antimicrobial activity, especially against B. subtilis and S. epidermidis with MIC value of 3.75 mg ml-1.

The essential oil obtained by hydrodistillation from the roots of Anthriscus nemorosa (Bieb.) Sprengel (Umbelliferae) was analyzed by GC and GC-MS. Among sixty-two compounds identified (representing 89.0% of the total oil), the main components were: n-nonane (12.1%), n-hexadecanol (6.9%), δ-cadinene (6.4%), β-pinene (6.0%) and germacrene D (5.4%). Furthermore, the antimicrobial activity of the oil was evaluated against the Gram-positive bacteria Staphylococcus epidermidis (ATCC 12228) and Bacillus subtilis (ATCC 6633), the Gramnegative bacterium Escherichia coli (ATCC 25922), and a yeast Candida albicans (ATCC 10259 and ATCC 24433) using the broth microdilution method.

The chemical composition of the hydrodistilled leaf essential oil from Pulicaria stephanocarpa Balf Fil was determined by GC-MS analysis, and its antimicrobial, antioxidant and anticholinesterase (AChE) activities were evaluated. Eighty-three compounds were identified representing 97.2% of the total oil. ( E)-Caryophyllene 13.4%, ( E)-nerolidol 8.5%, caryophyllene oxide 8.5%, α-cadinol 8.2% spathulenol 6.8% and τ-cadinol 4.7%, were the main components. Antimicrobial activity of the oil, evaluated using the disc diffusion and broth dilution methods, demonstrated the highest susceptibility on Gram-positive bacteria and Candida albicans. The free radical scavenging ability of the oil was assessed by the DPPH assay to show antiradical activity with IC50 of 330 μg/mL. Moreover, the oil revealed an AChE inhibitory activity of 47% at a concentration of 200 μg/mL using Ellman's method.

The chemical composition of the essential oils of Achillea holosericea, Achillea taygetea, Achillea fraasii was determined by GC/MS analysis. Among the ninety-five assayed constituents, camphor, borneol and 1,8-cineol were found to be the major components. The in-vitro antimicrobial activity of these essential oils was evaluated against six bacteria indicating that the first is totally inactive, while the other two possess moderate to strong activities mainly against the Gram negative strains. The essential oil of A. fraasii was also active against the tested pathogenic fungi
