Objective: Sarasvata ghrita (SG) is a polyherbal formulation in Ayurvedic Indian medicinal system, in which ghee is the main ingredient used for extraction. Ghee is 100% lipid, thus its regular use is limited, and there is a lack of quality control profile of SG. Thus, the objective of the study is to develop quality control method for standardization of SG and to analyze manufacturing process of SG and an effective method of extraction to extract phytoconstituents from herbs used in SG to overcome the limitation of SG.Methods: SG was processed as per the traditional method, whereas ethanolic extract (EE) and hydroalcoholic extract (HAE) were obtained by the conventional method and lipid-based extract (LE) was prepared by modern extraction method. SG and all extracts were standardized using newly developed high-performance liquid chromatography (LC) with respect to bebeerine, piperine, 6-shogaol, β-asarone, and chebulinic acid. All extracts were analyzed for pesticides, and heavy metal content by LC/mass spectrometry (MS/MS) and inductively coupled plasma/MS, respectively, screened for total polyphenols and flavonoids content, in vitro antioxidant potential, and for assessing its stability over time.Results: The better extraction was observed with maceration extraction using ethanol compared to ayurvedic method and LE method. All extracts were found to have a negligible amount of pesticide and heavy metals and found to be stable for 6 months under accelerated storage condition. Better polyphenols and flavonoid content and in vitro antioxidant potential were resulted in EE.Conclusion: EE showed a better potential in comparison with SG and LE.

The present study was conducted to evaluate the in vitro antioxidant activity of Ficus carica, commonly known as fig. Methanol and ethanol extracts of F. carica leaves were subjected to 2, 2-diphenyl-1-picryhydrazyl (DPPH) free radical scavenging activity assay where ascorbic acid being positive control had an IC50 value of 3.98±0.26 while methanol and ethanol fractions showed an IC50 of 101.76±1.12 and 93.12±1.17 respectively exhibiting their high antioxidant potential. DPPH assay was also performed on high performance liquid chromatography (HPLC) elutions. Most active antioxidant components in ethanol extract were eluted between 17–18 min, and those in methanol were eluted over 14–15 min and upon ultra-high performance liquid chromatography-mass spectrometery (Orbitrap Liquid Chromatography-Mass Spectrometry) were identified to be 13-Docosenamide, (Z)- for ethanol and ficusin for methanol fraction. Thus, it is concluded that these two components are most probable determinants of antioxidant potential of F. carica leaf extracts. © 2021 Friends Science Publishers

Currently, consumers’ demand for sunscreens derived from natural sources that provide photoprotection from ultraviolet (UV) radiation is pushing the cosmetic industry to develop breakthrough formulations of sun protection products by incorporating plant antioxidants as their active ingredients. In this context, the present study was initiated to evaluate the antioxidant and photoprotective properties of the underutilized Hylocereus polyrhizus peel extract (HPPE) using in vitro spectrophotometric techniques. The phytochemical screenings of HPPE conducted via high-performance liquid chromatography (HPLC) and ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) revealed the presence of phenolic acids and flavonoids as the major secondary metabolites in HPPE. The antioxidant potentials evaluated based on 2, 2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical and total antioxidant capacity assays were in the range of 22.16 ± 0.24%–84.67 ± 0.03% with 50% inhibitory concentration (IC50) of 36.39 ± 0.04 μg/mL and 23.76 ± 0.14%–31.87 ± 0.26% (IC50 = 21.93 ± 0.07 μg/mL), respectively. For the photoprotective evaluation, the results showed that HPPE had significantly high absorbance values (3.1–3.6) at 290–320 nm with an exceptional sun protection factor (SPF) value of 35.02 ± 0.39 at 1.00 mg/mL. HPPE also possessed a broad-spectrum shielding power against both UVA and UVB radiations. Hence, in terms of practical implications, our findings would offer an exciting avenue to develop a photoprotective formulation incorporating the ethanolic extract of Hylocereus polyrhizus peels as a synergistic active ingredient for its excellent UV absorption properties and the strong antioxidant activities.
