 Objective: The aim of this study is to determine the relaxation activity of the ethanolic extract of tetanus leaf (EETL) against contracting isolated guinea pig trachea induced by acetylcholine.Methods: Isolated trachea was equilibrated for 45 min until a stable condition in the Creb’s solution with a temperature of 37°C aerated with carbogen gas (O2:CO2=95%:5%). Test of the relaxation effect conducted after guinea pig trachea contracted with maximum acetylcholine concentration (EC80: 2.0449×10–3 mol) and then a cumulative with concentration of EETL (0.5 mg/mL–4 mg/mL) and concentration of atropine sulfate (6.95×10-6 mg/mL–2.08×10–2 mg/mL).Results: The results showed that EETL could lower the contraction smooth muscle of guinea pig trachea induced by acetylcholine. EETL at a concentration of 3.5 mg/mL had the ability to relax the muscle. It was not significantly different compared to that of atropine sulfate 6.95×10-3 mg/ mL (p>0.005).Conclusion: EETL has a relaxation effect on smooth muscle of guinea pig isolated trachea.

Titanus (Leea aequata L.) merupakan tumbuhan dari suku leeaceae yang digunakan dalam pengobatan tradisional di daerah Tanah Karo, Provinsi Sumatera utara, sebagai obat luka dan obat anti tetanus. Salah satu tanda utama dari penyakit tetanus adalah spasme otot atau kejang disebagian atau seluruh tubuh. Penelitian ini bertujuan untuk mengetahui efek antikejang berupa relaksasi dari ekstrak etanol daun titanus terhadap kontraksi ileum marmut terisolasi yang dikontrasikan dengan asetilkolin. Penelitian ini dilakukan secara in vitro menggunakan alat organ bath. Tahapan penelitian adalah persiapan bahan dan pengujian efek relaksasi ileum terpisah. Parameter yang diukur dalam penelitian ini adalah relaksasi otot polos ileum terpisah. Sebelum dilakukan pengujian, ileum marmut terpisah diekuilibrasi selama 45 menit sampai diperoleh kondisi yang stabil didalam larutan tyrode dengan suhu 37o C yang diaerasi dengan gas karbogen (O2:CO2) 95% : 5%. Pengujian efek relaksasi dilakukan setelah ileum marmut dikontraksi dengan asetilkolin, kemudian masing masing ileum diberikan konsentrasi kumulatif ekstrak daun titanus dan atropin sulfat . Hasil pengujian yang diperoleh menunjukkan bahwa pemberian ekstrak etanol daun titanus memiliki efek relaksasi. Ekstrak etanol daun titanus pada konsentrasi 2,5mg/ml (105,4203±2,9151) mempunyai kemampuan yang tidak berbeda dengan atropin sulfat 1 x 10-5 (113,9796±4,5825) dalam menurunkan kontraksi otot polos ileum yang diinduksi oleh asetilkolin 1,889 x 10-4 M (p>0,005). Jadi dapat disimpulkan bahwa ekstrak etanol daun titanus mempunyai efek relaksasi terhadap otot polos ileum terpisah dengan kemampuan yang tidak jauh berbeda dengan atropin sulfat. Titanus (Leeaaequata L.) is a plant from Leeaceae family used in traditional medication in Tanah Karo, North Sumatera as wound and anti-tetanus medicine. One of the main signs of tetanus is muscle spasm or seizure in some or all parts of body. This research aimed to evaluatethe anti-seizure effect of titanus leaves ethanol extractsuch as relaxation isolated ileum of marmot which was contacted with acetylcholine. This research was conducted byin vitro studyusing organ bath instrument. The stages of this research were the material preparation and isolated ileum relaxation effect evaluation. The parameter measured in this research was the isolated ileum smooth muscle relaxation. Before the test, isolated marmot ileum was equilibrated for 45 minutes until the stable condition in tyrode solution was obtained at 37°C temperature which aerated with carbogen gas (O2:CO2) 95%:5%. The relaxation effect test was conducted aftermarmot ileum was contracted with acetylcholine, and then, titanus leaves extract and atropine sulfate were added to each ileumswith cumulative concentration. The result indicated that the titanus leaves ethanol extract had relaxation effect. Titanus Leaves ethanol extract in 2.5 mg/ml concentration (105.4203±2.9151) had the same ability as atropine sulfate 1x10-5 (113.9796±4.5825)in decreasing the ileum smooth muscle contraction induced with acetylcholine 1.889x10-4 M (p>0.005).It could be concluded that titanus leaves ethanol extract hasrelaxation effect to isolated ileum smooth muscle and it was not so different with atropine sulfate.

Histamine produces constriction of tracheal smooth muscle via H1 receptors, but it also decreases tracheal smooth muscle tone via H2 and H3 receptors. In addition, it has already been reported that phenylethylamine is competitive antagonist of histamine N-methyl-transferase (HMT), enzyme responsible for rapid inactivation of histamine. Our results suggest possibility that phenylethylamine as competitive antagonist of histamine N-methyl-transferase leads to potentiation of histamine induced constriction of isolated guinea-pig trachea, which could be consequence of decreased histamine methylation and subsequent histamine inactivation. At the same time, phenylethylamine had no direct effect on basal tone of intact isolated trachea rings, as well as on other mechanisms leading to increased responsiveness of guinea-pig tracheal smooth muscle (acetylcholine, KCl, electro stimulation).

The effect of pretreatment with egg albumin was examined on the beta-adrenoceptors in guinea pig isolated trachea. Befunolol and carteolol acted as partial agonists and their pA2 values were significantly larger than their corresponding pD2 values in tracheae from both untreated guinea pigs and those treated with egg albumin, suggesting that the beta-adrenoceptors contain two different affinity sites. The Scatchard plot of specific [3H]befunolol binding showed two affinity sites of the receptor (high and low affinity sites) in tracheae from both untreated animals and those treated with egg albumin. The pKD values of befunolol for both low and high affinity sites were in agreement with their respective pD2 and pA2 values. The intrinsic activities of befunolol and carteolol and the pD2 values of the test drugs were decreased by the treatment with egg albumin. The treatment with egg albumin also decreased the total amount of the two affinity sites of the receptor without any change in affinity. The present results support the partial blockade of beta-adrenoceptors in asthma proposed by Szentivanyi.

  Objective: The excessive activity of the parasympathetic nervous system in the intestinal smooth muscle is important role in the increased intestinal motility, so antispasmodic medications are needed that can decrease intestinal motility such as atropine. Treatment may also use medicinal plants that are cheaper and easy to obtain, and also believed to have smaller side effects than modern antispasmodic drugs. This study aimed to determine the anticonvulsant or relaxation effects of the ethyl acetate fraction of titanus leaf (EAFTL) against contracted guinea pig colon.Methods: The parameters measured are smooth muscle relaxation. Before testing, guinea pig colon was equilibrated for 45 min to obtain a stable condition in Tyrode’s solution with a temperature of 37°C aerated with carbogen gas (O2:CO2) with a ratio of 95%:5%. The relaxing effect of the colon was tested after inducing by acetylcholine chloride; then, each colon was given cumulative concentration of EAFTL and atropine sulfate. The concentration of acetylcholine chloride required to increase the contraction of the guinea pig colon was 1.76×10−4 Mol. The cumulative concentration of EAFTL given was 0.5–4 mg/Ml and cumulative concentration of atropine sulfate given was 6.95×10−6–2.08×10−2 mg/Ml.Results: The EAFTL has a relaxing effect. Statistical analysis of EAFTL at a concentration of 4 mg/Ml (100.000±1.7417) in reducing the smooth muscle of colon contraction induced by acetylcholine chloride 1.76×10−4 Mol (p>0.005), has not statistically significant compared to that of atropine sulfate 6.95×10−3mg/Ml (105.7292±0.8161).Conclusion: EAFTL has relaxing effect on the smooth muscle of the colon.

Objective: The study was aimed to investigate the relaxation effect of ethanolic extract of Averrhoa bilimbi L. leaves (EEABL) and ondansetron against 5-HT-induced contraction of the 5-HT3 receptor.Methods: The study of the relaxation effect of the cumulative concentration EEABL leaves (0.5–4 mg/mL) and ondansetron (10−5–3×10−2) after the contracted with 5-HT (EC80: 1.158×10−6 M) was conducted in vitro using isolated rat’s ileum organ in the Tyrode solution.Results: The EEABL and ondansetron decreased ileum contraction after induced by 5- HT. EEABL has no difference in terms of ability as ondansetron in reducing the ileum smooth muscle contraction induced by 5-HT (p>0.05).Conclusion: The EEABL has relaxation effect on smooth muscle of rat’s isolated ileum which has induced by 5-HT.
