AbstractFor the first time, DNA sequence data were obtained for three species of Trichostrongylus from Russia. Internal transcribed spacer (ITS-2) of ribosomal DNA was sequenced for T. axei, T. colubriformis and T. probolurus from sheep from the Moscow region. ITS-2 rDNA length was estimated as 238 nucleotides for T. colubriformis and T. probolurus and 237 nucleotides for T. axei. The G+C content of the ITS-2 sequences of T. colubriformis, T. axei and T. probolurus were 31 %, 32 % and 34 % respectively. The level of interspecific differences in ITS-2 of rDNA of T. axei, T. probolurus and T. colubriformis ranged from 3 to 4 %. The ITS-2 sequences from the Russian specimens were compared with those of T. axei, T. probolurus and T. colubriformis from Australia and Germany. Intraspecific variation ranged from 0 % in T. colubriformis to 3.0 % in T. axei.

Lichens consist of a fungal component that associates with either a green alga or cynobacterium, resulting in diverse morphologies. Resynthesis experiments of isolated myco- and photobionts, as well as internal transcribed spacer (ITS) rDNA phylogenies, suggest that the notoriously dissimilar photomorphs of Lobaria fendleri (Tuck. ex Mont.) Lindau and Lobaria amplissima (Scop.) Forss. both have a single mycobiont, which can simultaneously lichenize two unrelated photobionts, a eukaryote and a prokaryote. In the phylogenetic analyses, the genera Sticta and Durietzia appear monophyletic; whereas Lobaria and Pseudocyphellaria are both nonmonophyletic. Furthermore, the phylogenetic relationships of Durietzia, Lobarina, and Sticta relative to Lobaria and Pseudocyphellaria are in need of re-evaluation.Key words: internal transcribed spacer (ITS), photosymbiodeme, phylogeny, lichen-forming fungi, Lobaria amplissima, Lobaria fendleri, Pseudocyphellaria, Sticta.

The genus Chrysoporthe includes important pathogens of plantation-grown Eucalyptus spp. and has been reported from several tree genera in the order Myrtales in tropical and subtropical areas of the world. During disease surveys in South China, fruiting structures typical of Chrysoporthe spp. were observed on cankers on Eucalyptus spp. and Syzygium cumini trees. The aim of this study was to confirm the identity of the Chrysoporthe cubensis infecting the Eucalyptus spp. and S. cumini and to test the pathogenicity of the fungus. Following glasshouse trials to select virulent isolates, field inoculations were undertaken to screen different commercial Eucalyptus genotypes for their susceptibility to the fungus. Isolates were characterized based on their morphology and DNA sequence data for the β-tubulin and internal transcribed spacer regions of the ribosomal DNA. Results showed that the putative pathogen represented the Asian form of C. cubensis, which occurred on numerous different Eucalyptus spp. and hybrid clones as well as S. cumini. Field inoculations showed that all six of the Eucalyptus genotypes tested are susceptible to infection by C. cubensis. Significant differences were observed between them, providing prospects to select disease-tolerant planting stock in the future.

Crinum latifolium and C. asiaticum are the two species of genus Crinum, Amaryllidaceae family and they have highly medicinal values. Distinction between two species is challenging due to many similar morphological characteristics. In this study, we used molecular makers to distinguish the two species Crinum latifolium and C. asiaticum. The phylogenetic tree was constructed based on the DNA sequence data of the ITS (internal transcribed spacer) nuclear ribosomal DNA (nrDNA) sequences of Crinum latifolium and C. asiaticum. As a result, C. latifolium and C. asiaticum were sorted into two different groups in the phylogenetic tree. Therefore, C. latifolium and C. asiaticum were carried out to solve the taxonomic ambiguity.

Collections of eleven Prunus specimens infected with Polystigma species from Belarus and Russia yielded two existing taxa: Polystigma fulvum (sexual morph) and Polystigma rubrum (asexual morph). DNA based phylogenies of large subunit nuclear rDNA (LSU) and nuclear ribosomal internal transcribed spacer (ITS) are provided for the first time for Polystigma fulvum and its placement is confirmed within Polystigmataceae. The concatenated LSU and ITS DNA sequence data for P. rubrum, analyzed to infer its potential relationship to other Polystigma species, also show that strains of P. rubrum are segregated into two subclades with sufficient genetic differences. No significant differences in morphology or morphometry among the strains of P. rubrum between the two subclades, especially vis-à-vis the conidiogenous cell and conidium sizes are observed (P>0.05). Subspecies concept of P. rubrum complex is discussed.

Abstract Objective Visualisation methods, primarily color-coded representation of sequence data, have been a predominant means of representation of DNA data. Algorithmic conversion of DNA sequence data to sound—sonification—represents an alternative means of representation that uses a different range of human sensory perception. We propose that sonification has value for public engagement with DNA sequence information because it has potential to be entertaining as well as informative. We conduct preliminary work to explore the potential of DNA sequence sonification in public engagement with bioinformatics. We apply a simple sonification technique for DNA, in which each DNA base is represented by a specific note. Additionally, a beat may be added to indicate codon boundaries or for musical effect. We report a brief analysis from public engagement events we conducted that featured this method of sonification. Results We report on use of DNA sequence sonification at two public events. Sonification has potential in public engagement with bioinformatics, both as a means of data representation and as a means to attract audience to a drop-in stand. We also discuss further directions for research on integration of sonification into bioinformatics public engagement and education.
