Abstract The main goal of this study was to evaluate the impact of the invasive common milkweed (Asclepias syriaca L.) on soil nematode communities. The research was carried out in 2018 and 2019 in an ecosystem of permanent grassland in the basin of the Laborec River in land registries of Drahňov, a Vojany village in southeastern Slovakia. The ecosystem contained a total of 64 species of free-living and parasitic nematodes. The most prevalent trophic groups were bacterial feeders (Acrobeloides nanus), followed by plant parasites (Helicotylenchus digonicus and Pratylenchus pratensis), fungal feeders (Aphelenchus avenae), and omnivores (Eudorylaimus carteri). The number of nematode species, the composition of trophic groups and the structure of communities in areas with invasive plants were similar to those in areas with native vegetation during the two years of observation.

Pepper (Piper nigrum L.) is a high economic value plant species that brings the main income to the people at Loc Hung commune. So the pepper’s yield plays a significant role in people’s life in this region. To assess the influence of environmental factors on the growth and development of pepper, we need to analyse the structural characteristics of soil nematode communities in order to promptly detect the levels of parasitic nematodes infection as well as assessment of soil environment status based on nematode communities. Study results at five sampling stations are analyses of 30 genera of nematodes belonging to 6 major trophic groups (bacterial feeders, fungal feeders, algal feeders, omnivores, carnivores and plant parasites). Calculation results of indices such as SUM(MI), SUM(MI2-5), MI and PPI showed that soil nematodes community is very little affected by the bacteria feeder group and the rate of parasitic nematode pepper quite severe. Analysis of correlation between trophic groups showed that nematodes are sensitive to environmental factors. This means the potential use of nematodes as a biological indicator for soil quality is possible. Hồ tiêu (Piper nigrum L.) là cây trồng có giá trị kinh tế cao và là nguồn thu nhập chính của người dân tại xã Lộc Hưng. Do đó, năng suất cây tiêu có ý nghĩa rất lớn đối với đời sống người dân trong vùng. Để đánh giá ảnh hưởng của các yếu tố môi trường lên sự sinh trưởng và phát triển của cây tiêu, cần phải phân tích đặc trưng cấu trúc quần xã tuyến trùng đất của vùng nhằm phát hiện kịp thời mức độ nhiễm tuyến trùng ký sinh trên hồ tiêu cũng như đánh giá nhanh hiện trạng môi trường đất dựa trên quần xã tuyến trùng. Kết quả nghiên cứu tại 5 điểm thu mẫu phân tích được 30 giống tuyến trùng thuộc 6 nhóm dinh dưỡng chính (ăn vi khuẩn, ăn nấm, ăn tảo, ăn tạp, ăn thịt và ký sinh thực vật). Kết quả tính toán các chỉ số ∑MI, ∑MI2-5, MI và PPI cho thấy quần xã tuyến trùng đất ở đây rất ít chịu ảnh hưởng của nhóm ăn vi khuẩn và tỷ lệ hồ tiêu nhiễm ký sinh khá nặng. Phân tích tương quan giữa các nhóm dinh dưỡng cho thấy tuyến trùng khá nhạy cảm với các yếu tố môi trường, điều này cho thấy tiềm năng sử dụng tuyến trùng như một sinh vật chỉ thị cho chất lượng môi trường đất.

Soil nematodes play an important role in indication for assessing soil environments and ecosystems. Previous studies of nematode community analyses based on molecular identification have shown to be useful for assessing soil environments. Here we applied PCR-DGGE method for molecular analysisoffive soil nematode communities (designed as S1 to S5) collected from four provinces in Southeastern Vietnam (Binh Duong, Ba Ria Vung Tau, Binh Phuoc and Dong Nai) based on SSU gene. By sequencing DNA bands derived from S5 community sample, our data show 15 species containing soil nematode, other nematode and non-nematode (fungi) species. Genus Meloidogyne was found as abundant one. The genetic relationship of soil nematode species in S5 community were determined by Maximum Likelihood tree re-construction based on SSU gene. This molecular approach is applied for the first time in Vietnam for identification of soil nematode communities. Tuyến trùng đất đóng vai trò chỉ thị quan trọng trong công tác đánh giá môi trường và hệ sinh thái đất. Các nghiên cứu trước đây đã cho thấy lợi ích của việc phân tích cộng đồng tuyến trùng đất bằng định danh sinh học phân tử đối với việc đánh giá môi trường đất. Ở đây, chúng tôi ứng dụng phương pháp PCR-DGGE dựa trên gene SSU để phân tích năm (ký hiệu từ S1 đến S5) cộng đồng tuyến trùng đất thuộc các vùng trồng chuyên canh cây hồ tiêu ở miền nam Việt Nam (Bình Dương, Bà Rịa Vũng Tàu, Bình Phước và Đồng Nai). Bằng cách giải trình tự các vạch của mẫu tuyến trùng S5, kết quả cho thấy cộng đồng tuyến trùng này có 15 loài gồm nhóm tuyến trùng đất, nhóm các loại tuyến trùng khác và nhóm không phải tuyến trùng (nấm) và trong đó Meloidogyne là giống ưu thế. Mối quan hệ di truyền của các các loài tuyến trùng đất thuộc cộng đồng S5 được xác định bằng việc thiết lập cây phát sinh loài Maximum Likelihood dựa trên gene SSU. Đây là nghiên cứu đầu tiên ở Việt Nam sử dụng kỹ thuật PCR-DGGE để phân tích các cộng đồng tuyến trùng đất trồng hồ tiêu.

Nematodes are ideal biological indicators to monitor soil biodiversity and ecosystem functioning. For this reason, they have been receiving increasing attention from a broad range of scientists. The main method to characterize soil nematode communities until at least genus level is still based on microscopic observations of nematode morphology. Such an approach is time-consuming, labor-intensive, and requires specialized personnel. The first studies on the potential use of DNA-metabarcoding to characterize nematode communities showed some shortcomings: under- or overestimation of species richness caused by failure to detect a number of nematode species or caused by intraspecific sequence variants increasing the number of OTUs (operational taxonomic units) or ‘molecular’ species, and flaws in quantification. We set up experiments to optimize this metabarcoding approach. Our results provided new insights such as the drastic effect of different DNA-extraction methods on nematode species richness due to variation in lysis efficacy. Our newly designed primer set (18S rRNA gene, V4-V5 region) showed in silico an improved taxonomic coverage compared with a published primer set (18S rRNA gene, V6-V8 region). However, results of DNA-metabarcoding with the new primer set showed less taxonomic coverage, and more non-nematode reads. Thus, the new primer set might be more suitable for whole soil faunal analysis. Species-specific correction factors calculated from a mock community with equal amounts of different nematode species were applied on another mock community with different amounts of the same nematode species and on a biological sample spiked with four selected nematode species. Results showed an improved molecular quantification. In conclusion, DNA-metabarcoding of soil nematode communities is useful for monitoring shifts in nematode composition but the technique still needs further optimization to enhance its precision.

This study evaluated the effect of a wildfire on soil nematode communities in the spruce forest ecosystem of the High Tatra Mountains 8 years after the fire. Two plots were established, an intact control plot (REF) and a fire-damaged plot after a windstorm (FIR). A total of 20 representative soil samples were collected; 10 from each site (five in June and five in October). A total of 64 species of nematodes were recorded. The mean number of species 8 years after the fire ranged from 24 to 36 and was always significantly higher in REF than in FIR (P < 0.05); with a significantly higher Shannon–Weaver diversity index in REF. Bacterivores were the most abundant trophic group at both sites. Omnivores were more abundant in FIR than in REF, and predators were more abundant in REF. The most abundant species at both sites were Acrobeloides nanus, Aglenchus agricola, Malenchus exiguus and Paratylenchus microdorus. Epidermal or root-hair feeders, mainly Malenchus exiguus, were the most abundant plant-parasitic nematodes at both sites, followed by the ectoparasitic P. microdorus. The migratory endoparasite, Pratylenchus pratensis, was found only in FIR in relatively high numbers, whereas the ectoparasitic species Criconemoides morgensis, Crossonema menzeli and Ogma cobbi occupied only REF. Nematode abundance and species diversity index differed significantly between the disturbed and undisturbed habitats, but ecological and functional indices and metabolic footprints indicate that the soil ecosystem 8 years after the fire had recovered, with moderate levels of nutrient enrichment comparable to those of the control plot.

A new species of Mesocriconema and a unique assemblage of plant-parasitic nematodes was discovered in a heath bald atop Brushy Mountain in Great Smoky Mountains National Park. Mesocriconema ericaceum n. sp., a species with males, superficially resembles M. xenoplax. DNA barcoding with the mitochondrial COI gene provided evidence of the new species as a distinct lineage. SEM revealed significant variability in arrangement of labial submedian lobes, plates, and anterior and posterior annuli. Three other nematodes in the family Criconematidae were characterised from the heath bald. Ogma seymouri, when analysed by statistical parsimony, established connections with isolates from north-eastern Atlantic coastal and north-western Pacific coastal wet forests. Criconema loofi has a southern Gulf Coast distribution associated with boggy soils. Criconema cf. acriculum is known from northern coastal forests of California. Understanding linkages between these species and their distribution may lead to the broader development of a terrestrial soil nematode biogeography.
