Effects of Bacterocin from MRSA and Nigella Sativa (seed oil) against Biofilm from MRSA

A many risk challenge in (settings hospital) are multi- bacteria are antibiotic-resistant. Some type strains that ability adhesion surface-attached bio-film census. Fifteen MRSA isolates were considered as high biofilm producers Moreover all MRSA isolates; M3, M5, M7 and M11 produced biofilms but the thickest biofilm seen M7strain. The MIC values of N. sativa oil against clinical isolates of MRSA were between (0.25, 0.5, 0.75, 1.0) μg/ml While MRSAcin (50, 75, 100, 125) µg\ ml. All biofilms treated with MRSAcin and Nigella sativa developed a presence of live cells after cultured on plate agar with inhibition zone between MIC (18 – 15) and (14- 11)mm respectively.Yet, results showed that MRSA supernatant developed a inhibitory effect then habitat oil . Significant differences (P less than 0.05) were found in O.D. and viable count between pre and post treatment of M7 strain biofilms. Unlike the Nigella oil treatment which left live some bacterial cells, MRSA supernatant (MRSAcin) left no live cells.

Objective: The effectiveness of a phytobiotic based on active metabolites of Nigella sativa L. against typical microorganisms of opportunistic microbiocenosis of the mucous membranes of highly productive cows was studied. Initially, the inhibitory activity of the phytobiotic was studied on cultures of wild multi-antibiotic-resistant isolates of P. aeruginosa and S. aureus isolated on a commercial dairy farm. It was found that the phytobiotic had the ability to inhibit the growth of isolates on the nutrient medium, but the severity of the inhibitory effect varied notably. At the second stage, an experiment was conducted with the local application of phytobiotics on cows that had inflammatory complications of the postpartum period. The results of the experiment showed a pronounced inhibitory effect of the phytobiotic on S. aureus, Str. uberis, P. aeruginosa, E. cloacae, C. albicans, and P. mirabilis. There was an involution of inflammatory symptoms and normalization of the clinical and microbiological state of the mucous membrane after the use of a phytobiotic preparation.

The aim of the present study was to determine the proper concentrations of whey protein isolate (WPI), Arabic gum (AG), and Zedo gum (ZG) for the encapsulation of Nigella sativa seed oil (NSO). WPI, AG, and ZG were used as wall materials for the encapsulation of NSO by mixture design and then the optimum sample with minimum Z-average, PDI, and a* and maximum zeta potential, conductivity, encapsulation efficiency, and b* was selected for examining FT-IR and SEM. The findings demonstrated that lower particle size and particle dispersion index were obtained at higher amounts of AG and WPI with the least a*. Desirability function results indicate that the mixture of 39.05% WPI, 16.758% AG, and 4.192% ZG led to the production of the optimum sample. Microencapsulation of Nigella sativa seed oil using the optimum wall composition was recognized to be useful to preserve bioactive compounds and provide flavor stability, enabling the use of microcapsules in food formulations.

Nigella sativa seed has a lot of beneficial health effect, such as immune-modulator, anti-histamine, anti-diabetic, anti-hypertension, anti-inflammatory, anti-microbes and antitumor. Numeral antimicrobial effect of Nigella sativa has been published, but only a few number of the information of the antimicrobial activity to the probiotic bacteria such as from the genus of Lactobacillus spp. However this research focused on the selectivity of the antimicrobial activity of N. sativa extract to the probiotic and pathogenic bacteria. This research begins with the extraction of N. sativa seed powder by ethanol 80% 1:3 (w/v). With rotary evaporator the macerates extracted till gained semi solid extract. In the other side probiotic and pathogenic bacteria broth cultures were prepared. Extract resoluted with concentration 5%; 2,5%; 1,25% and 0,625%. Into the extract were submerged some disk paper with 6 mm diameter. After that the disk put on to the solid medium were had cultured by bacteria then incubated overnight. The diameters of the clearing zone that seemed then measured. The result showing that the inhibitory zone occurred at 0,625% concentration. After general analysis there is no significant different inhibitory effect of the extract to the tested bacteria, but from the statistical analysis known that probiotik bacteria seemed more sensitive than pathogenic bacteria were tested.
