In present study, the effect of Nigella sativa seeds extract on the chicken peripheral blood mononuclear cells (PBMCs) was investigated. The chicken PBMCs were stimulated with two different doses (125?g/ml; 250?g/ml) of Nigella sativa seeds extract and the cells were harvested at different time points till 48h post-stimulation for analysis of iNOS gene expression by quantitative PCR and nitric oxide (NO) estimation at 24 and 48h post-stimulation. High dose (250?g/ml) of Nigella sativa seed extract showed remarkable induction of iNOS transcripts expression and NO production at 48h post-stimulation, which were 12.8040±1.03347 folds and 5.7089±.64535µM respectively. The results indicated the immunostimulatory potential of the Nigella sativa seed extract on the chicken PBMCs.

Background: Dental infections were caused by the bacterium overgrowth on the surface of the tooth, and treatment should always be set up to prevent this development.Antibiotics have long been used as a conventional antibacterial medication, but their overuse has resulted in microbes gaining resistance to many of the antibiotics, trying to make many commercialized therapeutic remedies ineffectual and resulting to infection recurrence. In this regard we aim to analyze the antibacterial activity of nigella sativa seeds’ and fenugreek seed extract rinses against S. mutans' colonies. Methodology: It was a preclinical experimental study conducted at Baqai Medical and Dental College Karachi., from January - June 2021. Calculated sample size was n = 80 Consecutive sampling technique was used. Plaque of study participants was collected on sterile strips that was transported to laboratory for culture in sterile containers.The extract of Fenugreek seed and nigella sativa seed was kept in an airtight bottle and stored in a refrigerator till usage. The extracts were diluted in distilled water in 1:4 (Extract: Distilled water). Study participants were instructed to not brush their teeth before sampling. Study participants were divided into four groups (negative control, positive control, fenugreek seed extract group and Nigella sativa group) each group had 20 participants. Diluted Fenugreek seed extract and Nigella sativa extract was given to experimental groups for rinses. Results: There was significant decrease in number of colonies in positive controls, fenugreek seed extract group and nigella sativa extract group and there was no change in number of colonies in control group. Furthermore, the analysis showed significant (p-value = 0.001) difference among the groups followed by post hoc analysis. Post hoc analysis showed no difference between positive control, fenugreek seed extract group and nigella sativa group. Conclusion: Fenugreek seed extract and Nigella Sativa seed extract showed comparable antibacterial properties. Also, the effect was found to be similar to commercially available mouth rinse.

The emergence of antibiotic-resistant bacteria is a major public health problem. Consequently, the development of new antimicrobials that act on novel bacterial targets and are less susceptible to resistance are of primary importance to researchers in academia and industry alike. The present study was aimed to determine the antimicrobial and antibiofilm activities of Nigella sativa seed extracts. For this, primary (qualitative) and quantitative antibacterial activities of Nigella sativa seed extracts were determined against ten human pathogenic bacteria including four biofilm producing bacterial strains Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Staphylococcus aureus. The antibacterial activities as well as minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the extracts were evaluated using disc diffusion and macro-broth dilution methods, respectively. The seed extracts of N. sativa exhibited the highest extract yields (w/v) 23.3% and 21.35% with ethanol and petroleum ether, respectively. The crude extract (essential oil) of petroleum ether exhibited superior antibacterial activity (36 mm in diameter zone of inhibition) against biofilm forming E. coli on a disc diffusion assay at a concentration of 1000 μg/disc when compared to that of ethanolic crude extract. Moreover, the lowest MIC and MBC values were determined as 500 μg/ml and 1500 μg/ml respectively with the same petroleum ether extract against E. coli. However, N. sativa essential oil obtained from petroleum ether extract of seed inhibited 94% biofilm formation of E. coli at 2×MIC concentration. Moreover, at the same concentration (2×MIC), the ethanol extract inhibited 56% biofilm formation of P. aeruginosa. These results consistently revealed that it is possible to isolate novel antimicrobial agents capable of completely eradicating microbial infections including antibiotic resistance. Therefore, bioactive natural products present in plant resources would represent a noteworthy alternative to commercial antibiotics helping treatment of human antibiotic resistant infections. In Siddha medicine system has been classified into Aga marunthu (Internal) and Pura marunthu (External) having 32 forms of medicines in each. The external remedies are very useful for topical ulcers, skin diseases like eczema, psoriasis bacterial and fungal infections. The purpose this study was Evaluate the efficacy of Antibacterial activity of aqueous extract of Nigella sativa seeds powder against Staphylococcus aureus by used disk diffusion method. The result has been evaluated as the diameter of the zone of inhibition of microbial growth showed that the different concentration (100%, 50%, 25%) of Nigella sativa seed powder’s aqueous extract more effective against Staphylococcus aureus. Mean value of the hot extract 100%, 50%, 25% respectively 12.19±1.91mm, 7.63±2.32 mm and 3.78±1.73mm and the mean value of Cold extract were 9.45±1.32mm, 4.21±0.92mm and 2.71±.64mm. Standard maintained 18mm of zone of inhibition .Among the hot and cold extract, 100% of hot extract was showed more significant affect against Staphylococcus aureus which was 12.19mm. Finally this study was concluded that the Nigella seeds powder can be used as an antibacterial agent.

Introduction: Cumin (Nigella Sativa) seed oil extract has some ingredients which have antimicrobial effects. The essential oils present in cumin act as antimicrobial agent and it influence on different type of Gram-negative and Gram-positive bacteria and also viruses, parasites and fungi. This study aimed to investigate the antimicrobial properties of cumin extract in disinfecting dentistry surfaces. Material and Methods: This study was performed experimentally and had three groups of cumin extract, Deconex and control group. For each of these groups, 12 culture media were prepared and we counted the colonies created in 24 hours and 48 hours and significance level was assessed using SPSS software and t-test.    Results: At 24 hours, there was a significant difference between the bacterial colony counts of the petri dishes from Cumin Seed (Nigella Sativa) Oil Extract at 5.83 and the Deconex at 0. And at 48 hours, there was also a significant difference since the bacterial colony count on the petri dishes with Cumin (Nigella Sativa) Oil Extract was too many to count and a 0.83 bacterial colony count for the petri dishes with the Deconex. Conclusion: The Cumin (Nigella Sativa) seed oil extract is not suitable to use as an alternative disinfectant of dental surfaces lonely. But some of its ingredients such as thymoquinone and hydroquinone can be used to produce a disinfecting solution.
