Hibiscus cannabinus or commonly known as kenaf is a versatile plant that serves as resources for numerous manufacturing and livestock industries. Originally planted in West Africa, kenaf is now distributed in many countries including Malaysia as its fibres were proved to be an ultimate alternative resource for major industries such as automotive, paper and bio-composite. In fact, in Malaysia, due to its adaptation to wide range of climatic conditions, kenaf has potentially be chosen as a new industrial crop replacing tobacco. There have been many interests on regenerating kenaf via micropropagation as the demand for this crop has been increasing tremendously since the past decades. Hence, this study is initiated with the objective to establish in vitro propagation system of H. cannabinus. The callus induction was achieved on Murashige and Skoog (MS) media supplemented with different concentrations of benzylaminopurine (BAP). It was observed that calli were successfully induced on all the BAP concentrations tested. The optimum concentration of BAP that induced the healthiest and biggest calli was 3.0 mg/l. Shoot and root induction from the calli were attempted using MS medium supplemented with different combinations and concentrations of IBA, BA and GA3. From the seven treatments, three treatments successfully induced formation of shoot; treatment T3 (MS + 1.0 mg/l IBA + 2.5 mg/l BA), treatment T5 (MS + 0.1 mg/l IBA + 2.0 mg/l BA + 0.3 mg/l GA3) and treatment T6 (MS + 1.0 mg/l IBA + 2.5 mg/l BA + 0.3 mg/l GA3). The results obtained in this study can paved for more research on tissue culture of H. cannabinus.

Multiple shoots were obtained from shoot tips (2 to 3 mm) derived from mature plants (5 to 6 years old) of Citrus reticulata Blanco cv. Khasi mandarin and C. limon Burm.f. cv. Assam lemon when cultured on Murashige and Skoog (MS) medium, supplemented with (mg·liter-1) 1.0 BAP, 0.5 kinetin, and 0.5 NAA. Root induction was observed when 7-week-old single shoots (≈ 2 cm long) of both Citrus species were cultured on MS medium supplemented with (mg·liter-1) 0.25 BAP, 0.5 NAA, and 0.5 IBA. These plantlets were successfully established in the soil. Chemical names used: naphthalene acetic acid (NAA), indole 3-butyric acid (IBA), and benzylamino purine (BAP).

Present investigation was undertaken to standardize technique for in vitro micro-propagation of chickpea( Cicer arietinum ) cultivar Vishwas (Phule G 12). Micropropagation method for chickpea was established and this method enabled much more efficient propagation of plants. The present work was aimed at evolving a protocol for rapid multiplication of chickpea using micropropagation technique. Explants from shoot tip and node segment were cultured on MS medium supplemented with different concentrations of BAP and Kinetin (1.0 to 2.5 mg/l) and their growth responses like shooting were elucidated. The maximum multiple response was observed with 2 mg/l concentration of BAP from both types of explant. The highest number of shoots (12.5 ± 0.3) was achieved on MS medium with 2 mg/l BAP using node segments. The medium supplemented with 2 mg/l of BAP was found better than all other concentrations. Individual shoots were transferred to IBA and IAA (1.0-1.5 mg/l) for root induction. MS medium supplemented with 2 mg/l of IBA proved better for rooting. Rooted plantlets were successfully hardened in greenhouse and established in the pot.

An efficient protocol for plant regeneration through multiple shoots induction from shoot tips of Polygonum hydropiper (L.) was established. The highest percentage (96.6) of multiple shoot induction and number of shoots (9.0) per culture were found on MS supplemented with 2.0 mg/l Kn. The induced shoots were excised and inoculated on to MS contains different concentrations of IBA or NAA for rooting. The highest percentage (90.0) of root induction and the highest number of roots per shoot (12.0) was found on MS having 1.0 mg/l IBA. AbstractThe year 2018 marked 40 years since the birth of Louise Brown, the first baby born as a result of pioneering in vitro fertilization (IVF) treatment. Since then, advances have seen a wide range of reproductive technologies emerge into clinical practice, including adjuvant treatments often referred to as IVF “add-ons.” However, these “optional extras” have faced growing criticism, especially when they have often come at additional financial cost to the patient and have little evidence supporting their efficacy to improve pregnancy or birth rates. Despite this, according to the latest national patient survey by the Human Fertilisation and Embryology Authority, three quarters of patients who had fertility treatment in the United Kingdom in the past two years had at least one type of treatment add-on highlighting the growing demand for these interventions. This article uses a psychosocial perspective to consider the motivations behind patient and clinician behavior along with the wider societal and economic factors that may be impacting upon the increase in the use of adjuvant treatments in fertility clinics more widely. It suggests the reasons fertility patients use unproven “optional extras” are complex, with interpersonal, psychological, and social factors intertwining to generate an increase in the use of IVF add-ons.

To evaluate an efficient protocol for the micropropagation of Tupistra albiflora K. Larsen, the effects of N6-benzylaminopurine (BA) and naphthalene acetic acid (NAA) concentrations on multiple shoot and root induction were examined. In vitro shoots were used as the explant materials which were cultured on Murashige and Skoog (MS) agar medium supplemented with 0, 1, 2, 3 and 4 mg/L BA for 4 weeks to induce multiple shoots. It was found that the MS medium containing 3 mg/L BA induced 100 % shoot formation with the highest number of 3.2 shoots per explant (2.4-fold significantly higher than the control). For root induction, in vitro shoots were cultured on MS agar medium supplemented with 0, 1, 2, 3 and 4 mg/L NAA for 8 weeks. The results showed that the MS medium containing 1 mg/L NAA induced 100 % root formation with the highest number of 6.6 roots per explant (1.8-fold significantly higher than the control).

Digitalis trojana Ivanina is a member of the Plantaginaceae family and known by its common name, Helen of Troy foxglove. It is perennial endemic to Çanakkale and Balıkesir, northwestern Turkey. In order to develop an efficient shoot regeneration protocol, the leaf explants of D. trojana were cultured on Murashige and Skoog (MS) medium containing 6-benzyl adenine (0.1, 0.5, 1.0, 3.0, 5.0 mg/L) and α-naphthalene acetic acid (0.1, 0.5, 1.0 mg/L), 3% (w/v) sucrose and 0.8% (w/v) agar. The highest number of regenerated shoots was obtained from leaf explants that were cultured on MS medium with 3.0 mg/L BA+0.1 mg/L NAA. Regenerated shoots were rooted on MS medium without plant growth regulators. Rooted plants (2–3 cm) were separately transferred to pots containing a mixture of peat and perlite (2:1 v/v) and acclimatized successfully in a growth chamber.

The aim of the work was to determine the effect of fungicides: Topsin M 70 WG, Bravo 500 SC, and Sportak 45 EC in two concentrations on the growth of fungi: Aspergillus niger, Epicoccum purpurascens, Cladosporium sphaerospermum, Mortierella isabellina and Penicillium verrucosum var. verrucosum on PDA and Murashige-Skoog medium (MS). The smallest index of linear growth rate of the fungi had Topsin M 70 WG for fungi: E. purpurascens and P. verrucosum var. verrucosum on PDA and MS medium and for M. isabellina and C. sphaerospermum on MS in concentrations of 71.42 mg/l and 107.13 mg/l. The highest index of linear growth had Bravo 500 SC in concentrations of 0.5 ml/l and 0.75 ml/l on PDA for E. purpurascens and A. niger. The lowest index of linear growth on MS for E. purpurascens, C. sphaerospermum and P. verrucosum var. verrucosum was noted down. The highest effectiveness in control of linear growth all tested species of fungi was found for fungicide Sportak 45 EC in concentration of 0.55 ml/l and 0.825 ml/l on MS and PDA. Among all tested fungicides used in vitro propagation fungicides from imidazoles group are recommended.

An efficient in vitro regeneration system was developed for Rauvolfia serpentina L. through direct and indirect organogenesis from nodal and leaf explants. Among the different growth regulators, MS medium supplemented with 2.0 mg/l BAP, 0.5mg/l IAA and 0.02mg/l NAA found best for the multiple shoot formation from nodal segments. In this combination 98% explants produced multiple shoots and the average number of shoots per explants is 13∙4. The frequency of callus induction and multiple shoot induction from leaves was highest 88% in MS medium supplemented with 2.0 mg/l BAP, where mean number of shoots/explants was 12.5. The highest frequency of root induction (80%) and mean number of roots/plantlets (10) were obtained on half strength of MS medium containing 0.2 mg/l IBA. In vitro study was carried out in an important medicinal plant Tinospora cordifolia (Willd.) Miers belonging to the family: Menispermaceae. Vegetative parts such as stem, leaf and nodal explants were excised from an elite in vivo grown mature plant and thereafter cultured on Murashige-Skoog (MS) medium supplemented with different hormonal concentrations for callus induction and organogenesis. Callus formation occurred from nodal segments, leaf and inter-node explants when planted on different combinations of hormones. Tinospora cordifolia showed response for in vitro shoot growth from the nodal segment. The best shoot growth was observed on MS medium supplemented with kinetin (1.5 mg/l). Similarly, the best result for root induction was obtained on MS medium supplemented with 6-benzylaminopurine (1.0 mg/l) and naphthaleneacetic acid (2.5 mg/l). Context: Orchid produces a huge number of minute seeds but the seeds can not germinate easily in nature due to the lack of endosperm in the seeds is an incompatibility barrier that limits its propagation in nature. Objectives: To develop in vitro culture techniques for quick propagation of Vanda tessellate, a commercially important orchid species. Materials and Methods: Shoot tips were used as experimental materials. The explants were surface sterilized and the shoot tips were excised. The isolated shoot tips were cultured in MS medium supplemented with different concentration and combinations of auxin and cytokinin. Results: The combination of 1.5 mgl-1 NAA and 1.0 mgl-1 BAP was proved to be the best medium formulation for multiple shoot formation as well as maximum shoot elongation. The single shoots were isolated from the multiple shoots and subcultured in MS medium having NAA and IBA individually and in combinations for root induction. Maximum root induction was obtained in MS agarified medium having 0.5 mgl-1NAA and 1.0 mgl-1IBA. The well rooted plantlets were hardened successfully in the potting mixture containing coconut husk, perlite, charcoal, brick pieces in the ratio of 2:1:1:1 and eventually established under natural condition.Conclusion: An efficient regeneration protocol for micropropagation in V. tessellata through shoot tip culture has been established.Key words: Shoot tip; micropropagation; orchid.DOI: 10.3329/jbs.v17i0.7122J. bio-sci. 17: 139-144, 2009
