ABSTRACT Myracrodruon urundeuva Fr. All. is a tree threatened with extinction, which has wood and medicinal potential. This study aimed to analyze the in vitro shoot regeneration in M. urundeuva, in order to increase the species multiplication. Two experiments were conducted: 1) concentrations of 6-benzylaminopurine (BAP) (0.0, 2.0, 4.0, 8.0 and 16.0 µM), in association with naphthaleneacetic acid (NAA) (0.0, 1.5 and 3.0 µM), in explants (cotyledon, hypocotyl and cotyledonary node); 2) concentrations of meta-topolin (mT) (0.0, 2.0, 4.0, 8.0, 16.0 and 32.0 µM) in explants (biaxillary, medial uniaxillary and apical basal nodal segment). The percentage of explants responsive to shoot regeneration, percentage of callus explants, number of shoots and shoot length were evaluated. In the first experiment, the shoot regeneration occurred only in explants of the cotyledonary node and hypocotyl type, with the highest responsiveness percentage (76.67 %) and number of shoots (1.97 and 1.63) obtained for the cotyledonary node in the presence of 3.0 µM of NAA in association with 2.0 (1.97 shoots/explant) and 4.0 µM (1.63 shoots/explant) of mT. In the second experiment, the resolution of the obtained quadratic equation indicates that the use of basal explant with 24.59 µM of mT added to the culture medium leads to the highest number of shoots (1.86). However, despite the mT having increased the mean number of shoots, all treatments containing this cytokinin showed callus formation. As a conclusion, it is possible to regenerate shoots in M. urundeuva from the cotyledonary node using BAP in association with NAA.

In vitro regeneration of pea explants (Pisum sativum L. var. ‘Lincon’) was done in 49 different combinations and concentrations of BAP, BAP and NAA, BAP and IBA, TDZ, TDZ and Adenine for shoot regeneration from hypocotyl, root, leaf and cotyledonary node. High frequency shoot regeneration was obtained in hypocotyl (81.43%), root(83.53%) and cotyledonary node(72.76%) on MS medium supplemented with 4.50 mg/l BAP and 1.86mg/l NAA, 2.00mg/l TDZ and4.50 mg/l BAP and 1.86mg/l NAA respectively. No shoot regeneration was obtained from leaf explants on any of the combination used. Shoot elongation was observed on the same medium used for shoot regeneration respectively.MS medium supplemented with 0.20 mg/l IBA was found best for root regeneration from in vitro raised shoots. The plantlets were able to regenerate within 6-7 weeks. The regenerated plantlets were acclimatized in pre-sterilized cocopeat.

The aim of this study is to establish an improved shoot regeneration of potential medicinal plants, Melaleuca alternifolia. The essential oil of these plants is useful in the cosmetic and pharmaceutical industries. Despite the importance of this species, in vitro regeneration was limited. Therefore, the present study use various concentrations of plant growth regulators to improve shoot regeneration of these plants. The present study shows that axillary shoot gave the best response to the treatment containing 0.5mg/L BAP and 0.1mg/L NAA. The average number of shoots is 23 with 77% of shooting. For rooting experiment, 35% of axillary shoot were rooted when cultured on MS medium supplemented with 1.5mg/L IBA. In vitro plantlet was later developed after 3 months.

A micropropagation method, through axillary shoot proliferation, was elaborated for <em>Inula royleana </em>DC. (Asteraceae), a medicinal plant native of Himalaya. Primary explants (cotyledonary node explants) and secondary explants (node explants of in vitro regenerated shoots) of the plant, inoculated on MS medium supplemented with 0.1 μM NAA and 5.0 μM kinetin, regenerated 3.4 ± 1.2 and 5.1 ± 1.9 axillary shoots per explant, respectively. The regenerated shoots were easily rooting and adapting to growth in soil.

Surface disinfested nodal and shoot-tip sections of chinkapin oak (Quercus muehlenbergii Engelm.), obtained from adult or juvenile source, when cultured on WFM supplemented with BA or kinetin (1.0 -5.0 mg l-1) produced greater number of axillary shoots per explant and shoot lengths than MS medium. Nodal and shoot-tip explants cultured in WPM containing cytokinins, BA or kinetin (0.1 - 5.0 mg l“) resulted in greater number of axillary shoots than media containing auxins, 2,4-D or NAA (1.0 - 5.0 mg l-1). In vitro grown shoot explants cultured in WFM shoot multiplication medium containing thidiazuron did not produce axillary shoots. Microshoots when cultured in WFM plus NAA or IBA (0.1 -2.0 mg l-1), or subjected to IBA (0.5 mg l-1) pulse treatment (0, 5, 10 or 15 min.) did not root.
