About invasion of grey mullet by myxosporidia Myxobolus episquamalis at Kerchenskyi preglass of the Black sea

Flathead grey mullet, Mugil cephalus, with mass whitish cystlike plasmodia on their scales were collected at Kerchensky preglass of the Black Sea in 2015. The prevalence of infected fish varied from 15% in spring, reached 100% in summer, and declined to 2.5% in autumn. No fish mortality was detected. The spores were oval in frontal view, tapering to a blunt apex. Two unequal polar capsules were pyriform and extended over the anterior half of spore. Spores were 8.2±0.03 µm (7.9-8.4) long, 5.9±0.23 µm (5.2-7.3) wide, and 4.4±0.17 µm (4.0-4.7) thick. Two pyriform and unequal polar capsules were observed (4.0±0.07 µm (3.3-4.5) long, and 1.5±0.24 µm (1.1-1.8) wide). The investigation of nucleotide sequences of the 18S rDNA gene of the myxosporean spores from scales with universal primer А (5′-ACCTGGTTGATCCTGCCAGT-3′) and В (5′-TGATCCTTCTGCAGGTTCACCTAC-3′) showed 100% identity with episquamalis, and 99 % with M. bizerti, M. ichkeulensis, M. spinacurvata and Myxobolus sp. previously detected in mullets. The results obtained from the present study reveal that M. episquamalis, found on the scales of M. cephalus from Kerchensky preglass of the Black Sea, is new record for Russian waters.

The environmental monitoring of chemical toxicants has been a widely studied topic in the last few decades. The main aim of the present study was to determine the total concentration of nine elements (Cd, Cr, Pb, Ni, Al, Cu, Fe, Mn, and Zn) in the fish species grey mullet (M. cephalus) and in the coastal marine waters collected from various sampling points along the Black Sea (Bulgaria) and the Ionian Sea (Italy). Further, those results were applied to predict the pollution degree in those coastal marine environments. The fish samples were subject to acid digestion followed by appropriate analytical determination. The metal concentrations in marine water samples collected from the Black Sea (Bulgaria) and the Ionian Sea (Italy) were also analyzed. Unpaired Student’s t-test and the one-way ANOVA were applied for the statistical analysis of the data. The statistical results revealed a significant variation (p < 0.0001) in the concentration of various fish tissues. The accumulation of toxic and essential elements differs significantly in grey mullet species caught from the Black Sea (Bulgaria) and the Ionian Sea (Italy). The results from this study may serve as a convenient approach during marine pollution programs set by both countries (Italy and Bulgaria).

The Black Sea offers numerous harnessing possibilities for the medical and pharmaceutical, agricultural, food industry and cosmetic fields. Collagen extraction from the Black Sea fish is a research area of great interest. The purpose of this paper is to optimize the collagen quantitative analysis method based on hydroxyproline reagent through visible molecular absorption spectrometry. The adapted method was validated, achieving the following performance criteria: linearity, detection and quantification limits, accuracy/fidelity, stability/sturdiness, repeatability, and measurement uncertainty. The validated method was applied for the quantitative determination of collagen content in Grey Mullet fish and for the evaluation of collagen extraction output.

The associations and ecological conditions of the existence of modern diatoms of the North-West (Pridneprovsky), Prikerchensky and Eastern regions of the subtidal zone of the Black Sea are considered. Based on the unity of the composition of the Present and Sarmatian-Meotian diatom flora, an attempt has been made to model some of the ecological c situation of the Late Miocene Euxinian basin.
