Berberine sulfate was shown to possess antimicrobial activity against a wide variety of microorganisms including Gram-positive and Gram-negative bacteria, fungi, and protozoa. The antibacterial activity against Vibrio cholerae and Staphylococcus aureus was dependent on the inoculum size of the test organism and pH of the medium. A method of microbiological assay sensitive to 5–10 μg/ml of the drug was developed. The drug was shown to exert a more rapid antibacterial activity than chloramphenicol and tetracycline on V. cholerae, the K values being 2.4 ×10−2 sec−1, 7.8 × 10−3 sec−1, and 5.2 × 10−3 sec−1 respectively. Berberine sulfate was shown to be bacteriocidal to V. cholerae and bacteriostatic to S. aureus, at concentrations of 35 and 50 μg/ml. In both these organisms concentrations of 35 and 50 μg/ml of the drug inhibited ribonucleic acid (RNA) and protein synthesis almost immediately after the addition of the drug. There was little effect on deoxyribonucleic acid (DNA) synthesis at these concentrations.

The plasmid contents of seven exfoliative toxin-producing strains of phage group 2 Staphylococcus aureus were analyzed by agarose gel electrophoresis and deoxyribonucleic acid-deoxyribonucleic acid hybridization. All strains were found to contain a large plasmid with a molecular weight of 27 X 10(6) except for strain RW1005. A comparison of the restriction endonuclease cleavage products by agarose gel electrophoresis showed that the number and size distribution of the fragments of all these Tox plasmids were similar, except for pRW002, which appeared to contain two deletions. Deoxyribonucleic acid-deoxyribonucleic acid hybridization studies confirmed that these plasmids were related to a plasmid which carried the genes for exfoliative toxin B and bacteriocin R1 biosynthesis and that they shared some sequence homology with the penicillinase plasmid pI258 isolated from a phage group 3 S. aureus.

Penelitian ini bertujuan mendeteksi gen tst isolat Staphylococcus aureus (S. aureus) susu sapi perah dan susu kambing melalui amplifikasi gen 23S rRNA sebagai langkah awal pencegahan beberapa kasus keracunan susu. Metode penelitian meliputi reidentifikasi bakteri, preparasi deoxyribonucleic acid (DNA), amplifikasi gen 23S rRNA, amplifikasi gen tst, dan pengurutan DNA. Hasil reidentifikasi bakteri melalui pewarnaan Gram, uji katalase, uji koagulase, uji fermentasi MSA, uji VJA, dan uji VP adalah positif bakteri S. aureus. Hasil amplifikasi gen tst terhadap 3 isolat S. aureus menunjukkan hanya 2 isolat memberikan hasil positif. Hasil positif tersebut ditandai dengan munculnya fragmen DNA yang memiliki panjang spesifik (350 bp) sesuai dengan produk polymerase chain reaction (PCR) dari referensi dan database GeneBank. Disimpulkan bahwa dengan menggunakan metode PCR dapat terdeteksi adanya gen tst pada isolat S. aureus asal susu sapi perah dan susu kambing dari Bogor.

The formula for a modified Vogel and Johnson Agar is presented. In addition to the ingredients found in Vogel and Johnson Agar, the new medium contains, per liter: 5 g of beef extract, 2 g of deoxyribonucleic acid, 2 g of phosphatidyl choline and 780 units of catalase spread on the plates before inoculation. This new medium is as effective as Baird-Parker Agar in enumeration of stressed Staphylococcus aureus, and in enumeration of staphylococci from naturally contaminated processed food samples.
