The research was carried out at the Laboratory of Agrobiotechnology of the Institute of Biology and Plant Biotechnology, Faculty of Agronomy of Vytautas Magnus University (till 2019 Aleksandras Stulginskis University). The effect of basal nutrition medium, subculture and growth regulators on the adventitious shoots formation from isolated explants of Miscanthus × giganteus was analysed. The apical meristem of in vitro developed adventitious shoots of Miscanthus × giganteus was cultivated in the Woody Plant Medium (WPM) and Murashige and Skoog (MS) nutrition medium supplemented with different 6-benzylaminopurine (BAP) and indole-3-butyric acid (IBA) or indole-3-acetic acid (IAA) combinations. It has been determined that the formation frequency of adventitious shoots on the WPM nutrition medium was from 5.0 to 18.5% (after the first subculture) and from 19.5 to 38.3% (after the second subculture) higher in comparison with the MS nutrition medium supplemented with the same combinations of growth regulators. It has been established that on the WPM nutrition medium the formation frequency of adventitious shoots after the second subculture was from 13.3 to 18.5% higher in comparison with the first subculture, while on the MS nutrition medium the effect of subculture was not significant. The most intensive frequency of shoots formation (48.8%) and the highest shoot number per explant (24.1 units) have been obtained in the WPM medium supplemented with 3.0 mg l–1 BAP + 0.4 mg l–1 IBA + 0.01 mg l–1 IAA after the second subculture.

The impact of the plant growth regulators (PGRs) Stimpo, Regoplant, and Charkor on the production of the second-generation energy crop Miscanthus × giganteus on marginal post-military soil was investigated during two vegetation seasons. The land, previously a tank training polygon, has not been in use since 1990 and has become marginal. Biological parameters (stem, shoot, and root lengths) and dry biomass values were evaluated in relation to the applied treatments. The multivariate general linear model (M-GLM) results showed a positive influence of Charkor on M. × giganteus development; the effect was markedly higher in the second year of vegetation. The impact of Stimpo and Regoplant was less noticeable; nevertheless, certain combinations of treatments showed satisfactory results. The M-GLM approach detected the inter-influence of the main factors of the production process, i.e., PGRs, soil, and year of growing. The results showed the predominant influence of year, PGRs and combined factor PGRs × year on the biological parameters; the other studied factors and their combinations were not as effective. Further research should focus on verifying the field-scale results for the M. × giganteus plantation established in a post-military area and compare the lab and field studies.

Research was carried out at the Institute of Biology and Plant Biotechnology of Aleksandras Stulginskis University and at the Laboratory of Agrobiotechnology of the Joint Research Centre. The effect of growth regulators on the direct organogenesis from isolated explants of Miscanthus × giganteus was analysed. Rhizome segments of Miscanthus × giganteus were cultivated in the Murashige and Skoog (MS) nutrient medium supplemented with different 6-benzylaminopurine (BAP) or thidiazuron (TDZ) and ɑ-naftilacetic acid (NAA) combinations. It has been determined that the isolated explants in the nutrient medium without growth regulators did not form adventitious shoots. In the nutrient medium supplemented with growth regulators the regeneration frequency varied from 5.6 to 21.1%. The adventitious shoots formation frequency due to the TDZ exposure, depending on the concentration, was from 5.4 to 10.0% higher (in combination with 0.05 mg l–1 NAA) and from 4.4 to 8.8% higher (in combination with 0.1 mg l–1 NAA) in comparison with the adequate concentration of TDZ. The most intensive shoot formation frequency and the highest shoot number per explant have been obtained in the medium supplemented with 2.0 mg l–1 TDZ + 0.05 mg l–1 NAA.

This long term study demonstrates for the first time that it is possible to propagate embryogenic Vigna trilobata and to subsequently initiate the differentiation of embryos into complete plantlets. Initiation of callus was possible on 2,4-D. Somatic embryos differentiated on modified MS basal nutrient medium with 1.0 mg/l  of 2,4-D and 0.5 mg/l  of Kn. Sustained cell division resulted in globular and heart shape stages of somatic embryos. Transfer of embryos on to a fresh modified MS basal medium with 0.5 mg/l of Kn and 0.5 mg/l of GA3 helped them to attain maturation and germination. However, the propagation of cells, as well as the differentiation of embryos, were inhibited by a continuous application of these growth regulators. For this reason, a long period on medium lacking these growth regulators was necessary before the differentiation of embryos occurred again.
