A protocol for multiple shoot induction and plant regeneration from nodal explants of Ocimum citriodorum has been developed. Nodal explants inoculated on Murashige and Skoog (MS) medium supplemented with 1.0 mg/l Benzyl adenine (BAP) and 0.025 mg/l indole -3- acetic acid (IAA) showed better growth response (80%) and produced 15.2 ± 1.28 shoots per explant with an average length of 6.17 ± 0.29 cm after 35 days. Roots were induced after transfer to half strength MS medium supplemented with 0.5 mg/l Indole -3- butyric acid (IBA) produced 6.0 ± 1.0 roots with an average height of 4.9 ± 0.26 cm after 30 days. Plantlets with well developed root and shoot systems were successfully acclimated (80 %) and established in earthen pots containing mixture of soil, vermiculite and farm yard manure (1:1:1). An efficient plant regeneration protocol was developed for Artemisia annua, an aromatic medicinal herb. Nodal explants inoculated on MS supplemented with 4.44 ?M BAP showed better growth response and produced 116.2 ± 0.1 micro-shoots of an average length 1.9 ± 0.3 cm after 35 days culture. The cluster of small shootlets were cultured on shoot elongation medium supplemented with 1.44 ?M GA3 and 10% coconut milk (CM) showed shoot elongation up to 4.6 ± 0.7 cm. Roots were induced after transfer to half strength MS supplemented with 2.46 ?M IBA produced 14.3 ± 0.2 roots with an average height of 4.3 ± 0.53 cm after 30 days. The rooted plantlets were transferred for hardening, 80 per cent of plantlets survived and resumed growth in the mixture of soil, vermiculite and farm yard manure (1 :  1 : 1). An efficient plant regeneration protocol was developed for Justicia gendarussa Burm.f. (Acanthaceae), an important medicinal shrub. Nodal segments grown on Murashige and Skoog (MS) medium containing 1.0 mg/l Benzyl adenine (BAP) with 10 % coconut milk showed better growth response and produced 10.5 ± 0.6 shoots per explant with an average length of 4.4 ± 0.3 cm after 35 days. Rooting of shoots was achieved on growth regulator free half strength MS medium produced 5.3 ± 0.25 cm roots with an average height of 4.8 ± 0.2 cm after 30 days. Shoot tip explants of Exacum travancoricum, an endangered herb mainly known for ornamental purpose grown on MS with 4.44 μM BAP and 1.34 μM NAA showed better growth response and produced 29.3 ± 0.3 shoots per explant with an average length of 4.6 ± 0.1 cm after 35 days. Roots were induced after transfer to half strength of MS supplemented with 2.46 μM IBA produced 4.8 ± 0.62 roots with an average height of 3.6 ± 0.10 cm after 30 days. The rooted plantlets were transferred for hardening, 80 per cent of plantlets survived and resumed growth in the mixture of soil, vermiculite and farm yard manure (1 : 1 : 1). Young nodal explants (0.5-1cm) of Salvia sclarea L. was used for synthetic seed preparation.Synthetic seeds were prepared using 5% sodium alginate and 1.11% calcium chloride. Seed germination was observed on MS medium fortified with 1.4µM GA3+4.4µM BA after twenty days of culture. Further multiple shoot induction was observed after fifteen days of shootinduction.

<p>Bauhinia purpurea L. is a leguminous plant moderate sized tree with multipurpose value. It is distributed in sub-Himalayan tracts. It has been cultivated in the plain region up to the elevation of 1350 m. Mature seeds of Bauhinia purpurea L. were cultured on half strength Murashige and Skoog (1962) (MS) medium. Nodal explants obtained from germinated seedlings were cultured on MS medium containing 0.5 M BAP produced multiple shoots which were used for experimental purposes. Nodal explants obtained from cultured were subcultured on different concentrations of N-Benzyl -9-(2-tetrahydropyranyl) (BPA) and Indole-3acetic acid (IAA). The best proliferation of nodes and shoots were observed on the MS medium supplemented with 0.5 M BPA and 0.1 M IAA. After 8 weeks of culture, the propagated plants were acclimatized and transferred to the sand box containing 1:1 soil and sand. Well rooted plants were then established in the field. All the data collected were worked out statistically with SPSS, a system of analytical procedure.</p>

Efficientin Vitro direct multiple shoot regeneration from Solanum pubescens was achieved from leaf explants on MS medium Sublimated with B5 vitamins and different concentrations and different combinations of PGRs like BAP, NAA and GA3. The maximum numbers of multiple shoots were achieved from leaf explants on 3.0 mg/l BAP + 1.0mg/l GA3. The regenerated shoots were transferred in to half strength MS medium fortified with IBA for root induction. Rooted plantlets were successfully acclimatized. This new and transfer into the field Conditions. Standardized and reproducible protocol useful the mass propagation of Solanum pubescens.

The shoot-tip explant harvested from ethyl methane sulphonate (EMS) and gamma ray (GR) mutagenized seedling was cultured over MS medium fortified with NAA and BAP for five generations to amplify the mutated sector. Mutagens reduced the regeneration efficiency of the explant and affected its plant growth regulator-dependence for multiple shoot induction. While the 12d-old shoot-tip from GR-treated seedling induced shoots with 0.5µM NAA+6.6µM BAP; that from EMS-treated seedling induced shoots with 8.8µM BAP. The present study establishes that the mutagens affect the regeneration process in the explant.

Efforts have been made to establish a protocol for direct multiple shoot regeneration from both in vitro grown seedlings and mature plants of Citrus macroptera. Both nodal and shoot tip explants taken from in vitro grown seedlings were cultured in MS supplemented with different concentrations of BAP and Kn either singly or in combinations. Both these explants are capable to regenerate and produce in vitro multiple shoots. Maximum number of shoots were obtained from nodal explants in MS supplemented with 1.0 mg/l BAP. BAP alone was found superior to Kn. On the other hand, only nodal explants from mature plants were used and 1.0 mg/1 BAP was also found best suitable for shoot induction and multiplication. A simple, repeatable and efficient protocol for direct multiple shoot regeneration from internodal explants has been defined in peppermint ( Mentha x piperita var. Indus). In vitro regenerated shoots of peppermint were excised into 4 to 8 mm long internodes and cultured on Murashige and Skoog's medium supplemented with different cytokinins. In the hormonal assay, 3.0 mg L-l zeatin or 6-isopentenyl adenine independently supplemented to half strength MS medium exhibited multiple shoot regeneration, while thiaduzorn (0.1-3.0 mg L−1) showed no morphogenetic effect. A maximum of 85% in vitro cultured explants showed multiple shoot formation with an average of 7 shoots per explant on MS medium supplemented with zeatin. Multiple shoots were initiated within three weeks of cultivation. Internodes with regenerated multiple shoots were transferred to half - strength MS medium without supplementing with any plant growth hormone for shoot elongation and rhizogenesis. Rooted plants acclimatized and grew to maturity under glasshouse conditions. The plantlets developed were phenotypically identical to the parent plant and exhibited 96 % survival.

In vitro embryo morphogenesis and micropropagation of Dendrobium aggregatum Roxb. were described. The gradual developmental stages of embryos to seedlings were traced out. Within two weeks of culture the cells of undifferented embryos underwent repeated aniclinal and periclinal division producing a compact, green parenchymatous cell mass called spherule that emerged out by rupturing the testa. The spherules subsequently differentiated into greenish protocorms were considered as typical seed germination. Germination occurred on both (MS and Phytamax (PM) medium but MS medium proved to be more efficient. The primary protocorms underwent profuse proliferation through production of secondary (2º) protocorms when transferred to different plant growth regulators (PGRs) supplemented MS; the medium fortified with 2.0 mg/l BAP and 1.0 mg/l NAA proved to be most effective for induction of 2º protocorms and seedling development. Multiple shoot buds (MSBs) were induced in pseudobulb segments of the in vitro grown seedlings when cultured on different PGRs supplemented media; and the maximum number of MSBs were obtained MS + 2.0 mg/l BAP + 0.5 mg/l picloram. The MSBs underwent elongation and then they rooted when they were transferred to half strength of MS + 0.5 mg/l IAA. The well rooted plantlets were finally transferred to outside natural environment with 80% survival. D. O.
