Juvenile leaf explants of Stevia rebaudiana Bertoni produced maximum callus than the nodal explants  cultured on MS containing 11.31 mM 2, 4-D and 2.22 mM BAP. Callus transferred to MS supplemented with 4.44 mM BA and 1.34 mM NAA showed better growth response and produced 14.0 ± 1.0 shoots with an average length of 5.6 ± 0.1 cm after 28 days. All plantlets produced profuse rooting within 25 days after transfer to half strength of MS basal medium supplemented with 2.46 mM IBA. An efficient plant regeneration protocol was developed for Justicia gendarussa Burm.f. (Acanthaceae), an important medicinal shrub. Nodal segments grown on Murashige and Skoog (MS) medium containing 1.0 mg/l Benzyl adenine (BAP) with 10 % coconut milk showed better growth response and produced 10.5 ± 0.6 shoots per explant with an average length of 4.4 ± 0.3 cm after 35 days. Rooting of shoots was achieved on growth regulator free half strength MS medium produced 5.3 ± 0.25 cm roots with an average height of 4.8 ± 0.2 cm after 30 days. Shoot tip explants of Exacum travancoricum, an endangered herb mainly known for ornamental purpose grown on MS with 4.44 μM BAP and 1.34 μM NAA showed better growth response and produced 29.3 ± 0.3 shoots per explant with an average length of 4.6 ± 0.1 cm after 35 days. Roots were induced after transfer to half strength of MS supplemented with 2.46 μM IBA produced 4.8 ± 0.62 roots with an average height of 3.6 ± 0.10 cm after 30 days. The rooted plantlets were transferred for hardening, 80 per cent of plantlets survived and resumed growth in the mixture of soil, vermiculite and farm yard manure (1 : 1 : 1). An efficient plant regeneration protocol was developed for Artemisia annua, an aromatic medicinal herb. Nodal explants inoculated on MS supplemented with 4.44 ?M BAP showed better growth response and produced 116.2 ± 0.1 micro-shoots of an average length 1.9 ± 0.3 cm after 35 days culture. The cluster of small shootlets were cultured on shoot elongation medium supplemented with 1.44 ?M GA3 and 10% coconut milk (CM) showed shoot elongation up to 4.6 ± 0.7 cm. Roots were induced after transfer to half strength MS supplemented with 2.46 ?M IBA produced 14.3 ± 0.2 roots with an average height of 4.3 ± 0.53 cm after 30 days. The rooted plantlets were transferred for hardening, 80 per cent of plantlets survived and resumed growth in the mixture of soil, vermiculite and farm yard manure (1 :  1 : 1). Leaf explants of Plumbago rosea L. an important medicinal plant inoculated on MS supplemented with 6.66 mM BAP and 2.69 mM NAA  produced numerous (105 ± 0.3) shootlets with an average length of 3.1 ± 0.0 cm. Small shootlets were transferred to shoot elongation medium supplemented with 1.11 mM BAP plus 1.44 mM GA3. The elongated shootlets transferred to half strength MS basal medium without any plant growth regulator produced 4.3 ± 0.2 rootlets per plants with average root length of 4.0 ± 0.0 cm after 25 days of culture. Rooted plantlets were transferred for hardening showed 80 - 90 per cent of plantlets success-fully established in the field. Potentially more than 50,000 plantlets could be produced within five subcultures from without callus phase obtained from leaf explant. Maximum root differentiation from leaf explants was obtained on MS supplemented with 5.38 mM IBA. An efficient indirect plant regeneration protocol was developed for Viola canescens, an important medicinal herb used in broad spectra of diseases in number of folk medicines since aeon. Excessive use of this plant without any rehabilitating measure has led to decline its natural population. Present investigation reports the use of zeatin to regenerate the plant from the callus on MS following its acclimatization on the soil condition. Calli of the plant responded positively to zeatin and maximum number of shoots 13.07 ± 2.01 were obtained when 9.12 μM concentration of zeatin was used. Regenerated shoots were subsequently rooted with IBA on MS and half strength MS and showed maximum number of roots 14.13 ± 1.64 after 60 days when medium was fortified with 4.92 μM IBA, followed by transferring them to soil condition, acclimatization of the plantlet was carried in growth chamber and then finally to the field for their survival where it showed 80% survival. Plant regeneration via somatic embryogenesis was assessed using immature leaf, petiole and apical meristem explants in Tetrapleura tetraptera. Somatic embryos were induced in the immature leaf using MS basal medium supplemented with 2,4-D and matured on MS basal medium containing BAP. Medium supplemented with 12 mg/l 2,4-D had the highest (43.1%) percentage of embryogenic calluses from immature leaf explants. Conversion of embryogenic callus to mature primary somatic embryo occurred in the medium that contained 1.2 mg/l BAP. Development of secondary embryogenic calluses to matured secondary embryos was highest (98.0%) in the medium with 0.4 mg/l BAP, while the highest average number of mature secondary embryos (6.0) was obtained in the same medium. Medium supplemented with 1.0 mg/l BAP and 0.5 mg/l IBA had the highest (38.7%) percentage of explants with shootbuds. The highest (18.1%) percentage of shoot elongation was obtained in medium with 1.0 mg/l BAP and 20 mg/l IBA. Shootbuds survived and produced roots on medium free of plant growth regulators. Shoots obtained on medium supplemented with 1.0 mg/l BAP and 20 mg/g IBA recorded the highest number of roots per plantlet (7.5) with no apparent morphological abnormality.

A protocol for multiple shoot induction and plant regeneration from nodal explants of Ocimum citriodorum has been developed. Nodal explants inoculated on Murashige and Skoog (MS) medium supplemented with 1.0 mg/l Benzyl adenine (BAP) and 0.025 mg/l indole -3- acetic acid (IAA) showed better growth response (80%) and produced 15.2 ± 1.28 shoots per explant with an average length of 6.17 ± 0.29 cm after 35 days. Roots were induced after transfer to half strength MS medium supplemented with 0.5 mg/l Indole -3- butyric acid (IBA) produced 6.0 ± 1.0 roots with an average height of 4.9 ± 0.26 cm after 30 days. Plantlets with well developed root and shoot systems were successfully acclimated (80 %) and established in earthen pots containing mixture of soil, vermiculite and farm yard manure (1:1:1). This study aims to determine the effect of interaction between several sterilants and types of explants and determine the best interaction with stevia culture's success rate. This study was designed using a Factorial Complete Randomized Design with two factors and three replications. The first factor is sterilant formulas i.e: fungicide 3% + alcohol 70% + Bayclin 5%; fungicide 3% + bactericide 6% + 70% alcohol + Bayclin 5%; sublimate 0.1% + 70% alcohol + Bayclin 5%; and fungicide 3% + bactericide 6% + sublimate 0.1% + alcohol 70% + Bayclin 5%. The second factor is explant types, i.e., stevia nodes and leaves. The variables observed are the percentage of contamination (%), percentage of alive explants (%), and browning percentage.  The interaction between sterilants and explant types only had a significant effect on the percentage of alive explants. The best interactions of sterilant formulas and explant types on the percentage of alive explants are 0.1% sublimate + 70% alcohol + 5% Bayclin and leaf explants.

Wedelia chinensis, belonging to the Asteraceae family, has been used in folk medicine in East and South Asia for the treatment of common inflammatory diseases and protection against liver toxicity. Previously, in vitro propagation through different tissue explants has been reported, including through nodal segments, axillary buds, and shoot tips, whereas leaf segments failed to proliferate. Here, we report on the in vitro propagation of W. chinensis by culturing young leaf explants in MS medium supplemented with 0.5 mg/L α-naphthaleneacetic acid (NAA), 0.75 mg/L thidiazuron (TDZ), 1 mg/L gibberellic acid (GA3), 3.75 mg/L adenine, 3% sucrose, and 0.8% agar at pH 5.8. Calli were observed in all explants derived from the youngest top two leaves, and the average percentage of shoot regeneration was 23% from three independent experiments. Then, several shoots were excised, transferred onto MS basal medium supplemented with 3% sucrose and 0.8% agar at pH 5.8, and cultured in a growth chamber for 1 to 2 months. Roots were easily induced. Finally, plantlets carrying shoots and roots were transferred into soil, and all of them grew healthily in a greenhouse. No morphological variation was observed between the regenerated plantlets and the donor wild-type plants. In addition, we also established root cultures of W. chinensis in culture medium (MS medium, 3 mg/L NAA, 3% sucrose, pH 5.8) with or without 0.8% agar. To the best of our knowledge, this is the first paper reporting plant regeneration from leaf explants in the herbal plant W. chinensis.
