Today, Construction based Industry is the prospering industry which has a high economical influence on any nation. Delay in the huge construction project increases the total project cost. Henceforth, uncertainties as well as risks must be significantly regarded during the project. For organizing and completing the projects in a financially, timely and qualitatively accountable manner, careful scheduling of projects is compulsory. Effectual scheduling of project assures project success. This study concentrates on qualitative analysis, risk identification, together with quantitative analysis. The targets are i) to ascertain the key risk aspects that disturb the project schedule, and ii) to find the probability of finishing the project within specified time. Questionnaires are distributed amongst 20 industry practitioners with disparate experience from [1] to [25] years.  Quantitative analysis is made by the methods like Monte Carlo simulation (MCS) and PERT. @RISK by Palisade corp. is utilized for MCS.

ABC Company is an agent of network construction, operation, and maintenance. ABC Company is currently implementing the STTF (Shit to the Front) project, which is the project to add FTTH (Fiber to the Home) networks in areas that can have high customer demand. One of the STTF project construction sites is the Indra Prahasta II housing location. However, the project is currently experiencing work delays due to the Covid-19 disaster in Indonesia. Delays in project execution can result in potential prospects choosing another company that provides similar services. The project schedule can be accelerated using the crashing method and TCTO (Time Cost Trade-Off) analysis to solve this problem. This research's acceleration will be carried out with alternatives for adding 3 hours, 2 hours, 1 hour, and an alternative to increasing workers' number. This project has an average duration of 55 working days with a total cost of Rp 604,124,460. The results obtained from data processing, on the alternative of adding 1 hour of overtime work, the total duration becomes 54 working days with total project cost is Rp 605,734,138. In addition to 2 hours of overtime work, the project's total duration can be reduced to 54 days with a total project cost Rp 606,803,619. And for the addition of 3 hours overtime, the total duration can be shortened to 54 days with a total cost of Rp 606,803,619. As for increasing the number of workers, project work duration can be shortened to 54 working days with a total project cost Rp 604,556,748

Single-cell RNA sequencing determines RNA copy numbers per cell for a given gene. However, technical noise poses the question how observed distributions (output) are connected to their cellular distributions (input). We model a single-cell RNA sequencing setup consisting of PCR amplification and sequencing, and derive probability distribution functions for the output distribution given an input distribution. We provide copy number distributions arising from single transcripts during PCR amplification with exact expressions for mean and variance. We prove that the coefficient of variation of the output of sequencing is always larger than that of the input distribution. Experimental data reveals the variance and mean of the input distribution to obey characteristic relations, which we specifically determine for a HeLa data set. We can calculate as many moments of the input distribution as are known of the output distribution (up to all). This, in principle, completely determines the input from the output distribution.

Prefabricated buildings are the direction of the future development of the construction industry and have received widespread attention. The effective execution of prefabricated construction project scheduling should consider resource constraints and the supply arrangement of prefabricated components. However, the traditional construction resource-constrained project scheduling implementation method cannot simultaneously consider the characteristics of the linkage between component production and on-site assembly construction. It cannot also fully adapt to the scheduling implementation method of the prefabricated construction projects. It is difficult to work out a reasonable project schedule and resource allocation table. In order to determine the relevant schedule parameters that can reflect the actual construction situation of the prefabricated building and meet the scheduling requirements of the prefabricated project, this study proposes a prefabricated construction project scheduling model that considers project resource constraints and prefabricated component supply constraints. Additionally, it improves the design of traditional genetic algorithms (GAs). Research results of the experimental calculation and engineering application show that the proposed project scheduling optimization model and GA are effective and practical, which can help project managers in effectively formulating prefabricated construction project scheduling plans, reasonably allocating resources, reducing completion time, and improving project performance.

In order to overcome the difficulty in quantifying rework by traditional project schedule management tools, this study proposes an innovative method, namely improved Critical Chain Design Structure Matrix (ICCDSM). From the perspective of information flow, the authors firstly make assumptions on activity parameters and interactions between activities. After that, a genetic algorithm is employed to reorder the activity sequence, and a banding algorithm with consideration of resource constraints is used to identify concurrent activities. Then potential criticality is proposed to measure the importance of each activity, and the rework impact area is implicated to indicate potential rework windows. Next, two methods for calculating project buffer are employed. A simulation methodology is used to verify the proposed method. The simulation results illustrate that the ICCDSM method is capable of quantifying and visualizing rework and its impact, decreases iterations, and improves the completion probability. In this vein, this study provides a novel framework for rework management, which offers some insights for researchers and managers.
