The main objective of cancer treatment with chemotherapy is to kill the cancerous cells without affecting the healthy normal cells. In the present study, bioactivity-guided purification of the n-chloroform soluble fraction from the methanol extract of Roscoea purpurea resulted in the identification of two new labdane diterpenes: coronarin K (1) and coronarin L (2), along with eight known compounds, coronarin A (3), bisdemethoxycurcumin (4), kaempferol 3-O-methyl ether (5), kaempferol (6), fenozan acid (7), 3-(3-methoxy,4-hydroxyphenyl)-2-propenoic acid ferulic acid (8), caffeic acid (9), and gallic acid (10). The structural identification of new compounds (1 and 2) were determined by detailed analysis of 1D (1H and 13C) and 2D NMR (COSY, HSQC, and HMBC) spectroscopic data. The relative configurations of 1 and 2 were determined with the help of NOESY correlations and comparison of optical rotations with known labdane diterpenes, with established stereochemistry, while structure of known compounds was established by direct comparison of their NMR data with those reported in the literature. This is the first report of isolation of this labdane diterpenes and phenolic classes of secondary metabolites in R. purpurea. In the preliminary screening, the methanol extract and its fractions were tested for the cytotoxic activity against a panel of four cancer cell lines (A549, HCT-116, Bxpc-3, and MCF-7); extract and its chloroform fraction were found to be active against the lung cancer cell line, A-549, with IC50 value <25 μg/ml. Owing to the notable cytotoxic activity of the chloroform fraction, the compounds (1–5) were screened for their cytotoxicity against all the cell lines by MTT assay. Coronarin K, 1 showed significant cytotoxic potential against lung cancer cell lines (A-549), with IC50 value of 13.49 μM, while other compounds did not show activity below 22 μM.

Sesamin extracted from Vismia baccifera var. dealbata was demonstrated to have cytostatic activity on the cancer cell lines tested, particularly the lung cancer cell line, with an IC50 of 1 g/L.

This study investigated the effects of lyophilized mare milk, human milk, and cow colostrum on both human lung cancer cell lines, called A549, and healthy lung cell lines, called MRC5. Mare milk, human milk, and cow colostrum varieties were applied to 6 replicates in both cell lines with lyophilized milk concentrations ranging from 50-3,200 ppm. The cell viability was monitored by optic microscopy and determined by the MTT test. ANOVA and Duncan's multiple range tests were used to analyze data. The results of this study indicated that the most effective milk type on reducing the A549 lung cancer cell line was human milk, followed by mare milk; however, cow colostrum showed little effect. It was observed that human milk and mare milk had anti-proliferative effects on lung cancer cell line at concentrations which were non-toxic to healthy lung cell line.

ABSTRACTIn the recent years, microRNAs (miRNAs) have been discovered to play a very important role in biological processes such as development, differentiation, and apoptosis. The miRNA expression levels in cells are associated with diverse diseases including cancers. MiRNA inhibitors have been widely employed for studying the functions and targets of miRNAs by transfecting the inhibitors into cells. The concentrations of miRNA inhibitors used for such studies can vary depending on the types of miRNAs being tested, the cell lines under study, and the analysis methods. Therefore, in order to obtain accurate results, appropriate amounts of miRNA inhibitors have to be used in the experiments. Apart from amounts, the evaluation of inhibitors may also have to be conducted for functional studies.Here we developed capillary electrophoresis with laser-induced fluorescence (CE-LIF) method for evaluating miRNA inhibitor and for optimizing miRNA inhibitor concentrations, in the A549 lung cancer cell line. The target miRNAs, miRNA-23a and miRNA-24 are biomarker candidates in lung cancer cell lines. Our results showed that miRNA-23a and miRNA-24 were effectively inhibited upon transfection with 20 nM miRNA inhibitors using CE-LIF method. Furthermore, these results demonstrated the potential of CE for fast, specific, sensitive and specific analyses for the evaluation and determination of the optimal concentration of miRNA inhibitors for functional studies.Abstract Graphics

 Objective: Cancer is a molecularly heterogeneous diseaseand the leading cause of death worldwide. The methanol extract of Clerodendrum phlomidis leaf has been reported for cytotoxicity. Hence, the current investigation was planned to evaluate the cytotoxic activity of the chemical constituents isolated from the methanol extract of the C. phlomidis leaf against the Michigan cancer foundation-7 (MCF-7) breast cancer and adenocarcinomic human alveolar basal epithelial cells (A549) lung cancer cell lines by the apoptotic study.Methods: Cytotoxic activity of the chemical constituents of the methanol extract of the C. phlomidis leaf was evaluated by the 3-(4,5-dimethylthiazol- 2-yl)-5-diphenyltetrazolium bromide (MTT) assay against breast cancer (MCF-7) and lung cancer (A549) cell lines. The MCF-7 and A549 cell lines were tested at different concentrations to determine 50% of growth inhibition (inhibitory concentration [IC50]) by MTT assay. Apoptosis of nuclei was detected by 4’,6-diamidino-2-phenylindole staining assay.Results: In MCF-7 breast cancer cell line study, Compounds 6 and 9 exhibited good cytotoxic activity with an IC50 value of 83.80 and 75.16 μg/ml, respectively. In A549 lung cancer cell line, again Compounds 6 and 9 exhibited good cytotoxic activity with an IC50 value of 84.46 and 78.60 μg/ml, respectively. Percentage of apoptosis induced by the Compounds 6 and 9 in the MCF-7 cancer cells was found to be 74.50 and 85.48, respectively.Conclusion: The results of the current study prove that the Compounds 6 and 9 are potential agents for breast and lung cancer. In future research, these potential agents can further be evaluated by animal studies and their mechanism of action apart from the induction of apoptosis of the cancer cells can be determined.

A new styryl lactone, 5 R,6 R-5-hydroxy-6-styryltetrahydropyrane-2-one 2 was isolated from the roots of an endemic Goniothalamus lanceolatus Miq. of Sarawak, Malaysia. Furthermore, seven previously undescribed diastereomers, 5 R,6 R-5-hydroxygoniothalamin 3, 5 R,6 R-5-acetylgoniothalamin 4, 6 S,7 S,8 S-goniodiol-7-monoacetate 5, 6 S,7 S,8 S-goniodiol-8-monoacetate 6, goniofupyrone B 7, deoxygoniopypyrone B 8 and 1 S,5 S,7 R,8 S,3- endo,7- endo-(+)-8- epi-9-deoxygoniopypyrone acetate 9, along with six known styryl lactones (1, 10–15) were also isolated and characterized. 6 S-goniothalamin 1 is reported for the first time from a Goniothalamus species. 1, 11 and 12 showed cytotoxic activity against human colon and lung cancer cell lines with IC50 values ranging from 2.38–7.59 μM.
