Sea pigs (Scotoplanes spp.) are deep-sea dwelling sea cucumbers of the phylum Echinodermata, class Holothuroidea, and order Elasipodida. Few reports are available on the microscopic anatomy of these deep-sea animals. This study describes the histologic findings of two, wild, male and female Scotoplanes sp. collected from Monterey Bay, California. Microscopic findings were similar to other holothuroids, with a few notable exceptions. Sea pigs were bilaterally symmetrical with six pairs of greatly enlarged tube feet arising from the lateral body wall and oriented ventrally for walking. Neither a rete mirabile nor respiratory tree was identified, and the large tube feet may function in respiration. Dorsal papillae protrude from the bivium and are histologically similar to tube feet with a large, muscular water vascular canal in the center. There were 10 buccal tentacles, the epidermis of which was highly folded. Only a single gonad was present in each animal; both male and female had histologic evidence of active gametogenesis. In the male, a presumed protozoal cyst was identified in the aboral intestinal mucosa, and was histologically similar to previous reports of coccidians. This work provides control histology for future investigations of sea pigs and related animals using bright field microscopy.

As part of an ongoing reassessment of the genus Callogorgia, we describe C. lucaya sp. nov. from the western Atlantic, with a distribution so far restricted to the Straits of Florida and northern Caribbean. The species was partially described in an unfinished manuscript of Dr. Frederick Bayer and differs from all other western Atlantic species by commonly lacking outer-lateral body wall scales, having a small number of abaxials, and having body wall sclerites that are externally almost smooth. The oceans make up about 70% of the earth’s surface and serve as habitats for many deep and shallow creatures. In depths of about 50 meters and more, deep-sea corals and sponges occur mostly along seamounts, continental margins, undersea canyons and ridges. They, deep-sea corals and sponges, play a key role in supporting the health of the ocean as they preserve the biodiversity and long-term sustainability of commercial and recreational fish species. With the many benefits that are attached to deep-sea corals and sponges, the Deep-Sea Corals and Research Technology Program (DSCRTP) has been collecting coral and sponge location data from hundreds of remotely operated vehicle (ROV) surveys. However, DSCRTP does not have a spatial representation of the area covered by each ROV while searching for corals and sponges in the deep-sea. A spatial representation would provide critical information to researchers and managers to understand where a survey for corals and sponges has happened, and where a survey is yet to be done in the deep-sea. Therefore, the goal of this study is to create a spatial representation of the ROV surveys that have been collected in Monterey Bay and Hawaii sections of the deep-sea.

Abstract Soft and biological materials are often composed of elementary constituents exhibiting an incessant roto-translational motion at the microscopic scale. Tracking this motion with a bright-field microscope becomes increasingly challenging when the particle size becomes smaller than the microscope resolution, a case which is frequently encountered. Here we demonstrate squared-gradient differential dynamic microscopy (SG-DDM) as a tool to successfully use bright-field microscopy to extract the roto-translational dynamics of small anisotropic colloidal particles, whose rotational motion cannot be tracked accurately in direct space. We provide analytical justification and experimental demonstration of the method by successful application to an aqueous suspension of peanut-shaped particles. Graphic abstract

Holothuria (Mertensiothuria) viridiaurantiasp. nov. is described based on specimens from rocky reefs of northern Chocó in the Colombian Pacific Ocean; however, it also occurs along the Eastern Pacific Ocean from Mexico and Panama. Although specimens from Mexico and Panama were previously identified as Holothuria (Mertensiothuria) hilla Lesson, 1830 the new species is easily distinguished morphologically and via mtDNA. In terms of morphology, the species can be identified by its olive-green background and white-orange papillae and tentacles, larger tentacles with deep indentations and also by larger buttons on the dorsal and ventral body wall, papillae and tube feet; large, thick and rough tentacle rods, and the absence of ossicles in the longitudinal muscles. The new species is included in the subgenus Mertensiothuria considering molecular evidence.

AbstractThe process of copulation in Trichuris globulosa (see Linstow, 1901) Ransom, 1911 has been studied using morphological, histological and scanning electron microscopical techniques (Keilley et al., 1973). The ventral coiling of the posterior part of the male body around the female is achieved by specialized muscles of the ventral body-wall. The single simple pointed spicule emerges with the everting cirrus and acquires a J-shape. The spicule may help in keeping apart the cuticular brim of the non-spiny noneversible part of the vagina during insemination. The cloacal tube is long with a detached internal cuticle which is eversible to the outside as a cirrus. Eversion is brought about by the protrusion of the spicule and contraction of the muscles of the spicule pouch. During eversion part of the proximal cloacal tube is also extruded and forms the globular part of the cirrus. The globular part is covered with spines, which may help in retaining the male and female in copula. The vagina is differentiated into the proximal vagina uterine and distal vagina vera. The vagina vera has a distal eversible spiny part and a proximal noneversible unarmed part. In Trichuris globulosa, the vagina vera is shorter than the spicule.

Pitx2, a bicoid-related homeobox gene, is involved in Rieger's syndrome and the left-right (L-R) asymmetrical pattern formation in body plan. In order to define the genomic structure and roles of Pitx2, we analyzed the genomic structure and generated Pitx2-deficient mice with the lacZ gene in the homeobox-containing exon of Pitx2. We were able to show that among three isoforms of Pitx2, Pitx2c shows asymmetrical expression whereas Pitx2a, Pitx2b and Pitx2c show symmetrical expression. In Pitx2(−)(/)(−) embryos there was an increase in mesodermal cells in the distal end of the left lateral body wall and an amnion continuous with the lateral body wall thickened in its mesodermal layer. These changes resulted in a failure of ventral body wall closure. In lung and heart in which Pitx2 is expressed asymmetrically, right pulmonary isomerism, atrioventricular canals with prominent swelling, and juxtaposition of the atrium were detected. The hearts failed to develop tricuspid and mitral valves and a common atrioventricular valve forms. Further, dysgenesis of the Pitx2(−)(/)(−) extraocular muscle and thickening of the mesothelial layer of cornea were observed in the ocular system where Pitx2 is expressed symmetrically, and these resulted in enophthalmos. The present study shows that Pitx2 expressed in various sites participates in morphogenesis through three types of actions: the involvement of asymmetric Pitx2 expression in the entire morphogenetic process of L-R asymmetric organs; the involvement of asymmetric Pitx2 expression in the regional morphogenesis of asymmetric organs; and finally the involvement of symmetric Pitx2 expression in the regional morphogenesis of symmetric organs.

Sclerothyone Thandar, 1989, currently restricted to two South African species, is here expanded to include two new species from off coast of Brazil. These new species expand the distribution of the genus to the southwestern Atlantic Ocean. Sclerothyone reichi sp. nov. is characterized by having body wall with multilocular two pillared tables with multiperforate disc and short spire, ending in a narrow crown of teeth; tentacles with rods and introvert with two pillared tables and rosettes; tube feet with only end plates. Sclerothyone oloughlini sp. nov. is distinctive in having a body wall with multilocular, two pillared tables; handles present and multilocular two-pillared tables; circular disc without handle; and plates (near the anus). The new species resemble each other in the tentacles with rods. However, Sclerothyone oloughlini sp. nov. stands apart from S. reichi sp. nov. in the introvert with rosettes; tube feet with two pillared supporting tables, arched plates and end plates. The possible assignment of Cucumaria nozawai Mitsukuri, 1912 and Thyone adinopoda Pawson & Miller, 1981 to Sclerothyone, and the position of Thyone neofusus Deichmann, 1941 in Temparena Thandar, 1989, are briefly discussed. A key to all genera and species of Sclerodactylinae is provided.

Rabbits were infected per os with 10 Eustrongylides ignotus L4 and with 50 Contracaecum multipapillatum L3 per rabbit, recovered from naturally infected freshwater fishes (Hoplias malabaricus) in order to evaluate the patogenicity of these two nematode species in mammalian host. Two rabbits (20%) infected with E. ignotus died before the fourth day post-inoculation (one after 51 and the other after 78 hours). Six rabbits (60%) were inappetent until the fifth day following experimental inoculation. No clinical signs in rabbits inoculated with C. multipapillatum were observed; nevertheless, eight (80%) animals were positive for this nematode species. Rabbits inoculated with E. ignotus, had gastric congestion with hematoma of the gastric wall in 60% of the cases. Peritoneum was congested in 20% of the animals with the presence of peritoneal abscess in 10% of the cases. All inoculated animals showed hyperemia of the gastric mucosa with hemorrhagic gastritis due to infections with E. ignotus. In C. multipapillatum inoculated animals, the hyperemia was followed by disruption of the epithelial mucosa in the sites of parasite attachment. In the gastric mucosa, miscellaneous leukocitary infiltrates, with multifocal necrosis reaching the submucosa in the infections with C. multipapillatum were observed under bright field microscopy. Perforating lesions in several organs, mainly in the gastric wall, pancreas and liver, always in the presence of a mixed inflammatory process, intensely fibrous, with hemorrhage and necrosis were observed in animals infected with E. ignotus.
