ABSTRACT DNA supercoiling in the chloroplast of the unicellular green algaChlamydomonas reinhardtii was found to change with a diurnal rhythm in cells growing in alternating 12-h dark–12-h light periods. Highest and lowest DNA superhelicities occurred at the beginning and towards the end of the 12-h light periods, respectively. The fluctuations in DNA supercoiling occurred concurrently and in the same direction in two separate parts of the chloroplast genome, one containing the genes psaB, rbcL, andatpA and the other containing the atpB gene. Fluctuations were not confined to transcribed DNA regions, indicating simultaneous changes in DNA conformation all over the chloroplast genome. Because the diurnal fluctuations persisted in cells kept in continuous light, DNA supercoiling is judged to be under endogenous control. The endogenous fluctuations in chloroplast DNA topology correlated tightly with the endogenous fluctuations of overall chloroplast gene transcription and with those of the pool sizes of most chloroplast transcripts analyzed. This result suggests that DNA superhelical changes have a role in the regulation of chloroplast gene expression in Chlamydomonas.

AbstractBecause the motives behind goal-directed behaviors are often complex, most behaviors result from the interplay between different motives. However, it is unclear how this interplay between multiple motives affects the neural computation of goal-directed behaviors. Using a combination of drift-diffusion modeling and fMRI, we show that the interplay between different social motives changes initial preferences for prosocial behavior before a person makes a behavioral choice. This increase in preferences for the prosocial choice option was tracked by neural responses in the bilateral dorsal striatum, which in turn lowered the amount of information necessary for choosing prosocial behavior. We obtained these results using a paradigm in which each participant performed the same behavior based on different, simultaneously activated motives, or based on each of the motives separately. Thus, our findings provide a model of behavioral choice computation in complex motivational states, i.e., the motivational setting that drives most goal-directed human behaviors.

The present study investigated whether ambiguity tolerance relates to personality traits that are theoretically grounded in fear (neuroticism) or attraction (openness to experience; extraversion) for the unknown. Our hypotheses were supported for self-report measures (and openness to experience predicted ambiguity tolerance controlling for intelligence), but behavioral choice measures of ambiguity tolerance demonstrated poor reliability and were unrelated to self-reported ambiguity tolerance and basic personality traits. An exploratory network analysis revealed that ambiguity tolerance was more strongly related to the intellectual curiosity (vs. aesthetic appreciation) facet of openness to experience, and the assertiveness (vs. energy or sociability) facet of extraversion. Our findings reinforce the fragmented literature in this area, and support predictions derived from psychological entropy theories of personality.
