Efficacy of different Neem (Azadirachta indica) parts in protecting susceptible soybean cultivar against parasitic nematode Meloidogyne javanica

ABSTRACT Management of nematodes is a very hectic job due to a highly diverse group of organisms. To find lead compounds for new nematicide development, five metabolites (1–5) were isolated from the culture broth of Chaetomium globosum YSC5 and tested for nematicidal activities against the second stage juveniles (J2s) of Meloidogyne javanica. The results revealed that chaetoglobosin A (1), chaetoglobosin B (2) and flavipin (3) exhibited strong adverse effects (91.6, 83.8 and 87.4%, respectively) on J2 mortality at 200 μg/mL with LC50 values of 88.4, 107.7 and 99.2 μg/mL after 72 h, respectively, while 3-methoxyepicoccone (4) and 4,5,6-trihydroxy-7-methylphthalide (5) showed moderate effects (78.0 and 75.5%, respectively) with LC50 values of 124.0 and 131.6 μg/mL, respectively. Furthermore, in pot assay compounds 1 and 2 appeared to be promising metabolites at 200 μg/mL that significantly reduced nematode reproduction and showed a positive influence on plant growth. Our findings could be helpful for development of new potential bio-based pesticides for integrated management of plant-parasitic nematode.

ABSTRACT Root-knot nematodes, Meloidogyne spp., and the root-lesion nematode, Pratylenchus brachyurus, are the most common phytoparasites in soybean crop, which usually occur in association in crop areas. However, few studies were conducted on the interaction between these parasites. Therefore, this study aimed to assess the interference of the increase in M. javanica population in the reproduction of P. brachyurus, and the effect of mixed populations on soybean (cultivar BRS/MT Pintado) development. The experiment consisted in inoculating a population of 1000 specimens of P. brachyurus per plant and changing the inoculum level of M. javanica, using zero, 1000, 2000 and 4000 eggs, in two different periods. After 65 days of inoculation, the increase in the initial population of M. javanica reduced by 79 and 73% the final population of P. brachyurus in the different experiments. However, both species significantly increased their populations. When subjected to the same inoculum level, M. javanica was more efficient in reproducing than P. brachyurus. The increase in the number of nematodes reduced the vegetative growth of the plant.

Meloidogyne enterolobii has become an economically important plant parasitic nematode worldwide because of its high aggressiveness, increasing geographic distribution, wide host range, and pathogenicity in pepper (Capsicum annuum) cultivars carrying resistance genes to Meloidogyne incognita, Meloidogyne arenaria, and Meloidogyne javanica. The objectives of this study were to identify landraces of peppers resistant to M. enterolobii and analyze the relationship between resistance indicators and the phenotype parameters of plant height, stem width, leaf length, leaf width, relative chlorophyll, and number of flowers. Ninety landraces of C. annuum were collected from several states of Mexico and were inoculated with 2,000 eggs of M. enterolobii. Eleven resistant landraces were selected and confirmed with a second inoculation experiment. Seventy-five days after inoculation, in both experiments, the resistance of landraces UTC66, UTC90, UTC67, UTC88, and UTC81 to M. enterolobii was consistent. Although genotypes UTC24, UTC79, UTC65, UTC68, UTC69, and UTC25 were susceptible, these landraces had a significantly higher proportion of resistant plants, less root galling, and a lower reproductive index, in comparison with the rest of the 79 genotypes and the susceptible control, which were highly susceptible in both experiments. There was no correlation between resistant indicators and phenotypic parameters, although plant height, relative chlorophyll, and number of flowers were in general significantly affected compared with noninoculated controls, indicating that the nematodes reduce the growth and yield of peppers. Results indicate that all resistant plants from these landraces are promising sources of resistance for the development of pepper cultivars resistant to M. enterolobii.
