Due to the harsh offshore exploitation environment, students are unable to conduct field investigation in offshore platforms. In this scenario, virtual simulation is an effective way to solve this problem. This paper introduces in detail a Virtual Simulation Experimental Software of Marine Platform Structure, which is closely integrated with the course content and engineering reality. Through the operation and learning of platform site selection, platform structure assembly, platform structure display, can realize the students' knowledge of platform structure composition mastery, and at the same time improve the students' participation and motivation. In addition, the system can automatically generate the experiment report and give the operation score reference to realize the teacher's quality control of the experiment course.

For representing the coupling relationships among various correlate elements in the formation process of assembly qualities, the mapping relations between the structure domain and the quality domain was established on the basis of analyzing the hybrid structure assembly. Through classifying the quality characteristics, the quality domain was divided into two sub-sets which are quality control points set and quality attributes set. Defining the relevant elements in the model, such as vertex, arc, etc by the theory of network flow, the coupling relations among various quality control points in the formation process of assembly qualities were reflected. According to the sequence of quality control points entering into assembly environment, their corresponding hierarchy in the model were determined, which presents the timing sexual of the formation process of assembly qualities. At last, a correlation-model between quality attributes and quality control points in the assembly process of complex product based on network flow was established, which would be the theoretical basis for the dynamic tolerance optimization of quality control points. An example of an automobile active gear axle assembly was given to demonstrate the feasibility of the correlation-model.

The construction of offshore oil and gas development platform is a system engineering with enormous cost, in order to make rational use of limited resources to achieve maximum economic benefits, based on the industrial site selection problem and with the analyses of decision variable in the process of offshore platforms site selection decision-making, Objective function and the Constraint condition a Bi-level Programming Model Of offshore platforms Site selection with a stochastic oil price are set up. According to the characteristics of the model, an adaptive genetic algorithm to solve the problems and a tabu search algorithm to solve the lower planning are designed to improve the Optimize efficiency and effect of the model. With the Optimize of the process, the problems such as Platform deliverability of the process of offshore oil platforms site, Cost, Optimal distribution of resources and so on are successfully resolved.

Based on the field investigation for secondary geological hazards induced by 5.12 Wenchuan Earthquake, the types of geological disasters during or after earthquake are analyzed in this paper. The main secondary geological hazards are rock fall and collapse, landslide, debris flow, dammed lake,sand liquefaction and so on. The development and distribution rules of secondary geo-hazards, with a long-term sustainability and a formation of hazard chains, are of relation with earthquake fault zone, and consistent with seismic intensity. The buildings located in secondary geo-hazards areas with a high risk are damaged severely, thus the site selection for post-earthquake reconstruction and related geotechnical engineering issue are discussed, which are useful in reconstruction of Wenchuan earthquake area.

AbstractLatent 5’ splice sites are highly abundant in human introns, yet, are apparently not normally used. Splicing at most of these sites would incorporate in-frame stop codons generating nonsense mRNAs. Importantly, under stress and in cancer, splicing at latent sites is activated generating nonsense mRNAs from thousands of genes. Previous studies point to an unresolved RNA quality control mechanism that suppresses latent splicing independently of NMD. They further demonstrated a pivotal role for initiator-tRNA in this mechanism, through its interaction with the AUG codon, independent of its role in protein translation. To further elucidate this mechanism, here we searched for nuclear proteins directly bound to initiator-tRNA in the nucleus. We identified nucleolin (NCL), a multifunctional, abundant, and conserved protein, as a novel regulator of splice site selection. Starting with UV crosslinking, we show that NCL is directly and specifically interacting with initiator-tRNA in the nucleus, but not in the cytoplasm. In support of NCL involvement in this mechanism, we show activation of latent splicing in hundreds of transcripts upon NCL knockdown, disrupting gene transcripts involved in several important cellular pathways and cell metabolism functions (e.g. transcription factors, oncogenes, kinases, splicing factors, translation factors, and genes affecting cell motility, proliferation, and cellular trafficking). We thus propose NCL, a component of the endogenous spliceosome, through its direct interaction with initiator-tRNA and its effect on latent splicing as the first documented protein of a nuclear quality control mechanism that regulates splice site selection to protect cells from latent splicing that would generate defective mRNAs.

AbstractLiterally translated as 'wind and water', Fengshui has been used as a special and important tool to harmonize people with their built environment and with the surrounding natural world. By examining some aspects of Fengshui as it was employed in traditional villages in southeast China, this paper aims to explore how Fengshui influences the interactions of Chinese people with their natural world. It focuses on two aspects of the employment of Fengshui in the outer spaces of villages in Southeast China: site selection and the entrance organisation. The methodology employed in the research involved both field investigation and archival study with a special emphasis on the historical records of family trees in the villages of Southeast China. Some modern psychological and architectural principles are borrowed as a frame of reference for analysing the Fengshui models. This analysis concludes that Fengshui satisfies not only physical needs, but also environmental, psychological and aesthetic ones. It demonstrates one of the most distinctive and sensitive treatments to aspects of the natural environment and landscape. It has been able, therefore, to improve the quality of the environment.

During translation initiation, 40S ribosomes scan the mRNA until they encounter the start codon, where conformational changes produce a translation-competent 80S complex. Destabilizing the scanning complex results in misinitiation at non-AUG codons, demonstrating its importance for fidelity. Here, we use a combination of biochemical and genetic analyses to demonstrate that the ability of the nascent subunit to adopt the scanning complex is tested during assembly via structural mimicry. Specifically, formation of the 80S-like assembly intermediate, which structurally resembles scanning complexes, requires the correct folding of two rRNA elements in the subunit head and the proper positioning of the universally conserved head proteins Rps3, Rps15, Rps20, and Rps29. rRNA misfolding impairs the formation of 80S-like ribosomes, and bypass of individual checkpoints using cancer-associated mutations produces ribosomes defective in accurate start-site selection. Thus, the formation of 80S-like assembly intermediates is a quality control step that ensures scanning competence of the nascent subunit.
