Carnation is a popular floricultural crop grown widely for its attractive cut flowers. Micro-propagation can be used to create large-scale carnation output. For growth and development, plants require some necessary nutrients as well as growth regulators. Due to the importance of carnation, the present work is carried out using leaf and nodal segments to examine the potential of several plant growth regulators for in vitro callus formation and adventitious shoot regeneration. Explants were sterilized properly with bavistin, sodium hypochlorite and mercuric chloride. The minor contaminated cultures were created by consecutively treating the explants with 0.25% bavistin, 0.50% sodium hypochlorite, and 0.1% mercuric chloride for ten, fifteen, and two minutes. MS media with 2.5 mg/l 2,4-dichlorophenoxy acetic acid (2,4-D) in combination with 0.75 mg/l naphthalene acetic acid (NAA) resulted in the maximum callus induction (90.47%) from leaf explants. Maximum shoots (76.47%) were produced in MS media supplemented with 2.0 mg/l Thidiazuron (TDZ) + 0.25 mg/l NAA. NAA at 1.25 mg/l was most efficient for maximum root induction (83.32%). In the present study, an effective protocol of carnation explants sterilization was optimized for successful callusing and shoot regeneration.

The influence of growth regulators, explants and their interactions on in vitro shoot bud formation from callus was studied in <I>Dianthus caryophyllus</I> L. The leaf and internode explants were cultured on Murashige and Skoog (MS) medium containing different concentrations of growth regulators. The highest callus induction was observed with 2 mg/l 2,4-dichlorophenoxy acetic acid (2,4-D) and 1 mg/l benzyl adenine (BA). Out of twenty seven shoot regeneration media tested, only 2 mg/l thidiazuron (TDZ) and zeatin alone or in combination with naphthalene acetic acid (NAA) and/or indole acetic acid (IAA) could differentiate calli. The highest average number of shoots was observed with 2 mg/l TDZ and 1 mg/l IAA. Significant differences were observed in calli producing shoots and number of shoots per callus in the explants of leaf and internode. The shoots were elongated and multiplied on MS medium supplemented with 1 mg/l BA and solidified with 1% agar. The shoots were rooted and hardened with 76% survival success in pots after six weeks of transfer to the pots.

The experiment was designed to evaluate the effect of growth regulators on leaf explant of Gerbera for callus induction. Various kinds of plant growth regulators such as 6-Benzylaminopurine (BAP), α-Naphthalene acetic acid (NAA), 2, 4-Dichlorophenoxyacetic acid (2, 4-D), Indole-3-acetic acid (IAA) were used to initiate cultures. These were added to Murashige and Skoog medium in different combinations and concentrations. Leaf explants cultured on MS medium supplemented with BAP+ 2, 4-D+ IAA in T4 treatment & BAP+ 2,4-D in T5 treatment showed the best results for callus induction. On the other hand callus was induced early in the combination of BA+ 2,4-D + IAA hormone in T5, T9 & T8 treatment respectively. The rate of callus induction was very low in BA + NAA combinations but it was much earlier.

Lulo is a species of great importance to the fruticulture of Colombia, but has significant phytosanitary problems that require an aggressive breeding program oriented toward the production of genotypes with tolerance to phytopathogens. These programs need to establish highly efficient mass plant propagation protocols, such as somatic embryogenesis. This study focused on research on the somatic embryogenesis of lulo using kinetin, naphthalene acetic acid-NAA (Plant Growth Regulators, PGRs), and different sucrose concentrations in a MS medium. Two lulo varieties, Solanum quitoense var. septentrionale and S. quitoense var. quitoense, and two explant types (hypocotyl and cotyledon) were used, incubated in dark conditions at 25±2°C. The highest production percentage of the embryos was obtained when 50 mM of NAA were added to the medium with sucrose (50.0 and 263.1 mM) for the two explant types used. In lulo with spines, the highest percentage of embryonic structures (50%) was observed with cotyledonary leaf explants and 50 mM of NAA ; while in the spineless lulo, the embryonic structures were observed in the same type of explant with 50 mM of NAA + 263.1 mM of sucrose (32%).

One of the goals of the experiment is to standardization of HgCl2 treatment for explants sterilization. The objectives also include developing a reproducible cost effective protocol for large scale production of Solanum tuberosum of Cardinal variety plantlets from selectively better clones through plant in vitro propagation methods. Selection of growth regulators for proper multiple shoots regeneration, elongation and root induction. To produce genetically uniform plantlets within a short time capable surviving in natural condition raised in in vitro environment. Shoot tip and nodal segment explants from field grown plants were used as experimental materials in this investigation. All explants were cultured on Murashige and Skoog medium supplemented with various plant growth regulators. For surface sterilization of explants, HgCl2 (0.1%) for 2 minutes was found to be most effective for complete destroying of surface pathogens and getting healthy tissues. Shoot regeneration was observed from both shoot tips and nodal explants for the studied plant. Maximum number of shoot per culture (17) was recorded and it also obtained the highest average length of the shoot (5cm) in Murashige and Skoog medium containing no hormone. On the other hand 6-benzyl amino purine (0.2mg/l) in 3 media showed the highest rate of shoot multiplication (73%) and the highest average length (4cm). In case of Gibberellic acid (0.1mg/l) in Murashige and Skoog media showed its highest rate of shoot regeneration (82%) and the highest average length (4.5cm). From the overall experiment it was observed that shoot tips are more responsive for micro propagation. In root induction Murashige and Skoog medium supplemented with different concentration (0.5, 1, 1.5 and 2mg/l) of indol-3-acetic acid and kinetin. Indol-3-acetic acid and kinetin (1.5+1.5 mg/l) showed its lowest rate of root regeneration (40%) and the average length of the root (1.5 cm). On the contrary Murashige and Skoog medium with no hormone showed the rate of root regeneration (96%) and the highest average length of the root (2.5 cm). Experiments were conducted to establish an efficient protocol for micropropagation of Chirita longgangensis W.T. Wang. Somatic embryos formed directly at the cut edges of C. longgangensis leaf explants on Murashige and Skoog (MS) medium supplemented with benzylaminopurine (BA) and α-naphthalene acetic acid (NAA). During the somatic embryo induction stage, leaf explants and basal leaf explants were used. Leaves were more appropriate explants than the basal leaf explants. The best medium was modified MS macronutrients and micronutrients supplemented with 0.5 mg·L−1 BA and 0.1 mg·L−1 NAA (the best mean number of somatic embryos per explants was 24.10 ± 1.63). The second stage was root induction and elongation. In vitro regenerated plantlets rooted best on MS medium containing 0.1 mg·L−1 indole-3-acetic acid (IAA) and 30 g·L−1 sucrose. Rooted plantlets, following acclimatization in a greenhouse, were successfully transferred to field conditions, and 95% of the plants survived. Application of this protocol has the ability for mass multiplication, in a limited time, of the endangered species C. longgangensis.

The experiment was carried out to study the effect of genotypes and growth regulators on root induction and shoot regeneration of brinjal (Solanum melongena) genotypes. Leaf segments of three varieties of Solanum were cultured on MS medium with different concentrations and combinations of plant growth regulators. Among the tested genotypes, Protab showed highest percentage of shoot regeneration (65.67%). Early and maximum rate of regeneration was found in MS + 1 mg/L NAA (naphthalene acetic acid) + 0.1 mg/LBAP (6-benzylamino purine) for all the genotypes. The highest number of roots per shoot was counted in Protab (73.33%) on ½MS + 2 mg/L IBA (indole butyric acid) + 0.4 mg/L BAP. Based on the overall performance, the variety Protab appeared as the best for shoot and root formation and ultimately successful regeneration of plants.Res. Agric., Livest. Suitable protocol for induction of callus and regeneration was developed from different explants viz., node, stem and leaves in Physalis minima. MS basal medium supplemented with various concentrations (1.0-4.0mg/l) of auxins like 2,4-Dichlorophenoxy acetic acid (2,4-D), α-naphthalene acetic acid (NAA) and Indole-3-acetic acid (IAA) and cytokinins (0.5-1.5mg/l) like BAP or Kn were used. All the three explants responded for induction of callus, however stem explants were found superior, followed by node and leaf. Callus induction was observed in all the auxins and combination of growth regulators used with varied mass (2010±1.10) and highest percentage of callus induction was observed from stem at 2.0mg/l 2,4-D (90%) followed by NAA (70%) and IAA (50%). Organogenesis was induced when nodal explants were transferred on MS medium supplemented with 2,4-D and Kn at various concentrations, maximum being on 2.0mg/l 2,4-D + 1.0mg/l Kn (90%). Regenerated shoots were elongated on 0.5mg/l GA3. The shoots were subsequently rooted on MS + 1.0mg/l IBA (95%) medium. Rooted shoots were hardened and acclimatized, later they were transferred to polycups containing soil, cocopeat and sand in the ratio 1:2:1.Keywords:Physalis minima, Node, Stem, Leaf, callus and growth regulators.

Digitalis trojana Ivanina is a member of the Plantaginaceae family and known by its common name, Helen of Troy foxglove. It is perennial endemic to Çanakkale and Balıkesir, northwestern Turkey. In order to develop an efficient shoot regeneration protocol, the leaf explants of D. trojana were cultured on Murashige and Skoog (MS) medium containing 6-benzyl adenine (0.1, 0.5, 1.0, 3.0, 5.0 mg/L) and α-naphthalene acetic acid (0.1, 0.5, 1.0 mg/L), 3% (w/v) sucrose and 0.8% (w/v) agar. The highest number of regenerated shoots was obtained from leaf explants that were cultured on MS medium with 3.0 mg/L BA+0.1 mg/L NAA. Regenerated shoots were rooted on MS medium without plant growth regulators. Rooted plants (2–3 cm) were separately transferred to pots containing a mixture of peat and perlite (2:1 v/v) and acclimatized successfully in a growth chamber.

An efficient in vitro regeneration system was developed for Rauvolfia serpentina L. through direct and indirect organogenesis from nodal and leaf explants. Among the different growth regulators, MS medium supplemented with 2.0 mg/l BAP, 0.5mg/l IAA and 0.02mg/l NAA found best for the multiple shoot formation from nodal segments. In this combination 98% explants produced multiple shoots and the average number of shoots per explants is 13∙4. The frequency of callus induction and multiple shoot induction from leaves was highest 88% in MS medium supplemented with 2.0 mg/l BAP, where mean number of shoots/explants was 12.5. The highest frequency of root induction (80%) and mean number of roots/plantlets (10) were obtained on half strength of MS medium containing 0.2 mg/l IBA. The study focused on the influence of the plant growth regulators (PGRs) benzyladenine (BA) and naphthalene acetic acid (NAA) on the production of volatile organic compounds (VOCs) from the flowers of two modern rose varieties, Hybrid Tea and Floribunda. Thirty-six plants of Hybrid Tea and Floribunda were tested. Benzyladenine and naphthalene acetic acid were applied at 0, 100 and 200 mg/L to both rose varieties. Gas chromatography, coupled with flame ionization detection and mass spectrometry, was used to analyze and identify the volatile organic compounds from the flowers. A three-phase fiber 50/30 µm divinylbenzene/carboxen/polydimethylsiloxane was used to capture VOCs, at 2, 4 and 8 weeks, and 4 weeks was selected as it had the highest peak area. In total, 81 and 76 VOCs were detected after treatment of both rose varieties with BA and NAA, respectively. In addition, 20 compounds, which had significant differences between different treatments, were identified from both rose varieties. The majority of VOCs were extracted after the application of 200 mg (BA and NAA) /L of formulation, and four important compounds, cis-muurola-4(141)5-diene, y-candinene, y-muurolene and prenyl acetate, increased significantly compared to the controls. These compounds are commercially important aroma chemicals. This study used the rapid and solvent-free SPME method to show that BA and NAA treatments can result in significant VOC production in the flowers of two rose varieties, enhancing the aromatic value of the flowers. This method has the potential to be applied to other valuable aromatic floricultural plant species.
