Centriole maturation is essential for ciliogenesis, but which proteins and how they regulate ciliary assembly is unclear. In this issue, Kumar et al. (2021. J. Cell Biol. https://doi.org/10.1083/jcb.202011133) shed light on this process by identifying a ciliopathy complex at the distal mother centriole that restrains centriole length and supports the formation of distal appendages.

The ARP2/3 complex promotes branched actin networks, but the importance of specific subunit isoforms is unclear. In this issue, Galloni, Carra, et al. (2021. J. Cell Biol. https://doi.org/10.1083/jcb.202102043) show that MICAL2 mediates methionine oxidation of ARP3B, thus destabilizing ARP2/3 complexes and leading to disassembly of branched actin filaments.

In this mSphere of Influence article, he reflects on how the papers “A promiscuous biotin ligase fusion protein identifies proximal and interacting proteins in mammalian cells” (K. J. Roux, D. I. Kim, M. Raida, and B. Burke, J Cell Biol 196:801–810, 2012, https://doi.org/10.1083/jcb.201112098) and “Proteomic mapping of mitochondria in living cells via spatially restricted enzymatic tagging” (H.-W. Rhee, P. Zou, N. D. Udeshi, J. D. Martell, et al., Science 339:1328–1331, 2013, https://doi.org/10.1126/science.1230593) impacted his thinking on how to determine where proteins from intracellular pathogens are located within host cells.
