AbstractRhizopus oryzae (R. oryzae) can effectively produce organic acids, and its pellet formation in seed cultures has been shown to significantly enhance subsequent fermentation processes. Despite advances in strain development, simple and effective methods for inducing pellet morphology and a basic understanding of the mechanisms controlling this process could facilitate substantial increases in efficiency and product output. Here, we report that 1.5% triethanolamine (TEOA) in seed culture medium can activate the growth of R. oryzae spores in compact and uniform pellets which is optimal for fermentation conditions. Analysis of fermentation kinetics showed that the production of fumaric and L-malic acid increases 293% and 177%, respectively. Transcriptomic analysis revealed that exposure of R. oryzae to 1.5% TEOA during the seed culture activated the phosphatidylinositol and mitogen-activated protein kinase signaling pathways. Theses pathways subsequently stimulated the downstream carbohydrate-active synthases and hydrolases that required for cell wall component synthesis and reconstruction. Our results thus provide insight into the regulatory pathways controlling pellet morphology germane to the viability of seed cultures, and provide valuable reference data for subsequent optimization of organic acid fermentation by R. oryzae.

Although Haematococcus lacustris has been developed for astaxanthin production for decades, the production cost is still high. In order to modify the production processes, we proposed a novel strategy of cultivation, featured by sequential indoor continuous mixotrophic cultivation for the production of green cells followed by outdoor phototrophic induction for astaxanthin accumulation. The continuous mixotrophic cultivation was first optimized indoor, and then the seed culture of mixotrophic cultivation was inoculated into outdoor open raceway ponds for photoinduction. The results showed that mixotrophically grown cultures could efficiently grow without losing their photosynthetic efficiency and yielded higher biomass concentration (0.655 g L−1) and astaxanthin content (2.2% DW), compared to phototrophically grown seed culture controls. This novel strategy might be a promising alternative to the current approaches to advance the production technology of astaxanthin from microalgae.

Sedirea japonica is becoming endangered, and even extinct, due to habitat destruction and illegal collection, and the development of an optimized artificial propagation system is necessary for its conservation and reintroduction. Thus, the effects of plant growth medium strength (Murashige and Skoog (MS) and Hyponex media) and the addition of activated charcoal (AC) and organic supplements on seedling growth of S. japonica were investigated through in vitro seed culture. The results showed that seedling growth was higher in half-strength (1/2) media than in full-strength media. After the addition of AC, the highest leaf area (2.14 cm2) was recorded in the seedlings grown in 1/2 Hyponex medium, and after the addition of organic supplements, root development increased regardless of the media type. Among the sixteen suitable media tested at later seedling growth stages, 1/2 MS medium with the addition of 0.6 g·L−1 AC, 30 g·L−1 banana homogenate and 10 g·L−1 apple homogenate was generally effective in fresh weight (6.13 g) and root length (9.59 cm). We demonstrated which organic supplements are preferred for in vitro growth of seedlings developed from S. japonica protocorms by asymbiotic seed culture, which can be used for mass production and conservation of this rare epiphytic orchid.
