We have developed and used high-time-resolution, single-molecule field-effect transistors (smFETs) to characterize the con-formational free-energy landscape of RNA stem-loops. Stem-loops are some of the most common RNA structural motifs and serve as building blocks for the formation of more complex RNA structures. Given their prevalence and integral role in RNA folding, the kinetics of stem-loop (un)folding has been extensively characterized using both experimental and computational approaches. Interestingly, these studies have reported vastly disparate timescales of (un)folding, which has been recently in-terpreted as evidence that (un)folding of even simple stem-loops occurs on a highly rugged conformational energy landscape. Because smFETs do not rely on fluorophore reporters of conformation or on the application of mechanical (un)folding forces, they provide a unique and complementary approach that has allowed us to directly monitor tens of thousands of (un)folding events of individual stem-loops at a 200 μs time resolution. Our results show that under our experimental conditions, stem-loops fold and unfold over a 1-200 ms timescale during which they transition between ensembles of unfolded and folded conformations, the latter of which is composed of at least two sub-populations. The 1-200 ms timescale of (un)folding we observe here indicates that smFETs report on complete (un)folding trajectories in which relatively extended unfolded con-formations of the RNA spend long periods of time wandering the free-energy landscape before sampling one of several mis-folded conformations or, alternatively, the natively folded conformation. Our findings demonstrate how the combination of single-molecule sensitivity and high time resolution makes smFETs unique and powerful tools for characterizing the con-formational free-energy landscape of RNA and highlight the extremely rugged landscape on which even the simplest RNA structural elements fold.

Abstract A revolutionary, film theorist, and screenwriter, Xia Yan (1900–1995) is known as one of the pioneers of Chinese cinema. Xia’s pursuit of a national style and international status for Chinese cinema and his aspirations for the prosperity of the Chinese nation are in line with the basic ideas and goals of the nascent “Chinese School of Film.” In the context of the under-theorized and problematic production practices of current Chinese cinema, it is high time to revisit Xia’s professional and academic contributions to cinematic art, which shed light on the construction of both the “Chinese School of Film” and “shared aesthetics.”

Abstract The last 50 years have witnessed ecolinguistics come into bloom as a mature domain. This paper aims to examine the half-century development of ecolinguistics by reviewing its backgrounds, definitions, strands, and approaches, and also briefly previewing its future horizons. The birth of ecolinguistics can be attributed to such ecological necessities as the ecological crisis as an essential root, and an ecological perspective for linguistics as a linguistic necessity, together with six ecolinguistic turns in this domain (Section 2). Since the emergence of ecolinguistics in the 1970s, various definitions for ecolinguistics as an evolving concept have come into being, involving the geographical, conceptual, disciplinary, methodological, and practical sides (Section 3). Figures who have contributed to the development of this domain can be divided into old strands like Haugenian and Hallidayan ecolinguistics, as well as new strands such as strong ecolinguistics and the latest radical embodied ecolinguistics (Section 4). Given the diverse definitions and strands, a set of approaches have taken shape, ranging from the Haugenian approach to ecological discourse analysis (Section 5). Due to major problems found in reviewing four parts of ecolinguistics, it is high time three shifts in perspective be put into effect in ecolinguistics that can promise its future horizons.

2020 has been marked by a ‘once in a century crisis' that influenced the dynamics of the globe deeply. Soon after the COVID-19 pandemic, most daily practices had to be transferred to online platforms as humanity was supposed to adopt social and physical distancing to avoid the risk of infection. Even technologically illiterate people were abruptly charged with online tasks as part of their jobs or responsibilities. It suddenly turned out to be high time to go online and have a digital identity to keep pace with the new normal life. Thus, internet has taken its place among the basic needs more specifically than before. This chapter is about the technology-driven supervisory social credit system which is said to have contributed to Chinese state to manage the COVID-19 crisis in a short time. Exploring the foundations, motives, and highlights of the system, this chapter proposes a framework for a potential digital governance model coined as the Cyber Leviathan and bears importance in terms of crisis management.

The most probable scenario for the development of experimental high-energy physics in the next 50 years is the creation of a family of Future Circular Colliders (FCC) at CERN, a Circular Electron–Positron Collider at China, and a Future Electron-Ion Collider at Brookhaven (USA), which continue the Large Hadron Collider (LHC) scientific program within the framework of the Standard Model and beyond it. The first generation of colliders to be put into operation will utilize the electron beam as one of the colliding species to provide precise mass spectroscopy in a wide energy range. Similarly to the measurements at the high luminosity phase of the LHC operation, the most important property of the detectors to be used in the experimental setup is a combination of the short response of the detectors and their high time resolution. The radiation tolerance to a harsh irradiation environment remains mandatory but not the main factor of the collider’s experiments using electronic beams. A short response in combination with high time resolution ensures minimization of the influence of the pile-up and spill-over effects at the high frequency of collisions (higher than 50 MGz). The radiation hardness of the materials maintains the long-term high accuracy of the detector calibration. This paper discusses the prospects for using modern inorganic scintillation materials for calorimetric detectors at future colliders.

Psychiatrization not only affects adults. Ever more children in Western countries are being diagnosed with a mental disorder of behavior, such as ADHD. Children may often be labelled with the best intentions, for example in order to be able to provide them with suitable care and guidance. However, this labelling can have exclusionary effects and often entails the consequence that important discussion about contextual factors that give rise to (the perception of) unwelcome behavior or academic underperformance rarely, if at all, takes place. In this article we contend that although children are of central concern to schools and the design of pupils’ education, it is important not to make pupils the sole owner of problems that arise. It is therefore high time that a far more critical normative stance towards inclusive education is taken, in which the presently widespread biomedical approach is met with a school community response that focuses not on the nature of individual disorders but on the special need for additional capacity that schools and teachers have in meeting (perceived) deviant behaviors and emotions and/or academic underperformance. We argue that teaching should not set out to remedy individual diagnoses, but that teachers should be supported to extend their professional competence to the benefit of all pupils.
