With the aim of obtaining potential IL-6 inhibitor for the treatment of rheumatoid arthritis, four 3-aryloxylindolizine derivatives were synthesized from the 2-bromopyridine and aryl propargyl ether through the Sonogashira coupling/5-endo-trigcycloisomerization domino strategy. These compounds were characterized by NMR, IR, EI-MS and elemental analysis. Their biological activities were evaluated by the bacterial lipopolysaccharide (LPS) stimulation of mouse cells on the RAW264.7 inflammation model. All the compounds revealed good anti-inflammatory activities with the inhibitions of 71~82% against IL-6 at 100 μM. The bioactive 3-aryloxylindolizines might be used as potential anti-inflammatory drugs.

Various heterocyclic compounds along their derivatives were evaluated for their biological activities as antiviral, antitumor, anticonvulsant, antibacterial, antifungal, antituberculosis, analgesic, anti-inflammatory, antidiabetic, antihistamine and other biological activities. The triazole moiety seems to be very small, but in the biological profile has attracted the attention of many researchers to explore this skeleton to its multiple potential against several activities. The triazole derivatives possess a great importance in medicinal chemistry and can be used for the synthesis of numerous heterocyclic compounds with different biological activities. This review article covers the information of triazoles derivatives having different psychopharmacological actions. Thus triazole acts as a promising medicinal agent for the scientists working over this field. This review can be helpful to develop various more new compounds possessing triazoles moiety that could be better in terms of efficacy and lesser toxicity.

The presence of three nitrogen hetero-atoms in five-membered ring systems defines an interesting class of compounds, the triazole. This may be of two types, the 1,2,3-triazoles and the 1,2,4-triazoles. Out of the two triazoles, 1,2,4-triazole have drawn great attention due to its wide variety of activities, low toxicities and good pharmacokinetic and pharmacodynamic profiles. Chemistry of triazoles and their fused heterocyclic derivatives has received considerable attention owing to their effective biological activities such as anti inflammatory, analeptic, sedatives, antianxiety, antimicrobial, antimycotic and other pharmacological and chemical properties. In this article, antiviral and antiparasitic activities of triazole derivatives are discussed.
