Short-Term culture of in vitro produced bovine preimplantation embryos with insulin-like growth factor-i prevents heat shock-induced apoptosis through activation of the Phosphatidylinositol 3-Kinase/Akt pathway

Certain reproductive techniques culture embryos in vitro; however, little is known about the impact of culture on fetal growth. Coculture of day 1ovine zygotes on a bovine granulosa cell layer to blastocysts followed by transfer to synchronous recipients increased fetal weight by 11 and 40% at days 61 and 125, respectively, compared with the transfer of in vivo-produced blastocysts. Plantaris muscle weights were increased by 40% in cultured fetuses at day 125. Examination of myogenesis in plantaris muscle showed that primary fiber number was unchanged at day 61 by culture but that primary fiber area was increased significantly by 15 and 25% at days 61 and 125, respectively; secondary fiber area was increased by 40% at day 125 by culture, and the ratio of secondary to primary fiber numbers was 18–20% greater in the cultured groups compared with the controls at days 61 and 125. The results show that coculture of preimplantation embryos may alter myogenic programming. These changes may contribute to the abnormally large muscles observed in oversize fetuses.

e13003 Background: We have demonstrated that cancer cells from patients (Pts) with recurrent or metastatic Breast CA frequently show as much or better APOP with Generics compared to Proprietaries (separately submitted to ASCO, 2012). We have compared these observations to in vitro APOP in Pts with NSCLC and Colon CA. Methods: Purified tumor cells from Pt biopsies were placed into short term culture using the microculture kinetic (MiCK) assay. APOP was analyzed every five minutes over 48 hours. APOP was defined in kinetic units (KU) of apoptosis. Significant APOP was > 1.0 KU, significant differences between individual assays were defined as > 0.57 KU based on replicate analyses. Results from Breast CA, Colon CA and NSCLC were compared. Results: 41 Pts with NSCLC, 8 Pts with Colon CA and 67 Pts with Breast CA had successful cultures. Generics produced APOP greater than Proprietaries in 25/32 Pts with NSCLC (78%), 4/7 Pts with Colon CA (57%) and 36/43 Pts (84%) with Breast CA. Generics produced APOP = Proprietaries in 5 Pts with NSCLC (16%), 1 Pt with Colon CA (14%) and 6 Pts (14%) with Breast CA. Proprietaries produced APOP greater than Generics in 2 Pts with NSCLC (6%), 2 Pts with Colon CA (29%) and 1 Pt (2%) with Breast CA. There were 0 Pts with NSCLC, Colon CA or Breast CA in whom no drug produced significant APOP (KU less than 1.0). Proprietaries produced more APOP in Colon CA than in Breast CA (p<0.05). Detailed comparisons of drugs by disease will be presented. Conclusions: Generic drugs can produce APOP in vitro equal to or better than Proprietary drugs in most Pts with NSCLC, Colon CA, and Breast CA. The frequency of Generic drugs being at least as active as Proprietary drugs varies by disease, and was higher in Breast CA compared to Colon CA. However, the MiCK APOP assay can identify which individual Pts might require use of Proprietary drugs. These conclusions justify prospective clinical trials to confirm these in vitro results. Increased use of Generic drugs based on the APOP assay may help to control healthcare costs.

ABSTRACT Proliferative activity was measured in rat anterior pituitary cells in short-term culture by calculating the labelling index (LI), based on the immunohistochemical detection of cells incorporating the thymidine analogue bromodeoxyuridine. Basal LI was reproducible in the test system. Arginine vasopressin (AVP) induced a dose-related increase in LI up to 20 ng/ml. Corticotrophin-releasing factor-41 (CRF-41) had no effect at doses up to 20 ng/ml. However, in the presence of 10 ng CRF-41/ml, AVP induced a greater increase in LI at lower doses than did AVP alone. Fibroblast growth factor also induced a significant increase in LI. In the system used, epidermal growth factor and insulin had no effect on proliferation. The effect of herbal components of Tokishakuyakusan on somatomedin C/insulin-like growth factor I (IGF-1) level in medium from rat corpora lutea incubated in vitro was examined. Hoelen + peony root + Japanese angelica root, hoelen + peony root, hoelen + Japanese angelica root or peony root + Japanese angelica root decreased the IGF-1 level. The data suggest that constituent herbal components of Tokishakuyakusan regulate the IGF-1 level by rat corpora lutea.
