When observing small objects such as cellular organelles by scanning electron microscopy, it is often valuable to use the techniques of transmission electron microscopy. The common practice of mounting and coating for SEM may not always be necessary. These possibilities are illustrated using vertebrate skeletal muscle myofibrils.Micrographs for this study were made using a Hitachi HFS-2 scanning electron microscope, with photographic recording usually done at 60 seconds per frame. The instrument was operated at 25 kV, with a specimen chamber vacuum usually better than 10-7 torr. Myofibrils were obtained from rabbit back muscle using the method of Zak et al. To show the component filaments of this contractile organelle, the myofibrils were partially disrupted by agitation in a relaxing medium. A brief centrifugation was done to clear the solution of most of the undisrupted myofibrils before a drop was placed on the grid. Standard 3 mm transmission electron microscope grids covered with thin carbon films were used in this study.

Familial exudative vitreoretinopathy (FEVR)is a rare genetically heterogeneous disease with multiple types of inheritance (autosomal dominant, autosomal recessive, X-linked) and widely varying clinical features. Up to 40 % of cases of FEVR are associated with mutations of the FZD4 gene.Purpose: to investigate the clinical manifestations of FEVR in children with nucleotide sequence alterations in the FZD4 gene. Material and methods. The Helmholtz National Medical ResearchCenter of Eye Diseases and the ResearchCentre for MedicalGenetics conducted a joint in-depth ophthalmological examination of 18 patients aged from 3 weeks to 17 years with a diagnosis of FEVR, which included a detailed ophthalmoscopy under drug mydriasis, ultrasound and electrophysiological examination, photographic recording of fundus changes using RetCam and Fundus Foto. Molecular genetic examination was carried out by direct sequencing according to Sanger. Results. Nucleotide sequence alterations in the FZD4 gene were detected in 3 patients(16.7 %)from two unrelated families. In one family, a 12-year-old girl wasfound to display the firstsymptoms of ophthalmic pathology (reduced vision, strabismus) at the age of 3.5 years. In another family, the clinical manifestations of FZD4 gene mutations were observed in two children during the first year of life (at the age of 5 and 11 months).Conclusions. The clinical picture of 3 patients with detected changes in the nucleotide sequence of the FZD4 gene is characterized by early manifestation and bilateral asymmetric ophthalmoscopic damage. The results of the study indicate the need for a timely diagnosis of FEVR in young children, recommend an interdisciplinary approach to the study of the disease, which should contribute to a better understanding of pathogenesis, and the development of an effective diagnostic, treatment and rehabilitation algorithm.

Introduction. The research goal is to identify and analyze the architectural features of the orthodox church buildings, designed and built in Ryazan since the early 2000ies. A number of objectives are to be accomplished to achieve this pre-set goal: one must identify and study the church buildings constructed in Ryazan, analyze the space-planning solutions and break them down into typological groups; besides, one should study the biography of their architect, as his professional track record influences the city’s historical and architectural appearance. This research is focused on contemporary church architecture exemplified by orthodox church buildings. Materials and methods. Field studies serve as the backbone of this research which encompasses fact finding and photographic recording of the source material, information analysis and generalization, tabulation, making conclusions and formulating the opinion. Results. The co-authors have analyzed the problems of contemporary church architecture and made a brief analysis of the history of orthodox church building in Ryazan. The overview encompasses 12 orthodox church buildings constructed in 2000–2014, as well as the key facts and dates associated with their construction. The co-authors have also identified compositional and other unique features of the new church buildings. They have outlined the milestones in the creative biography of the architect who designed these items of contemporary church architecture. Conclusions. The research findings comprise a scholarly insight into contemporary church architecture. The analysis of new church buildings has helped to identify the features, peculiarities and architectural techniques, applied by the architect. The features, identified by the co-authors, define the appearance/typology of contemporary church buildings and their constructions.

Modifications were made in the design and filling procedure of the Staaland device used to study salinity preference in fish. Two photographic recording techniques that eliminate the need for direct observation of fish activity and allow for continuous monitoring of fish movement throughout several daily photoperiodic cycles are described.
