When observing small objects such as cellular organelles by scanning electron microscopy, it is often valuable to use the techniques of transmission electron microscopy. The common practice of mounting and coating for SEM may not always be necessary. These possibilities are illustrated using vertebrate skeletal muscle myofibrils.Micrographs for this study were made using a Hitachi HFS-2 scanning electron microscope, with photographic recording usually done at 60 seconds per frame. The instrument was operated at 25 kV, with a specimen chamber vacuum usually better than 10-7 torr. Myofibrils were obtained from rabbit back muscle using the method of Zak et al. To show the component filaments of this contractile organelle, the myofibrils were partially disrupted by agitation in a relaxing medium. A brief centrifugation was done to clear the solution of most of the undisrupted myofibrils before a drop was placed on the grid. Standard 3 mm transmission electron microscope grids covered with thin carbon films were used in this study.

As part of a program to develop iron particles for next generation recording disk medium, their structural properties were investigated using transmission electron microscopy and electron diffraction. Iron particles are a more desirable recording medium than iron oxide, the most widely used material in disk manufacturing, because they offer a higher magnetic output and a higher coercive force. The particles were prepared by a method described elsewhere. Because of their strong magnetic interaction, a method had to be developed to separate the particles on the electron microscope grids.

A new unsymmetrical photochromic diarylethene 1-(2,4-dimethyl-3-isoxazol)-2-[2-methyl-5-(2-chlorophenyl) -3-thienyl] perfluorocyclopentene (1a) has been synthesized. Its photochromic properties in solution and PMMA film were investigated. Diarylethene 1a changed the color from colorless to red upon irradiation with 297 nm UV light, in which absorption maxima were observed at 515 nm in hexane and at 526 nm in PMMA film, The results demonstrated that it can be potentially used as polarization holographic optical recording medium.
