Martínez spruce (Picea martínezii T.F. Patterson) is a conifer currently passing through a bottleneck, reduced to a few relict populations totaling less than 800 trees. We used isozyme markers to analyze the mating system and survey the level of genic diversity in two populations. The mating system was characterized by a high frequency of selfing. The multilocus outcrossing rates (tm) and 95% confidence intervals were only 0.399 (0.197 < tm < 0.601) for the smallest population and 0.589 (0.475 < tm < 0.703) or 0.685 (0.465 < tm < 0.905), depending on year, for the largest. These are among the lowest rates of outcrossing observed in conifers. The fixation indices for the two populations were -0.058 and 0.121, less than expected for such high levels of selfing. Expected heterozygosity, unbiased He, based on 22 loci in 13 enzyme systems, was 0.121 and 0.101 in the two populations. The proportion of the total genic diversity between populations, FST, was 2.4%. Nm, the number of migrants per generation, was about 1.00 or 10.17, depending on the method of estimation. The time since the two populations were isolated was estimated from Nei's genetic distance as only 150 to 15 000 years, which is consistent with a hypothesis of population collapse during late Pleistocene or Holocene warming. We discuss the implications for conservation.

Effects of asexual reproduction as a primary reproductive strategy on population structure and levels of variability were investigated electrophoretically in natural populations of a woody plant species, trembling aspen (Populus tremuloides Michx.), from Alberta. As expected, levels of genic diversity, 42%, and proportion of polymorphic loci, 92%, averaged over all clones are considerably greater than those reported for comparable samples of sexually reproducing plant and animal species. These measures of genic variability of a primarily asexual plant species are similar to those reported for asexual species of insects, fish and bacteria. In addition, each of the 222 clones was electrophoretically unique. Since neutral theory would predict each individual clone to be heterozygous for a unique mutation at each gene locus at equilibrium, these results can be interpreted in a number of ways: (i) insufficient time to reach equilibrium, (ii) inability of electrophoresis to detect all variation at a locus, (iii) periodic establishment of sexually derived propagules in the population, and (iv) selection for similar genotypes at each location or against mutations at particular gene loci. Re-invasion of Pleistocene-glaciated areas by trembling aspen likely was by sexual means, with subsequent reproduction being primarily asexual.

Blood samples were collected from Pantaneiro Horses in five regions of Mato Grosso do Sul and Mato Grosso States. Arabian, Mangalarga Marchador and Thoroughbred were also included to estimate genetic distances and the existing variability among and within these breeds by RAPD-PCR (Random Amplified Polymorphic DNA - Polymerase Chain Reaction) molecular markers. From 146 primers, 13 were chosen for amplification and 44 polymorphic bands were generated. The analysis of molecular variance (AMOVA) indicated that the greatest portion of detected variability was due to differences between individuals within populations (75.47%). Analysis of the genetic variability between pairs of populations presented higher estimates for the five Pantaneiro populations with the Arabian breed, while lowest estimates were presented by pairs formed among the Pantaneiro populations with the Mangalarga Marchador. Highest genic diversity was shown by the Pantaneiro (0.3396), which also showed highest genetic distance with the Arabian and lowest with Mangalarga Marchador breed. UPGMA dendrogram showed distinct differences between naturalized (Pantaneiro and Mangalarga Marchador) and exotic (Arabian and Thoroughbred) breeds. In the dendrogram generated by UPGMA method, the similarity matrix generated by the Jaccard coefficient showed distinction between the naturalised breeds, Pantaneiro and Mangalarga Marchador, and the exotic breeds, Árab and English Thoroughbred. Results suggest that the Pantaneiro presents a higher genetic variability than the other studied breeds and has a close relationship with the Mangalarga Marchador.

From the early Miocene, the uplift of the Andes Mountains, intense volcanic activity and the occurrence of successive periods of dryness and humidity would have differentially influenced the modification of Altiplano watersheds, and consequently the evolutionary history of the taxa that live there. We analyzed Orestias populations from the Caquena and Lauca Altiplanic sub-basins of northern Chile to determine their genetic differentiation and relationship to their geographical distribution using mitochondrial (D-loop) and nuclear (microsatellite) molecular markers and to reconstruct its biogeographic history on these sub-basins. The results allowed reconstructing and reevaluating the evolutionary history of the genus in the area; genic diversity and differentiation together with different founding genetic groups suggest that Orestias have been spread homogeneously in the study area and would have experienced local disturbances that promoted isolation and diversification in restricted zones of their distribution.

Abstract The effects of chromosomal rearrangements on recombination rates were tested by the analysis of chiasma distribution patterns in wild house mice. Males and females of two chromosomal races from Tunisia differing by nine pairs of Robertsonian (Rb) fusions (standard all-acrocentric, 2N = 40 and 2N = 22) were studied. A significant decrease in chiasma number (CN) was observed in Rb mice compared to standard ones for both sexes. The difference in CN was due to a reduction in the number of proximal chiasmata and was associated with an overall more distal redistribution. These features were related to distance of chiasmata to the centromere, suggesting that the centromere effect was more pronounced in Rb fusions than in acrocentric chromosomes. These modifications were interpreted in terms of structural meiotic constraints, although genic factors were likely involved in patterning the observed differences between sexes within races. Thus, the change in chromosomal structure in Rb mice was associated with a generalized decrease in recombination due to a reduction in diploid number, a lower CN, and a decrease in the efficiency of recombination. The effects of such modifications on patterns of genic diversity are discussed in the light of models of evolution of recombination.
