Settlement behavior and metamorphosis of oyster larvae (Crassostrea gigas) in response to bacterial supernatants

This study evaluated the larval settlement inducing effect of sugars and a conspecific cue from adult shell extract of Crassostrea gigas. To understand how the presence of different chemical cues regulate settlement behavior, oyster larvae were exposed to 12 types of sugars, shell extract-coated and non-coated surfaces, and under varied sugar exposure times. Lectin-glycan interaction effects on settlement and its localization on oyster larval tissues were investigated. The results showed that the conspecific cue elicited a positive concentration dependent settlement inducing trend. Sugars in the absence of a conspecific cue, C. gigas adult shell extract, did not promote settlement. Whereas, in the presence of the cue, showed varied effects, most of which were found inhibitory at different concentrations. Sugar treated larvae exposed for 2 h showed significant settlement inhibition in the presence of a conspecific cue. Neu5Ac, as well as GlcNAc sugars, showed a similar interaction trend with wheat germ agglutinin (WGA) lectin. WGA-FITC conjugate showed positive binding on the foot, velum, and mantle when exposed to GlcNAc sugars. This study suggests that a WGA lectin-like receptor and its endogenous ligand are both found in the larval chemoreceptors and the shell Ethylenediaminetetraacetic acid (EDTA) extract that may complementarily work together to allow the oyster larva greater selectivity during site selection.

Costa Rica muestra una condición incipiente en el desarrollo de la acuicultura marina, a pesar de los esfuerzos invertidos por más de veinte años. La experiencia en el cultivo de Crassostrea gigas alcanzada por comunidades, la institucionalidad pública y ONG´s, desde el ámbito social- económico y ambiental, evidencian que el país cuenta con las condiciones idóneas, para direccionar la ostricultura como un nuevo sector socio-económico con y desde las comunidades marino costeras del litoral pacífico. El presente trabajo expone los logros obtenidos por comunidades y actores locales; además los retos que ambas partes se han propuesto para hacer de esta actividad una herramienta más que contribuye con la sustentabilidad y empoderamiento de familias y comunidades que actualmente se encuentran en condiciones de pobreza.

For the past decade, the Osterberg testing method (O-cell test) has been proved advantageous over the conventional pile load testing method in many aspects. However, because the O-cell test uses a loading mechanism entirely different from that of the conventional pile loading testing method, many investigators and practicing engineers have been concerned that the O-cell test would give inaccurate results, especially about the pile head settlement behavior. Therefore, a bidirectional load test using the Osterberg method and the conventional top-down load test were executed on 1.5-m diameter cast-in-place concrete piles at the same time and site. Strain gauges were placed on the piles. The two tests gave similar load transfer curves at various depth of piles. However, the top-down equivalent curve constructed from the bidirectional load test results predicted the pile head settlement under the pile design load to be approximately one half of that predicted by the conventional top-down load test. To improve the prediction accuracy of the top-down equivalent curve, a simple method that accounts for the pile compression was proposed. It was also shown that the strain gauge measurement data from the bidirectional load test could reproduce almost the same top-down curve.
