Abstract Collaborative studies at the University of Iowa and the National Center for Voice and Speech aim to help the voices of teachers. Investigators study how cells and tissues respond to vibration doses simulating typical vocalization patterns of teachers. A commercially manufactured instrument is uniquely modified to support cell and tissue growth, to subject tissues to vocalization-like forces, and to measure viscoelastic properties of tissues. Through this basic science approach, steps toward safety limits for vocalization and habilitating rest periods for professional voice users will be achieved.

In the last thirty years we have witnessed a burgeoning of research in thanatology. As in most disciplines, the revolution has been followed by a period of filling-in. As we confront the possibility of nuclear and biological annihilation, we need to recover the pioneering vision and spirit that heralded the beginning of thanatology. The authors in this volume call for broad-based interdisciplinary, multi-method, collaborative studies that not only contribute to the development of thanatological theory and research, but also are of use to clinicians, social planners and legislators.

Abstract Raceophenidol in feed at concentration levels intended for growth promotion of poultry can be estimated by a curvature inversion measurement related to the ultraviolet spectrum of derived p-methylsulfonylbenzaldehyde. The drug can be accurately measured at the 0.0005% level. Collaborative studies on the method showed an average value of 94 ± 10% of claim. The method is recommended for adoption as official, first action.

Abstract A colorimetric method for the determination of hydroxyproline as a measure of collagen in meat and meat products has been collaboratively studied in 18 laboratories. The method includes hydrolysis with sulfuric acid, oxidation with chloramine- T, and formation of a reddish purple complex with 4- dimethylaminobenzaldehyde. Five frozen and 3 freeze-dried samples were tested, ranging in content from 0.11 to 0.88% and from 0.39 to 4.0% hydroxyproline, respectively. The mean values of 2 identical samples were 0.245 and 0.251 %. The average recovery from a spiked sample was 96.1 %. The hydroxyproline content of a known sample (a mixture of 2 samples in the ratio 5:2) was calculated to 1.42%, which agrees well with the analytical result, 1.40%. In comparison with other collaborative studies, based on the ISO analytical method, the repeatability and reproducibility of this method agree well with the other results. This method was accepted as an official NMKL method by all national Committees, and has been adopted official first action by AOAC as an NMKLAOAC method.

Abstract Antioxidants (BHA and BHT) in different types of breakfast cereals were studied by argon ionization gas chromatography. Samples containing antioxidants were packed into the column, and antioxidants were eluted with redistilled petroleum ether. EIuates were concentrated under N2 and injected into a Barber-Colman Model 10 gas chromatograph. Recoveries ranged from about 92 to 110%. Results obtained by steam distillation-colorimetry methods were erratic. Colorimetric procedures were not free from interferences and could lead to highly erroneous results at the lower ppm levels. The gas chromatographic method is faster, simpler, and more accurate. Collaborative studies on corn and rice ready-to-eat breakfast cereals are recommended.

Abstract The AOAC Roese-Gottlieb and Rapid Detergent (TeSa) methods for the determination of butterfat in pasteurized, homogenized milk and in chocolate milk were compared in a collaborative study. The results by the detergent method on both products were comparable to those by the Roese-Gottlieb method. The Babcock method gave low results on the homogenized milk.

The development, by collaborative study, of standardised method for the determination of mono- and diacylglycerols in vegetable oils and fats is described. The method involves separation of mono- and diacylglycerols by normal phase high-performance liquid-liquid chromatography (HPLC) and evaporative light scattering detection of a solution of oil, fat or a commercial mono- and diacylglycerol preparation in a organic solvent.

e16187 Background: Bile tract cancers are genetically and clinically heterogenous with a poor prognosis. Identifying novel biomarkers for targeted therapy is required to improve the clinical outcome of bile tract cancer patients. Methods: Tumor tissue samples of 482 Chinese biliary tract cancer (BTC) patients were genetically profiled using targeted next generation sequencing. Tumor mutation burden (TMB) was calculated by counting all nonsynonymous mutations per megabase of coding sequences. The R package ReactomePA was used in pathway enrichment analysis. Genomic instability was characterized by an in-house developed NGS-based Homologous Recombination Deficiency (HRD) panel and a HRD score was an unweighted sum of loss of heterozygosity (LOH), telomeric allelic imbalance (TAI), and large-scale state transitions (LST) scores. Results: The BTC cohort consisted of 135 gallbladder cancer (GBC), 73 intrahepatic cholangiocarcinoma (iCCA), 18 distal cholangiocarcinoma (dCCA), 14 perihilar cholangiocarcinoma (pCCA), while the remaining 242 BTC patients of no specific subtype information. Most frequently mutated genes included TP53 (56%), KRAS (25%), ARID1A (17%), SMAD4 (11%), and CDKN2A (10%) . A preliminary pathway analysis revealed that mutations of DNA damage repair (DDR) pathway genes were enriched in the cohort ( p< 1e-10), accounting for over 70% of the patients, particularly in homologous recombination repair (HRR), Fanconi anemia (FA), mismatch repair (MMR), and base excision repair (BER) genes. More specifically, approximately 50% of the cohort carried at least one mutation of the HRR genes (43%) or MMR genes (14%). Patients with impaired MMR had increased microsatellite instability status (MSI) comparing to those with wildtype MMR (33% vs. 3.1%, p< 0.0001), and patients harboring HRR mutations demonstrated elevated genomic instability than those without such mutations (median HRD: 18 vs.14, p < 0.05), indicative of potential response to poly (ADP-ribose) polymerase (PARP) inhibitors and other DNA-damage agents. Furthermore, high TMB was found to be highly correlated with DDR gene alterations ( p =0.004). In addition, we observed higher mutation frequencies of BRCA1/2 genes (including somatic and germline) in GBCs in contrast to other BTC subtypes. Conclusions: We herein reported the genomic features of 482 Chinese BTC samples and highlighted the role of DDR pathways including HRR and MMR. These findings could be useful to establish treatment and diagnostic strategies for BTC patients based on genetic information.
