Blackgram (Vigna mungo L. Hepper, syn. Phaseolus mungo L.) is one of the four major pulse crops. So far, no reports of tissue culture studies aiming at genetic improvement of this crop are available in Bangladesh. The present study comprised of experiments for direct shoot regeneration and plantlet formation in blackgram. Shoot tip explants were cultured on different concentrations of BAP (1.0, 5.0 and 10.0 mg L-1) and Kinetin (1.0, 5.0 and 10.0 mg L-1). The highest percentage (85.00%) of multiple shoot initiation was observed in the MS medium supplemented with 5.0 mg L-1 kinetin. The regenerated shoots were transferred to rooting medium supplemented with different concentrations of NAA. A high frequency of 85% rooting was observed with 0.2 mg L-1 NAA. Among the in vitro derived different explants such as cotyledonary leaf, hypocotyl, shoot tip and root of two local varieties, namely Singhnath and Kazla (BARI Begun-4) of eggplant (Solanum melongena L.) cotyledonary leaf was found to be the best for multiple shoot regeneration. High frequency direct organo-genesis of shoots was achieved from cotyledonary leaf in MS supplemented with 1.0 mg/l BAP and 1.0 mg/l Kn. Anatomical studies using freezing microtome supported the formation of shoots through organogenesis. Proliferation and elongation of such shoots were obtained in hormone free MS. Moreover, the regenerated shoots produced healthy roots when they were cultured on MS without hormonal supplements. Following the formation of roots the in vitro raised plantlets were successfully established in soil. Effects of different hormonal concentration on multiple shoot formation of RABI Strawberry-3 were studied. Where the explants were runner tips that cultured on MS basal medium supplemented with 6-Benzyl Adenine and Kinetin. The maximum percentage 80% of shoot was obtained from Benzyl Adenine concentrations of 0.5 mg/l and 1.0 mg/l whereas Kinetin concentrations were 1.5 mg/l and 2.0 mg/l separately. More promising result was found by combining two impressive concentrations of each hormone. When MS basal media was supplemented with Kinetin 2.0mg/l + Benzyl Adenin A 0.5 mg/l, Kinetin 1.5 mg/l + Benzyl Adenine 0.5 mg/l and Kinetin 1.5 mg/l + Benzyl Adenine 1.0 mg/l respectively then 100% shoots were obtained.
