Development of nanoemulsion from Vitex negundo L. essential oil and their efficacy of antioxidant, antimicrobial and larvicidal activities (Aedes aegypti L.)

The essential oils from the leaves of Annona salzmannii and A. pickelii (Annonaceae) growing in Sergipe, northeastern region of Brazil, were obtained by hydrodistillation using a Clevenger-type apparatus, and analyzed by GC/MS and GC/FID. Thirty-four compounds were identified in the essential oil of A. salzmannii and twenty-seven in that of A. pickelii; sesquiterpenes predominated in both essential oils. Bicyclogermacrene (20.3%), ( E)-caryophyllene (19.9%), δ-cadinene (15.3%), α-copaene (10.0%), and allo-aromadendrene (5.7%) were the main components of A. salzmannii, and bicyclogermacrene (45.4%), ( E)-caryophyllene (14.6%), and α-copaene (10.6%) of A. pickelii. The essential oils showed significant antioxidant capacity in the ORACFL and DPPH assays. The antimicrobial activity of these essential oils was also evaluated against bacteria and fungi, as well as the larvicidal activity against Aedes aegypti larvae.

The chemical constituents of essential oils extracted by hydrodistillation from Piper klotzschianum, P. hispidum, and P. arboreum, collected from the remaining Atlantic forest in Sergipe State, were characterized and submitted to cytotoxic and larvicidal activity assays. The major constituents identified (stems, fresh, and dried leaves) were ( E)-caryophyllene (16.8%) and bicyclogermacrene (21.6%). Germacrene D (22.8%) was identified only in P. klotzschianum and P. hispidum. A high percentage of β-pinene (27.2%) and α-pinene (7.2%) were present in P. klotzschianum stems and δ-3-carene in the fresh and dried leaves of P. hispidum (17.4% and 19.1%, respectively). Piper klotzschianum showed the best cytotoxic activity, inhibiting human hepatocellular carcinoma (27.3 µg/mL), human promyelocytic leukemia (33.8 µg/mL), and murine melanoma (37.9 µg/mL) cell lines. Larvicidal activity was also tested and the essential oil from the dried leaves of P. klotzschianum was the most potent against Aedes aegypti larvae, with an LC50 value of 122.4 µg/mL.

The larvicidal activities of essential oils from the leaves of Artemisia vulgaris L., Cymbopogon flexuosus(Nees ex Steud.) Wats. and Piper tuberculatum Jacq. were evaluated using third-instar larvae of Aedes aegypti. The essential oils were obtained by hydrodistillation and analyzed by GC and GC-MS. The essential oil of P. tuberculqatum had the lowest LC50 value (106.3 ± 2.2 μg/mL), followed by that of A. vulgaris(114.1 ± 1.7 μg/mL) and C. flexuosus(121.6 ± 0.8 μg/mL). The results show that these essential oils may be potent sources of natural larvicides.

Blue tansy essential oil (BTEO) ( Tanacetum annuum L.) was analyzed by GC-MS and GC-FID using two different capillary column stationary phases. Sabinene (14.0%), camphor (13.6%), myrcene (8.0%), β-pinene (7.7%), and chamazulene (6.9%) were the main components using an SE52 column (non-polar). On a polar CW20M phase column, sabinene (15.1%), camphor (14.4%), α-phellandrene (7.9%), β-pinene (7.7%), and myrcene (6.9%) were the most abundant compounds. To assess the oil for potential applications in integrated pest management strategies, behavioral bioassays were conducted to test for repellency against yellow fever mosquito Aedes aegypti, and for attractant activity for Mediterranean fruit fly Ceratitis capitata. Results showed that BTEO was not effective in repelling Ae. aegypti (minimum effective dosage [MED]: 0.625 ± 0.109 mg/cm2 compared with the standard insect repellent DEET (N,N-diethyl-3-methylbenzamide). In assays with male C. capitata, BTEO displayed mild attraction compared with two positive controls (essential oils from tea tree Melaleuca alternifolia and African ginger bush Tetradenia riparia). Additional studies are needed to identify the specific attractant chemicals in BTEO and to determine if they confer a synergistic effect when combined with other known attractants for C. capitata. To the best of our knowledge, this study represents the first investigation of BTEO for repellency against the mosquito vector Ae. aegypti and for attractancy to C. capitata, a major agricultural pest worldwide.
