The aim of this study was to determine the prevalence and intensity of gastrointestinal parasites in donkeys slaughtered in Kinamba, Mogotio, and Lodwar slaughterhouses and their association with several host factors. A survey was done between July and September 2017 in three slaughterhouses. Faecal samples were collected per rectum from all the study donkeys for faecal egg counts (EPG) and morphological identification of the eggs. At slaughter, the gastrointestinal tracts were opened and examined visually, and all helminth parasites collected were subjected to morphological identification. Prevalence and intensity were calculated based on the helminth identified and EPG. A total of 282 donkeys were sampled. A majority of the donkeys (89%) were in poor body condition. Ten helminth parasite species were identified in 85.5% donkeys. They were Strongylus vulgaris (52.8%), Parascaris equorum (20.2%), Strongylus edentatus (12.1%), Anaplocephala perfoliata (10.3%), Setaria equina (3.5%) Anaplocephala magna (2.5%), Cylicocyclus auriculatus (2.1%), Cyathostomum species (1.8%), Strongylus equinus (0.4%), and Triodontophorus serratus (0.4%). A significant percentage (55.3%) had no eggs in their feces, 39% had low infection, 5% had medium, and only 0.7% were heavily infected. Prevalence rates via use of the EPG showed Strongyles (44.7%), Parascaris equorum (5.3%), Oxyuris equi (11%), Triodontophorus tenuicolis (0.7%), Habronema species (0.7%), and cestodes eggs (0.4%). No significant differences were observed between fecal Strongylus egg count and age, sex, and pregnancy status. However, donkeys with poor body condition shed more Strongylus eggs in feces. Helminth infections are prevalent in donkeys in Kenya; however, this is not reflected in coprological analyses. These helminth parasites may contribute to poor body condition, ill health, and poor productivity of donkeys.

With the aim of studying the endoparasite fauna of horses from the Formiga city, located in center-west region of the state of Minas Gerais, 25 animals that were naturally infected with helminths were evaluated. By means of parasitological necropsies, different endoparasites were found. The subfamily Cyathostominae presented the highest incidence, followed by Trichostrongylus axei, Oxyuris equi, Triodontophorus serratus, Strongyloides westeri, Strongylus edentatus, Habronema muscae, Parascaris equorum, Probstmayria vivipara, Strongylus vulgaris, Gasterophilus nasalis, Anoplocephala magna and Anoplocephala perfoliata. In the present study, if the species Probstmayria vivipara was not considered in the prevalence, the frequency of Cyathostominae was equivalent to 94.85%. The results obtained in this study allowed us to detect and identify different species of helminths in horses, and confirmed the high incidence of nematodes belonging to the subfamily Cyathostominae in the center-west region of Minas Gerais.

Abstract The present research is a continuation of studies conducted periodically over 40 years on transmission of natural infections of internal parasites in the same horse herd on pasture (Field 10) on a farm in Central Kentucky. It included 12 mixed light horse foals born in 2013 and euthanatized between July, 2013 and April, 2014 for collection of internal parasites. Parasites found: Gasterophilus intestinalis, Strongyloides westeri, Parascaris equorum, Anoplocephala perfoliata, small strongyles (cyathostomes), Strongylus vulgaris, Strongylus edentatus and Thelazia lacrymalis. Prevalence generally was related to age of the foals. Overall prevalence and number of specimens were lower than in earlier studies except for P. equorum. There were 15 species (much fewer than previously) of small strongyles found and recorded by location in the large intestines. All stages of small strongyles encysted in the mucosa of the large intestine were recovered by artificial digestion and in significantly lower numbers in older foals

AbstractThere is a vital need for authentic COVID-19 animal models to enable the pre-clinical evaluation of candidate vaccines and therapeutics. Here we report a dose titration study of SARS-CoV-2 in the ferret model. After a high (5 × 106 pfu) and medium (5 × 104 pfu) dose of virus is delivered, intranasally, viral RNA shedding in the upper respiratory tract (URT) is observed in 6/6 animals, however, only 1/6 ferrets show similar signs after low dose (5 × 102 pfu) challenge. Following sequential culls pathological signs of mild multifocal bronchopneumonia in approximately 5–15% of the lung is seen on day 3, in high and medium dosed groups. Ferrets re-challenged, after virus shedding ceased, are fully protected from acute lung pathology. The endpoints of URT viral RNA replication & distinct lung pathology are observed most consistently in the high dose group. This ferret model of SARS-CoV-2 infection presents a mild clinical disease.

Purpose: The purpose of this drug review was to explore the safety and efficacy of the newly approved benzodiazepine, remimazolam, in order to evaluate its place in therapy. Summary: Remimazolam has a faster onset of action and recovery time than midazolam when given as single IV doses. Additionally, it has no known CYP450 interactions that would contribute to drug-drug interactions. Patients with severe hepatic impairment may require dose titration as well as the elderly who should be closely monitored. Although remimazolam vials should be protected from light and must be reconstituted immediately before use, the reconstituted vial may be stored for later use at room temperature for up to 8 hours. Remimazolam is more expensive than current options used in practice, as such individual institutional formulary and provider preference will require review to see if its advantages are worth the additional cost and to determine its place in therapy. Conclusion: Remimazolam is a novel option when choosing a benzodiazepine for procedural sedation that has pharmacokinetic and pharmacodynamic advantages when compared to other commonly prescribed sedatives. Remimazolam has proved superior to midazolam when analyzing drug-drug interactions, onset, and time to alertness. Remimazolam also has a shorter elimination half-life and decreased volume of distribution when compared to midazolam.

OBJECTIVE: To assess the stability of naltrexone oral liquid prepared from tablets and powder, and to evaluate its use in precipitating rapid withdrawal from methadone. DESIGN: Naltrexone 1 mg/mL oral liquids were prepared from tablets and powder and stored in the dark at 4, 25, and 70 °C. Similar formulations containing 5 mg/mL were stored at 70 °C. The 1-mg/mL formulation prepared from tablets was clinically evaluated in inducing rapid withdrawal in two drug-dependent individuals receiving methadone maintenance treatment using a naltrexone dose titration protocol. SETTING: A university pharmacy school and affiliated urban teaching hospital. MAIN OUTCOME MEASURES: Samples removed at six time points were analyzed for naltrexone concentration to assess decomposition over 90 days. An opioid withdrawal symptom checklist was used to assess the severity of the withdrawal symptoms prior to, and 30 minutes after, each dose of naltrexone. RESULTS: Decomposition of naltrexone in all formulations stored at 4 and 25 °C was not significant over 90 days. Both patients tolerated naltrexone 1 mg/mL oral liquid, but found it bitter and gritty. Withdrawal symptoms were experienced immediately after the first dose, but were resolving by the end of day 3 of naltrexone treatment, at which stage both patients were able to tolerate a 50-mg tablet of naltrexone as maintenance. CONCLUSIONS: Naltrexone 1 mg/mL oral liquids prepared from tablets or powder are stable when stored in the dark for 60 days at 4 °C and for 30 days at 25 °C. The formulation prepared from tablets provides flexible dosing in patients undergoing rapid withdrawal from methadone.

AbstractObjectiveAssess cardiovascular effects of once-daily mixed amphetamine salts extended release (MAS XR) in adolescents (13–17 years of age) with attention-deficit/hyperactivity disorder (ADHD).MethodsBlood pressure (BP), pulse, and electrocardiograms were assessed in 327 healthy subjects during a 4-week, randomized, double-blind, placebo-controlled, forced dose-titration study. Placebo (n=69) or once-daily MAS XR(10, 20, 30, or 40 mg) was administered to subjects ≤75 kg (n=233); 50- and 60-mg MAS XR was administered to subjects >75 kg (n=25). One hundred thirty-eight subjects participated in a 6-month, open-label extension study.FindingsChanges in BP and QTcB (Bazett's formula) intervals at 4 weeks with MAS XR were not significantly different from the placebo group. Pulse increased by 5.0 and 8.5 bpm after 3 weeks with MAS XR 20 and 50 mg/day, respectively (P≤.002). After 6 months of open-label MAS XR treatment, mean increases in systolic BP (1.7 mm Hg; P=.0252) and pulse (4.4 bpm; P<.0001) were statistically, but not clinically, significant diastolic BP was not significantly changed (0.6 mm Hg) A decrease in QTcB interval (-4.6±19.9 msec) was statistically (P=.009), but not clinically, significant. There were no serious cardiovascular adverse events.ConclusionCardiovascular effects of short- and long-term MAS XR treatment (≤60 mg/day) were minimal in otherwise healthy adolescents with ADHD.
