The aim was to evaluate the effect of subclinical mastitis by somatic cell count (SCC) on follicular dynamics, ovulation, oocyte and cumulus cell quality, exosome size and concentration in milk-producing cows. Crossbred cows (Bos taurus × Bos indicus; that is, Holstein × Gyr) were randomly allocated to control (SCC <200,000 cells mL−1] and mastitis (SCC >400,000 cells mL−1) groups. In experiment 1 (follicular dynamics), cows (n = 57) were submitted to ultrasonographic evaluations every 24 h, after removal of an intravaginal progesterone device (Day 8) up to Day 10. From Day 10, ultrasound evaluations were performed every 12 h, until ovulation or until 96 h after progesterone device withdrawal, in order to follow final dominant follicle growth and ovulation. In experiment 2 (oocyte, cumulus cells, and follicular fluid evaluation), cows (n = 23) were submitted to follicular aspirations, preceded by synchronization of the emergence of the follicular wave. The levels of target genes in cumulus cells (BCL2, BAX, PI3K, PTEN, FOXO3) were evaluated by RT-qPCR. In the follicular fluid, the exosomes were isolated for evaluation of particle size. Data were analysed by the Glimmix procedure of SAS (SAS Institute Inc., Cary, NC, USA). Ovulation rate (P = 0.09) was higher in control cows [control 77.42% (24/31) and mastitis 57.69% (15/26)]. Viable oocyte rate (P = 0.01) was also higher in control cows [control 59.1% (130/220) and mastitis 41.9% (125/298)]. The dynamics of follicular growth did not differ between groups. The number of degenerate oocytes (P = 0.001) was higher in cows of the mastitis group. In the evaluation of cumulus cell gene expression, there was a higher abundance of BAX transcripts (P = 0.003) in cells of mastitis cows. Additionally, the mean and mode of exosome diameter in mastitis cows were smaller (P = 0.03 and P = 0.02, respectively). In conclusion, ovulation rate, oocyte quality, and follicular fluid exosome diameter were lower in cows with subclinical mastitis, demonstrating a link between mammary gland sanitary status and reproduction.

<p class="abstrak2">Superovulation treatment on PO cattle (Bos indicus) was less responsive compared to Bos taurus breed. It might due to the difference of their follicular dynamic. This study was conducted to investigate the follicular dynamics and its repeatability in PO cattle. Follicular dynamics observations conducted on 9 cows trough ultrasound scanning every day. Observations of wave patterns repeatability were performed in 6 cows which its wave pattern already known on the next consecutive IOI.  Research result indicated that PO cattle had 3 (66%) and 4-waves (34%) pattern. The first wave of 3 and 4-waves pattern emerged on day -0.4+0.9 and 1.4+1.1 respectively.  The second wave of 3 and 4-wave pattern emerged on day 9.8+1.5 and 7.4+1.9 respectively.  The pattern of 3 waves has a longer follicle dominant duration (11.6+1.5 day) in the first wave of estrous cycle, compared with 4 waves pattern (10+2.92 and 7+1.00 day respectively). The growth rate of dominant follicle was not different significantly between the 3 and 4-waves pattern (0.87+0.23 and 0.94+0.25 mm/day respectively). Similarly, ovulatory follicle diameter between 3 and 4-waves pattern was also not different significantly (12.24+12.34 and 12.30+12.23 mm respectively). Observation of wave patterns repeatability in 6 PO cows indicated that PO cattle had high repeatability in follicular wave pattern (0.88) and the number of growing follicle was 0.91.  This study resulted data for dynamic of follicular development, wave pattern, its repeatability which be expected to design the protocol of superovulation treatment or other reproduction technologies based on follicular dynamic to improve its result in PO cattle.</p><p> </p>

Two experiments in Nelore cows subjected to conventional protocol for timed AI (TAI) were designed to compare the influence of antral follicle count (AFC; 1) on the ovarian follicular dynamics and (2) conception rates. First, multiparous cows with high (G-high, ≥45 follicles; n = 43) and low AFC (G-low, ≤15 follicles; n = 32) were selected to receive a conventional oestradiol/progesterone-based TAI protocol for monitoring ovarian follicular dynamics. Second, AFC from 962 multiparous suckling cows (40 ± 15 days postpartum) subjected to a conventional protocol for TAI were classified as G-high (≥45 follicles; n = 194), G-intermediate (≥20 and ≤40 follicles, n = 397), and G-low AFC (≤ 15 follicles; n = 243). Pregnancy status was evaluated 30 to 35 days later. Data were analysed using ANOVA followed by the Tukey test and model of logistic regression (P ≤ 0.05). In the first study, the AFC were greater (P ≤ 0.05) in G-high than in G-low, but remained stable in both groups during the evaluations. The ovarian diameter, perimeter, and area were greater (P ≤ 0.05) in G-high than in G-low (28.3 ± 3.9 v. 20.5 ± 3.2 mm, 100.9 ± 13.2 v. 73.8 ± 13.9 mm, and 67.5 ± 16.4 v. 37.7 ± 11.7 mm2; respectively). Follicular growth rate, time of ovulation, corpus luteum diameter, and progesterone concentration were similar (P > 0.05) for cows with low or high AFC. Dominant follicles showed greater (P ≤ 0.05) diameter in G-low than G-high at D4 (7.3 ± 2.2 v. 6.2 ± 1.4 mm; P = 0.069, tendency), D8 (11.2 ± 1.8v. 9.5 ± 1.8 mm), D9 (12.3 ± 1.7 v. 10.6 ± 1.7 mm), and D10 (13.4 ± 1.3 v. 12.2 ± 1.8 mm), and the estimated diameter of the ovulatory follicle tended (P = 0.08) to be higher in G-low than in G-high (14.4 ± 1.5 v. 13.4 ± 2.1 mm). In the second study, AFC of G-low cows showed higher (P ≤ 0.05) conception rate (61.7%) than those of G-intermediate (52.9%) and G-high (49.5%) cows. In Nelore cows subjected to ovulation synchronization protocol, cows with high AFC showed higher ovarian measurements, but those with low AFC had large follicular diameters. Additionally, the cows with low AFC showed higher conception rate than groups with intermediate and high counts.

Abstract We evaluated the effect of the antral follicle count (AFC) on ovarian follicular dynamics, pregnancy rates, progesterone concentrations, and transcriptional patterns of genes in Nelore cattle (Bos taurus indicus) after a timed artificial insemination (TAI) programme. Cows were separated based on the AFC, and those with a high AFC showed a larger (P < 0.0001) ovarian diameter and area than those with a very low AFC. Females with a very low AFC exhibited a larger (P < 0.01) diameter of the dominant follicle at TAI (13.6 ± 0.3 vs. 12.2 ± 0.4 mm) and a tendency (P = 0.06) to have different serum progesterone concentrations (2.9 ± 0.3 vs. 2.1 ± 0.3 ng/mL; on day 18, considering day 0 as the beginning of the synchronization protocol) than those with a high AFC. The pregnancy rate was higher (P ≤ 0.05) in animals with a very low (57.9%) and low (53.1%) AFC than in those with a high AFC (45.2%). The expression of genes related to intercellular communication, meiotic control, epigenetic modulation, cell division, follicular growth, cell maintenance, steroidogenesis and cellular stress response was assessed on day 5. In females with a low AFC, 8 and 21 genes in oocytes and cumulus cells, respectively, were upregulated (P < 0.05), while 3 and 6 genes in oocytes and cumulus cells, respectively, were downregulated. The results described here will help elucidate the differences in ovarian physiology and the reproductive success of Bos indicus females with a low or high AFC.

Ultrasound-guided follicular puncture (OPU) has become the most used technique to recover cumulus-oocyte complexes (COCs) from valuable donors for in vitro embryo production, because of the low risk and the possibility of collecting COCs at intervals as short as twice-a-week. However, repeated aspiration of ovarian follicles may induce endocrine abnormalities due to partial luteinization of punctured follicles and interference with follicular development. The use of exogenous progestagens is an alternative used to control these side effects, and is under evaluation. The aim of this study was to evaluate whether the effect of norgestomet treatment on intra-follicular and systemic steroid concentrations and on ovarian follicular dynamics is related to changes in LH releasing pattern. Pluriparous non-lactating Gir breed (Bos indicus) cows (n = 10) were randomly distributed between treatment (norgestomet ear implants, replaced weekly) and control (no hormone used) groups, and had their ovarian follicles larger than 3 mm in diameter aspirated twice a week, during the next two consecutive weeks. Follicular dynamics were evaluated every 12 h between OPU sessions, and the largest follicles present were used to recover samples of follicular fluid. Blood samples were collected daily for progesterone evaluation in all cows, and 3 times within a 4-h window interval, at 24, 48, 72, and 96 h after follicle puncture for LH evaluation, in 6 cows (3 from treated and 3 from control groups). LH was measured by a standardized RIA procedure. Data was analyzed by ANOVA, and means were compared by Tukey's test. Results are presented as means ± SEM. Treatment with norgestomet reduced mean progesterone plasma concentration during the evaluated period (36.3 ± 14.0 vs. 250.3 ± 49.3 pg/mL; P < 0.0001), the incidence of follicles growing above 9 mm (30% vs. 65%; P < 0.05) in the intervals between OPU sessions, and intrafollicular estradiol and progesterone concentrations in the largest follicles (n = 27) present (265.5 ± 47.4 and 34.9 ± 5.4 ng/mL vs. 765.2 ± 169.1 and 173.3 ± 43.4 ng/mL, respectively; P < 0.05). Plasma LH concentrations were consistently lower during the 3 session intervals in cows treated with norgestomet (0.16 ± 0.04, 0.22 ± 0.03, 0.22 ± 0.09 and 0.17 ± 0.01 vs. 0.44 ± 0.15, 0.53 ± 0.04, 0.42 ± 0.05 and 0.39 ± 0.11 for 24, 48, 72, and 96 h after OPU, respectively; P < 0.05). These results confirm the theory that norgestomet treatment is associated with a reduction in the LH-release pattern, as expected due to the reduction in both luteinization of punctured follicles and in the steroidogenic activity of growing follicles observed during the experiment. The use of norgestomet ear implants can be an alternative in the management of donor cows undergoing oocyte pickup. This work was sponsored by FAPEMIG and CNPq. Follicular dynamics of Zebu Cattle are characterized by lower maximum diameter and persistence of dominant follicles, when compared to European breeds. However, the main effects of follicular dominance on subordinated follicle population, and consequently on oocyte recovery and embryo production, have been shown to be similar among breeds. The aim of this study was to evaluate the dominant follicle establishment period, indicated by follicle divergence, in Zebu cows. Pluriparous, non-lactating Guzera breed (Bos indicus) cows (n=5), presenting a mean body weight of 518.0±48.5kg and similar body score condition (ranging from 3.5 to 4, in a 1 to 5 scale), were used. In a first trial, follicular dynamics were evaluated daily during 1 (n=2) or 2 (n=3) estrous cycles, using a portable ultrasound device equipped with a linear rectal 5MHz transducer (Aloka SSD500, Aloka Co., Tokyo, Japan). In a second trial, estrous cycles were synchronized using progesterone intravaginal-releasing devices (CIDR, Pharmacia, Sao Paulo, Brazil), and follicular dynamics were evaluated every 6h after ovulation, during the first follicular wave, until the largest follicle achieved a diameter of 10mm. During each evaluation ovaries were mapped, and evolution of follicles followed individually. Data are normalized for the moment of follicular divergence, and analyzed by ANOVA. Results are presented as mean±SD. Data from the first evaluations confirmed the occurrence of cycles with three (4/8) and four (1/8) follicular waves, despite a mean cycle length of 19.1±1.9 days, with a maximum diameter of 11.6±2.4mm for non-ovulatory follicles, and 14.4±0.5mm for ovulatory follicles. Follicular deviation during the first follicular wave occurred 49.2±5.0h after ovulation, when the largest follicle presented a diameter of 7.3±0.6mm, and the second largest follicle 6.8±0.7mm. Growth rates of the dominant and the largest subordinated follicles were similar before (0.35mm/6h vs. 0.33mm/6h, respectively;; P&gt;0.05), but significantly different after deviation (0.52mm 6h v. −0.04mm 6h; P&lt;0.05). These results shows that the lower maximum diameter and persistence of dominant follicles, in Zebu cattle, is partially compensated by an earlier dominant follicle deviation.
