Two-level fractional factorial design experiments were conducted in growth chambers to determine the influence of environmental factors and adjuvants on the foliar penetration of the sodium salt of acifluorfen in velvetleaf. Relative humidity (RH) was found to have the greatest effect on penetration. Temperature was the second most important factor. Although soil moisture significantly affected penetration, this effect was much less than that produced by RH and temperature. Diammonium phosphate (DAP) was the most effective adjuvant tested. The use of DAP at 1.12 kg/ha significantly increased penetration. The effect of DAP was less than either RH or temperature. Oxysorbic (5 POE) surfactant also significantly increased penetration. Glycerol and oxtoxynol (7 to 8 POE) surfactant had no effect on penetration. In separate experiments, acifluorfen penetration at low and high RH and with the addition of DAP was measured to verify these results. Acifluorfen penetration into velvetleaf increased approximately ninefold with changes in RH from low to high. Penetration increased sevenfold when DAP was added. DAP was by far the most effective adjuvant increasing acifluorfen penetration into velvetleaf under conditions of low RH. Oxysorbic (5 POE) surfactant slightly increased penetration under these conditions.

Introduction. The creating an effective and safe domestic neuroprotective medicines with a complex of pleiotropic effects realized through specific orphan receptors (SUCNR1, HCA2) of glial cells, is a pressing problem of modern pharmacology and a promising possibility of pharmacotherapy of brain injury and cerebrovascular diseases. Ideally, the medicine should helps restore lost cognitive functions and physical performance after damage to the central nervous system, and its use should improve the quality of life of patients and reduce the risk of complications.Aim. To develop composition and technology of a new medicine tablets with neuroprotective effect, using fractional factorial design of experiment and the Harrington desirability function.Materials and methods. The shape and size of particles, physicochemical (solubility, melting point) and technological properties (bulk density, compressibility factor, fractional composition, hygroscopicity) of the pharmaceutical substance of DEAE derivative were studied according to the methods described in the State Pharmacopoeia of the Russian Federation (14th edition). A three-factor fractional plan based on the 4 × 4 Latin square design for selection a scientifically based composition of solid dosage form was chosen. 16 experiments to test the significance of the factors were carried out. The tablets obtained according to the planning matrix were investigated for disintegration; tablet crush resistance, friability, and hygroscopicity. To optimize the quality of the tablets, the generalized Harrington desirability function was used.Results and discussion. The study of the physicochemical and technological properties of the DEAE derivative substance showed that it is a highly hygroscopic, amorphous, white or pale yellow, odourless powder, prone to the formation of agglomerates. The powder is very easily soluble in water. Taking into account the values of the general desirability function, the best result was shown by sample № 4 consisting of mannitol, calcium stearate, and partially pregelatinized maize starch.Conclusion. The physicochemical and technological properties of the DEAE derivative were studied. The DEAE derivative is a highly hygroscopic substance. Via the method of mathematical planning of the experiment the composition of DEAE derivative tablets was selected and scientifically grounded: DEAE derivative 60 mg, mannitol, partially pregelatinized maize starch, calcium stearate. The average tablet weight is 300 mg. The parameters of pressing tablets were selected.

The optimization of the supercritical fluid extraction (SFE) of cannabinoids, using supercritical carbon dioxide (scCO2), was investigated in a fractional factorial design study. It is hypothesized that four main parameters (temperature, pressure, dry flower weight, and extraction time) play an important role. Therefore, these parameters were screened at predetermined low, medium, and high relative levels. The density of scCO2 was used as a factor for the extraction of cannabinoids by changing the pressure and temperature. The robustness of the mathematical model was also evaluated by regression analysis. The quantification of major (cannabidiol (CBD), cannabidiolic acid (CBDA), delta 9-tetrahydrocannabinol (Δ9-THC), delta 8-tetrahydrocannabinol (Δ8-THC), and delta 9-tetrahydrocannabinol acid (THCA-A)) and minor (cannabidivann (CBDV), tetrahydrocannabivann (THCV), cannabigerolic acid (CBG), cannabigerol (CBGA), cannabinol (CBN), and cannabichomere (CBC)) cannabinoids in the scCO2 extract was performed by RP-HPLC analysis. From the model response, it was identified that long extraction time is a significant parameter to obtain a high yield of cannabinoids in the scCO2 extract. Higher relative concentrations of CBD(A) (0.78 and 2.41% w/w, respectively) and THC(A) (0.084 and 0.048% w/w, respectively) were found when extraction was performed at high relative pressures and temperatures (250 bar and 45 °C). The higher yield of CBD(A) compared to THC(A) can be attributed to the extract being a CBD-dominant cannabis strain. The study revealed that conventional organic solvent extraction, e.g., ethanol gives a marginally higher yield of cannabinoids from the extract compared to scCO2 extraction. However, scCO2 extraction generates a cleaner (chlorophyll-free) and organic solvent-free extract, which requires less downstream processing, such as purification from waxes and chlorophyll.
