AbstractWntless (WLS), an evolutionarily conserved multi-pass transmembrane protein, is essential for secretion of Wnt proteins. Wnt-triggered signaling pathways control many crucial life events, whereas aberrant Wnt signaling is tightly associated with many human diseases including cancers. Here, we report the cryo-EM structure of human WLS in complex with Wnt3a, the most widely studied Wnt, at 2.2 Å resolution. The transmembrane domain of WLS bears a GPCR fold, with a conserved core cavity and a lateral opening. Wnt3a interacts with WLS at multiple interfaces, with the lipid moiety on Wnt3a traversing a hydrophobic tunnel of WLS transmembrane domain and inserting into membrane. A β-hairpin of Wnt3a containing the conserved palmitoleoylation site interacts with WLS extensively, which is crucial for WLS-mediated Wnt secretion. The flexibility of the Wnt3a loop/hairpin regions involved in the multiple binding sites indicates induced fit might happen when Wnts are bound to different binding partners. Our findings provide important insights into the molecular mechanism of Wnt palmitoleoylation, secretion and signaling.

Serine recombinases are often tightly controlled by elaborate, topologically-defined, nucleoprotein complexes. Hin is a member of the DNA invertase subclass of serine recombinases that are regulated by a remote recombinational enhancer element containing two binding sites for the protein Fis. Two Hin dimers bound to specific recombination sites associate with the Fis-bound enhancer by DNA looping where they are remodeled into a synaptic tetramer competent for DNA chemistry and exchange. Here we show that the flexible beta-hairpin arms of the Fis dimers contact the DNA binding domain of one subunit of each Hin dimer. These contacts sandwich the Hin dimers to promote remodeling into the tetramer. A basic region on the Hin catalytic domain then contacts enhancer DNA to complete assembly of the active Hin tetramer. Our results reveal how the enhancer generates the recombination complex that specifies DNA inversion and regulates DNA exchange by the subunit rotation mechanism.

How can we merge interactive design processes and the development of interactive prototypes for first-semester students without prior programming experience? The authors provide a selection of relevant contructionism-inspired programming tools and provide indications on how one of them can enrich a user-centred design project for first-semester software and IT engineering students. They do this by describing the experiences from two runs of a HCI course and the concurrent semester projects. The students developed interactive touch-based learning apps for children in the fourth to sixth grade using App Inventor. Most of the project groups managed to do three iterations of specifying requirements, doing conceptual design, physical and interactive prototyping, and user evaluation. The groups implemented quite complex programs with multiple-screen switching, multiple interfaces, media such as pictures, animations and sound, database connection, Web-server connection, and integrated sensors, such as camera, accelerometer, etc. The students did a lot more project iterations and spent more time on the creative designs in real-life situations than the authors expected. This also allowed for the students' professional reflections on their prototypes, usability, interaction, and the design processes. All in all, this gave them a more profound real-life experience in the user-centred design process. The authors compare the two runs and suggest how to introduce contructionist prototype programming in a HCI course curriculum and conclude that contructionist programming tools can be a valuable addition to the teaching of HCI, and they suggest that further research should be conducted to explore how to best integrate these tools in order to optimize the students' learning capabilities.

In recent years, the development and the deployment of different generation systems such as 3G (UMTS, Wi-Fi), 4G (LTE, WIMAX), and 5G have become a reality for different telecommunication operators. At the same time, the development of new mobile devices with multiple interfaces have shifted the behavior of users concerning the utilization of the internet. Additionally, several streaming servers are available to provide real time applications such as e-commerce transactions, video streaming, online gaming, etc. As a result, the users have the privilege to use different multimedia applications at anytime and anywhere. The major trend in Beyond 4G networks is determining the best access network for the end user in terms of quality of service (QoS) during the network selection decision. Thus, in this article, the authors investigate graph theory and the AHP approach to deal with the network selection issue. The experimental results show that the proposed policy can achieve a significant performance in terms of QoS metrics for real-time streaming than conventional algorithms.
