Abstract Amitriptyline, administered orally, is currently one of the treatment options for the management of neuropathic pain and migraine. Because of the physicochemical properties of the molecule, amitriptyline is also a promising candidate for delivery as a topical analgesic. Here we report the dermal delivery of amitriptyline from a range of simple formulations. The first stage of the work required the conversion of amitriptyline hydrochloride to the free base form as confirmed by nuclear magnetic resonance (NMR). Distribution coefficient values were measured at pH 6, 6.5, 7, and 7.4. Solubility and stability of amitriptyline were assessed prior to conducting in vitro permeation and mass balance studies. The compound demonstrated instability in phosphate-buffered saline (PBS) dependent on pH. Volatile formulations comprising of isopropyl alcohol (IPA) and isopropyl myristate (IPM) or propylene glycol (PG) were evaluated in porcine skin under finite dose conditions. Compared with neat IPM, the IPM:IPA vehicles promoted 8-fold and 5-fold increases in the amount of amitriptyline that permeated at 24 h. Formulations containing PG also appear to be promising vehicles for dermal delivery of amitriptyline, typically delivering higher amounts of amitriptyline than the IPM:IPA vehicles. The results reported here suggest that further optimization of topical amitriptyline formulations should be pursued towards development of a product for clinical investigational studies. Extracellular vesicles (EVs) are nanosized lipid bilayer-delimited particles released from cells that mediate intercellular communications and play a pivotal role in various physiological and pathological processes. Subtypes of EVs may include plasma-membrane ectosomes or microvesicles and endosomal-origin exosomes, although functional distinctions remain unclear. EVs carry cargo proteins, nucleic acids (RNA and DNA), lipids, and metabolites. By presenting or transferring this cargo to recipient cells, EVs can trigger cellular responses. Here, we summarize what is known about EV biogenesis, composition, and function, with an emphasis on the role of EVs in cardiovascular system. Additionally, we provide an update on the function of EVs in cardiovascular pathophysiology, further highlighting their potential for diagnostic and therapeutic applications.

The present paper deals with the curvature (or splay) elasticity of lipid bilayers and its possible consequences for the microscopic organization of membranes. Experimental studies of the microstructure of lipid–lipid and lipid–protein mixtures are reported and discussed in terms of this type of membrane elasticity. In particular, evidence is provided that the elastic distortion of lipid bilayers caused by the incorporation of proteins can lead to mechanisms of the indirect protein–protein interaction. Finally the question is discussed whether these mechanisms could also play a role for the formation of enzyme complexes in biological membranes.

Organizational strategic feasibility studies are critical in mitigating uncertainty in the business environment and enhancing growth and performance. This study sought to ascertain the role of feasibility studies in the sustainability of supermarkets in Nairobi County, Kenya. The study's specific objectives were to determine the impact of technical feasibility, economic feasibility, operational feasibility, and legal feasibility on the sustainability of supermarkets in Nairobi County, Kenya. A descriptive research design was used for the study. The study's target population was seven major supermarkets: Naivas, QuickMart, Cleanshelf, The Game Store, Chandarana, Eastmatt, and Carrefour. A questionnaire was used to collect primary data. Findings indicated that technical feasibility is positively and significantly related to the business sustainability of supermarkets in Nairobi County. The findings also revealed that economic feasibility is positively and significantly related to the business sustainability of supermarkets in Nairobi County. The findings revealed a positive and significant relationship between operational feasibility and business sustainability of supermarkets in Nairobi County. Finally, the findings revealed that the legal feasibility and business sustainability of Nairobi County supermarkets are positively and significantly related. The study concluded that technical feasibility, economic feasibility, operational feasibility, and legal feasibility all have a positive impact on supermarket business sustainability. The study recommends that supermarkets should become acquainted with the technical technology that is available on the market and evaluate those that are appropriate for their needs. Managers of Nairobi County supermarkets increase the level of internal and external benchmarking in their supermarkets for economic benefit assessments. Background: The rapid flow of globalization makes the world free from barriers so that the existence of globalization gives birth to a new concept of "World Without Borders". This situation puts the character of millennials on the brink of collapse and needs to be straightened out. Based on the problems above, the researcher determines the question: How is the role of civic education in shaping millennial character in the era of globalization? Purpose: The purpose of this study is to determine the impact of the current globalization on society, as a form of knowledge of the importance of citizenship education as a shield in the era of globalization, knowing the role of civic education in shaping the character of the millennial generation. Method: The method used in this research is a literature review. The search for journals was carried out using the google scholar database with the keywords “citizenship educations”, “millennial generation”, and “globalization”. Feasibility studies are assessed based on title, abstract, and full text. Data analysis using qualitative analysis based on research findings. Results: The results of this study show the influence of globalization causes character problems in millennials increasing every year. The high nature of individualism, consumerism, and westernization shows the characteristics of Indonesia began to erode and eventually also impacted the moral damage. Therefore, prevention is necessary through citizenship education as the basis for character formation by the values of Pancasila.
