Integrative approach reveals new species of water bears (Pilatobius, Grevenius, and Acutuncus) from Arctic cryoconite holes, with the discovery of hidden lineages of Hypsibius

AbstractKristianstads Vattenrike Biosphere Reserve [KVBR] is a UNESCO designated area of Sweden possessing high biological value. Although several studies on tardigrades inhabiting Sweden have been performed, the KVBR area has been neglected. The current study investigates the tardigrade fauna of five areas of the biosphere reserve and includes 34 samples of different substrates analysed quantitatively and qualitatively. In total, 33 species of tardigrades were found in the samples, including 22 new records for the Skåne region, 15 new records for Sweden, and four species new to science. Mesobiotus emiliae sp. nov., Xerobiotus gretae sp. nov., Itaquascon magnussoni sp. nov., and Thulinius gustavi sp. nov. were described with an integrative approach (when possible) using morphological characters (light, electron scanning, and confocal laser scanning microscopies) and molecular markers (ITS2, 18S, 28S, cox1). A new protocol to increase morphological data was developed recovering mounted specimens within old slides for SEM analysis. Emended diagnoses for the genus Itaquascon and the transfer of Platicrista itaquasconoide to the genus Meplitumen are proposed. This study enriches the knowledge of the tardigrade biodiversity both within the KVBR and in Sweden and contributes to the rapidly increasing number of tardigrade species reported worldwide. The 33 species identified in the KVBR area represents 28% of all water bear species found in Sweden so far. The restricted study areas and limited number of samples collected suggests that the KVBR is very rich of tardigrades.

The Neotropical water scavenger beetle genus Chasmogenus Sharp, 1882 is a diverse lineage with 34 described species. Here, three new species from the southwestern margin of the Guiana Shield are described using an integrative approach combining adult morphology and molecular data: Chasmogenus clinatus sp. n. (Brazil: Amazonas, Venezuela: Amazonas), C. gironae sp. n. (Venezuela: Amazonas), and C. inpa sp. n. (Brazil: Amazonas). Examination of the male genitalia is essential for confident identifications and thus unassociated females may prove extremely difficult to identify. New distributional records in Brazil for C. acuminatus Smith & Short, 2020 are provided as well as habitus images of newly described species and an updated key to the species from Venezuela, Suriname, Guyana, French Guiana and northern Brazil.

AbstractThe new species Ramalina fleigiae from Brazil is described growing on rocks in riverbeds in high altitude grasslands of southern Brazil. It grows in areas with constant water flow, sometimes almost immersed, and always in exposed habitats. Through an integrative approach, the detailed description of R. fleigiae includes morphological, anatomical, ecological, chemical and molecular data. Ribosomal DNA-based phylogenies suggest that R. fleigiae is more closely related to a species that shares its habitat preference (R. laevigata) than to the morphologically and chemically similar R. exiguella and R. gracilis. Ramalina fleigiae and R. laevigata can be distinguished by thallus morphology (irregularly flat branches in R. fleigiae vs. flat to canaliculate in R. laevigata) and pattern of chondroid tissue, as genetic distances between them are compatible with the interspecific range. It is possible that many species of Ramalina still remain hidden within the morphological or chemical variation of currently accepted species. Combining ecological, anatomical and molecular data will improve our future understanding of this genus.

A new species, Diplolepis valtonyci Zhu, Wang & Pujade-Villar sp. nova, is described from Rosa rugosa Thunb. (1784) and R. davurica Pall. (1788) from China using an integrative approach based on molecular and morphological data. Diagnosis, distribution, and biology of the new species are given and illustrated. The phylogenetic relationship between D. valtonyci sp. nova and other Diplolepis species are assessed based on COI and Cytb genes. A key to the Chinese species of Diplolepis is provided.

Abstract Piresia, a small genus of herbaceous bamboos, has a geographical disjunction between the Caribbean and northern/western South America and the north-eastern Atlantic Forest in Brazil. Piresia leptophylla is reported from western Amazonia (WA) and the north-eastern Atlantic Forest (NAF), but its occurrence in western Amazonia is questionable. Using an integrative approach, we combined traditional morphological analysis, anatomy and niche modelling. The results revealed few macromorphological differences between WA and NAF specimens (only plant height, leaf length, lodicule dimensions, shape and position), contrasting with consistent differences in leaf anatomy (macrohairs and cruciform silica bodies in the costal zone of the adaxial/abaxial leaf surfaces, crenate silica bodies on the abaxial leaf surface, lack of panicoid hairs on the abaxial leaf surface, bicellular microhairs and lobed papillae over the abaxial leaf surface, and sparse but elongated fusoid cells in the mesophyll of WA specimens) and in niche patterns. The anatomical/micromorphological characters suggest environmental adaptations to the Amazonian and ‘restinga’ forests, respectively. We therefore propose the segregation of the WA populations into a new species, Piresia tenella sp. nov. We provide a formal description, photographs, a line illustration, a distribution map and discussion of the conservation status for the new species.

Integrative taxonomy tests the validity of taxa using methods additional to traditional morphology. The existence of two different morphotypes in specimens identified as Chrysotoxum vernale Loew (Diptera: Syrphidae) prompted their taxonomic study using an integrative approach that included morphology, wing and male-surstylus geometric morphometrics, genetic and ecological analyses. As a result, a new species is recognised, Chrysotoxum montanum Nedeljković & Vujić sp. nov., and C. vernale is re-defined. A lectotype and paralectotypes are designated for C. vernale to stabilize this concept. An additional species, Chrysotoxum orthostylum Vujić sp. nov., with distinctive male genitalia is also described. The three species share an antenna with the basoflagellomere shorter than the scape plus pedicel and terga with yellow fasciae not reaching the lateral margins. This study confirms the value of integrative approach for resolving species boundaries.

Members of the Monoctonina subtribe have long been neglected in applied studies of the subfamily Aphidiinae, due to their low economic importance, as they do not parasitize pests of cultivated plants. Consequently, data about this group are scarce, including its taxonomy and phylogeny. In the present study, we explore inter- and intraspecific genetic variation of Monoctonina species, including genera Monoctonus Haliday 1833, Monoctonia Starý 1962, Falciconus Mackauer 1959 and Harkeria Cameron 1900. We employ two molecular markers, the barcode region of the mitochondrial cytochrome c oxidase subunit I (COI) and the D2 region of the 28S nuclear gene (28S rDNA), to analyze genetic structuring and phylogeny of all available Monoctonina species, and combine them with morphological data for an integrative approach. We report one new species, and three potentially new species which can be formally described when further specimens are available. Analysis of phylogenetic relationships within the subtribe shows a basal position for the genera Falciconus and Monoctonia, and the close relatedness of Harkeria and Monoctonus.

The genus Obama Carbayo et al., 2013 includes 38 species, being the most species-rich within Geoplaninae. Species of this genus show a similar anatomy regarding their copulatory apparatus, which may hinder species differentiation. In this study, we describe two new species, presenting a marbled colour pattern, found in two different phytophysionomies of the Atlantic Forest, namely Semi-deciduous Forest and Araucaria Forest. Both species can be distinguished from their congeners, as well from each other, by colour pattern and eye arrangement combined with characteristics of the pharynx, penis papilla and prostatic vesicle, confirmed by molecular analyses from cytochrome c oxidase subunit I gene (COI). Phylogenetic analyses suggest that both species herein studied are closely related to another species that occurs in areas of Araucaria Forest (O. maculipunctata). Results also indicate the need to use at least 600 bp of the gene COI in the definitions of interspecific divergences and for species delineation, at least for the genus Obama.

Two new species of flatworm, collected from a beach at eastern Shenzhen, China, were studied through an integrative approach by combining morphological, histological, histochemical (acetylcholinesterase, AChE), and molecular (18S r- DNA) data. These species belong to two genera of marine triclads, previously unrecorded from China, viz. Nerpa Marcus, 1948 and Paucumara Sluys, 1989.        Nerpa fistulata Wang & Chen, sp. nov. is characterized by: transparent body; principally pentamerous intestine with three distinct commissures; two very large, prepharyngeal testis follicles; a semi-circular lens in each eye cup; a penis papilla provided with a chitinized, pointed stylet; lateral bursae communicating with the oviduct and opening ventrally to the exterior via a duct. Phylogenetically N. fistulata groups with one member of the family Bdellouridae. This new, Chinese species of Nerpa introduces a major geographic disjunction, as the type species N. evelinae was described from the bay of Santos, Brazil, so that the genus is now known from both Atlantic as well as Pacific coasts.        The species Paucumara falcata Wang & Li, sp. nov. is characterized by: three distinct pale yellow transverse pigmentation bands on its dorsal side, between which some snowflake-like specks are randomly distributed, and a brown transverse band anteriorly to the eyes; 8–11 testicular follicles on either side of the body, the follicles extending from immediately behind the ovaries to half-way along the pharyngeal pocket; a musculo-parenchymatic organ with a sclerotic, curved tip projecting from the anterior wall of the male atrium, ventrally to the root of the penis papilla. Phylogenetically P. falcata groups with its congener P. trigonocephala, with the genus Paucumara forming the sister taxon of the genus Ectoplana. Comparison of the nerve structure of P. falcata, as revealed by AChE histochemistry, with that of eight other species of triclad suggested that the nervous system of marine planarians is simpler than that of species of freshwater planarians, but revealed also that the nerve structure is rather variable among species.        The copulatory position exhibited by two partners in Paucumara falcata is remarkable in that they intertwine, with their heads pointing downwards and the tails pointing upwards, the entire process lasting about 10 min. Such a copulatory position has never before been reported for triclad flatworms.
