While studies have indicated an association between socioeconomic status (SES) and neuroimaging measures, weaker SES effects are shown for Blacks than Whites. This is, in part, due to processes such as stratification, racism, minoritization, and othering of Black people in the United States. However, less is known about Latino youth. This study had two aims: First, to test the association between parental education and the right and left nucleus accumbens (NAcc) resting-state functional connectivity with the frontoparietal network (FPN) in children; and second, to investigate ethnic heterogeneity in this association. This cross-sectional study used data from the Adolescent Brain Cognitive Development (ABCD) study. We analyzed the resting-state functional connectivity data (rsFC) of 10,840 US preadolescents who were between 9 and 10 years old. The main outcomes were the NAcc resting-state functional connectivity with FPN separately calculated for right and left hemispheres. Parental education was our independent variable. Family structure, sex, and age were covariates. Furthermore, ethnicity (Latino vs. non-Latino) was regarded as the moderator. We used mixed-effects regression for data analysis with and without interaction terms between parental education and ethnicity. Most participants (n = 8690; 80.2%) were non-Latino and 2150 (19.8%) were Latino. Parental education was associated with higher right and left NAcc resting-state functional connectivity with FPN. Ethnicity showed statistically significant interactions with parental education, suggesting that the positive associations between parental education and right and left NAcc resting-state functional connectivity with FPN were different in non-Latino and Latino children. For right hemisphere, we found significantly stronger and for left hemisphere, we found significantly weaker association for Latino compared with non-Latino preadolescents. Preadolescents’ NAcc resting-state functional connectivity with FPN depends on the intersections of ethnicity, parental education, and laterality.

AbstractExtensive research has demonstrated that rs1360780, a common single nucleotide polymorphism within the FKBP5 gene, interacts with early-life stress in predicting psychopathology. Previous results suggest that carriers of the TT genotype of rs1360780 who were exposed to child abuse show differences in structure and functional activation of emotion-processing brain areas belonging to the salience network. Extending these findings on intermediate phenotypes of psychopathology, we examined if the interaction between rs1360780 and child abuse predicts resting-state functional connectivity (rsFC) between the amygdala and other areas of the salience network. We analyzed data of young European adults from the general population (N = 774; mean age = 18.76 years) who took part in the IMAGEN study. In the absence of main effects of genotype and abuse, a significant interaction effect was observed for rsFC between the right centromedial amygdala and right posterior insula (p < .025, FWE-corrected), which was driven by stronger rsFC in TT allele carriers with a history of abuse. Our results suggest that the TT genotype of rs1360780 may render individuals with a history of abuse more vulnerable to functional changes in communication between brain areas processing emotions and bodily sensations, which could underlie or increase risk for psychopathology.

Abstract Background The growing abuse of amphetamine-type stimulants leads to new challenges to human health. A possible addiction mechanism has been proposed by altered functional architecture of the nucleus accumbens (NAc) during resting state. NAc contains different subdivisions and they may play different roles in addiction. The aim of the present study was to examine whether there are common or distinct patterns of functional connectivity of the NAc subdivisions in amphetamine-type stimulant abusers (ATSAs). Methods The present study recruited 17 male ATSAs and 22 healthy male controls. All the subjects underwent resting-state functional magnetic resonance imaging (fMRI) with their eyes closed. The NAc was divided into core-like and shell-like subdivisions. We used seed-based resting-state functional connectivity (RSFC) analyses to identify differences in brain functional architecture between ATSAs and healthy controls (HCs). Results ATSAs had lower positive RSFCs with all of the NAc subdivisions over the left orbital part of superior frontal gyrus and higher positive RSFCs with the NAc subdivisions over the left opercular part of inferior frontal gyrus than HCs, which indicates common abnormalities across the NAc subdivisions in ATSAs. In addition, the RSFCs between the NAc subdivisions and the left orbital part of superior frontal gyrus were negatively correlated with the addiction severity in ATSAs. Conclusion These results provide evidence that there are common RSFC patterns of the NAc subdivisions in ATSAs. The abnormality indicated by disrupted functional connectivity between the NAc subdivisions and prefrontal cortex suggests abnormal interaction between the rewarding process and cognitive control in ATSAs. Our results shed insight on the neurobiological mechanisms of ATSA and suggest potential novel therapeutic targets for treatment and intervention of ATSAs.

Background: Changes in resting state functional connectivity between the insula and dorsal anterior cingulate cortex as well as between the insula and nucleus accumbens have been linked to nicotine withdrawal and/or administration. However, because many of nicotine’s effects in humans appear to depend, at least in part, on the belief that nicotine has been administered, the relative contribution of nicotine’s pharmacological actions to such effects requires clarification. Aims: The purpose of this study was to examine the impacts of perceived and actual nicotine administration on neural responses. Methods: Twenty-six smokers were randomly assigned to receive either a nicotine inhaler (4 mg deliverable) or a nicotine-free inhaler across two sessions. Inhaler content instructions (told nicotine vs told nicotine-free) differed across sessions. Resting state functional connectivity between sub-regions of the insula and the dorsal anterior cingulate cortex and nucleus accumbens was measured using magnetic resonance imaging before and after inhaler administration. Results: Both actual and perceived nicotine administration independently altered resting state functional connectivity between the anterior insula and the dorsal anterior cingulate cortex, with actual administration being associated with decreased resting state functional connectivity, and perceived administration with increased resting state functional connectivity. Actual nicotine administration also contralaterally reduced resting state functional connectivity between the anterior insula and nucleus accumbens, while reductions in resting state functional connectivity between the mid-insula and right nucleus accumbens were observed when nicotine was administered unexpectedly. Changes in resting state functional connectivity associated with actual or perceived nicotine administration were unrelated to changes in subjective withdrawal and craving. Changes in withdrawal and craving were however independently associated with resting state functional connectivity between the nucleus accumbens and insula. Conclusions: Our findings highlight the importance of considering non-pharmacological factors when examining drug mechanisms of action.
