Metabolome and transcriptome analyses unravel the inhibition of embryo germination by abscisic acid in pear

This study was initiated to test the embryogenic potential of immature cotyledons (3-5 mm long) of Texas bluebonnet (Lupinus texensis). The embryo initiation medium consisted of B5 salts and vitamins with 3% sucrose and 22.6 μM 2,4-D alone or in combination with 1-15 μM of various cytokinins. Within 15-20 days, globular embryos were formed on the distal end of the cotyledons. Eventually the entire cotyledon surface was covered by embryo-like structures. Addition of cytokinins to the medium did not increase the percentage of cultures which formed embryos. In fact, addition of thidiazuron severely inhibited embryogenesis. Following transfer to an embryo maturation medium (MS medium with 0.38 μM abscisic acid alone or in combination with benzyladenine or zeatin) for 10-14 days, the embryos were placed in MS medium supplemented with GA (2.9 μM) or glutamine (200 mg/liter) with or without activated charcoal (0.5%) for embryo germination and plantlet development. Most of the embryos exhibited precocious germination and well-developed roots but failed to produce normal shoots. Therefore, additional work is needed to improve embryo conversion frequency.

AbstractThis work presents the results of physiological studies developed to understand modifications linked to the reduction of seed dormancy provoked by domestication processes. The experiments performed compared wild and domesticatedCucurbitasubspecies and their hybrids developed by reciprocal crossings. Seeds of two accessions of the wild subspecies presented dormancy, but it was largely reduced in seeds from the domesticated genotype, and partially reverted in hybrids, especially in those obtained when the domesticated genotype was used as the mother plant. In addition, naked embryos of all subspecies did not display dormancy when incubation was performed at 28°C, but embryo germination was progressively reduced only in the wild genotype under decreasing incubation temperatures (22 and 16°C). In the embryos, abscisic acid (ABA) concentrations were similar in both domesticated and wild subspecies, whereas in the seed coat, it was threefold higher in the wild subspecies. The naked embryos from the wild subspecies were far more responsive to ABA than those from the domesticated subspecies. These results indicate that dormancy in the wild subspecies is imposed by the seed coat tissues and that this effect is mediated by their high ABA content and the sensitivity of the embryos to ABA. These physiological aspects were apparently removed by domestication along with the temperature-dependent response for germination.

An enantioselective synthesis of (+)-8′-demethyl ABA (2) is described. The chiral intermediate 7 was prepared by yeast reduction of a substituted monoprotected cyclohexa-2,5-dien-1,4-dione (9) synthesized through a phenol oxidation. The scope and limitations of the phenol oxidation is described. 8′-Demethyl ABA shows ABA-like activity in wheat embryo germination inhibition, showing that the 8′-methyl group is not essential for biological activity.
