ObjectiveThe aim of this study was to assess age-related changes in cardiac autonomic modulation and heart rate variability (HRV) and their association with spontaneous and pharmacologically induced vulnerability to cardiac arrhythmias, to verify the translational relevance of mouse models for further in-depth evaluation of the link between autonomic changes and increased arrhythmic risk with advancing age.MethodsHeart rate (HR) and time- and frequency-domain indexes of HRV were calculated from Electrocardiogram (ECG) recordings in two groups of conscious mice of different ages (4 and 19 months old) (i) during daily undisturbed conditions, (ii) following peripheral β-adrenergic (atenolol), muscarinic (methylscopolamine), and β-adrenergic + muscarinic blockades, and (iii) following β-adrenergic (isoprenaline) stimulation. Vulnerability to arrhythmias was evaluated during daily undisturbed conditions and following β-adrenergic stimulation.ResultsHRV analysis and HR responses to autonomic blockades revealed that 19-month-old mice had a lower vagal modulation of cardiac function compared with 4-month-old mice. This age-related autonomic effect was not reflected in changes in HR, since intrinsic HR was lower in 19-month-old compared with 4-month-old mice. Both time- and frequency-domain HRV indexes were reduced following muscarinic, but not β-adrenergic blockade in younger mice, and to a lesser extent in older mice, suggesting that HRV is largely modulated by vagal tone in mice. Finally, 19-month-old mice showed a larger vulnerability to both spontaneous and isoprenaline-induced arrhythmias.ConclusionThe present study combines HRV analysis and selective pharmacological autonomic blockades to document an age-related impairment in cardiac vagal modulation in mice which is consistent with the human condition. Given their short life span, mice could be further exploited as an aged model for studying the trajectory of vagal decline with advancing age using HRV measures, and the mechanisms underlying its association with proarrhythmic remodeling of the senescent heart.

Abstract Objectives The present work was designed to study the modulatory effects of algogen-induced vasosensory reflex responses on short-term heart rate variability (HRV) parameters in naïve and vagotomized rat models. Methods In this study, vasosensory reflex responses were elicited by instilling algogens (bradykinin/histamine), a component of inflammatory mediators into a local segment of medium-sized peripheral blood vessel (femoral artery) while a continuous electrocardiogram (ECG) was recorded. Short-term (5 min) ECG segments obtained from original recordings were examined in detail and relevant data of HRV parameters were pooled. Time domain and frequency domain analyses were performed using dedicated software (LabChart 8, AD Instruments®, Australia) and results were analyzed. Results Bradykinin-induced vasosensory reflexes caused significant alterations in both time domain and frequency domain HRV parameters as compared to the time-matched saline control group. Instillation of bradykinin caused a transient increase in NN interval, RMSSD, TSP, HF power (HFP) along with a decrease in the standard deviation of all normal NN intervals (SDNN), SDNN/RMSSD, LF power (LFP), LFP/HFP. Histamine produced a similar pattern of responses, but HRV alterations were less pronounced compared to those with bradykinin. Further analysis revealed that algogen-induced vasosensory reflex responses caused an increase in the parasympathetic influence on the heart accompanied by a decrease in sympathetic influence. In addition, HRV modulation by algogen-induced vasosensory reflexes was significantly attenuated in vagotomized rats, illustrating the principal role of vagus in the reflex HRV modulation. Conclusions The present study proposes a novel hypothesis regarding the cardio-protective role of inflammatory mediators during acute stress, by potentiating the vagal impact and attenuating the sympathetic impact on the heart.

Introdução: Existem poucos estudos que evidenciam a manipulação vertebral relacionada à modulação autonômica cardíaca. Objetivo: Revisar a literatura sobre os efeitos da manipulação vertebral sobre a modulação autonômica cardíaca. Métodos: Foi realizada uma busca bibliográfica nas bases de dados da saúde Medline, Pubmed e Cinahl, no período correspondido entre setembro e novembro de 2014. Foram utilizados os descritores em inglês Spinal Manipulation, Cardiac Autonomic Modulation, Autonomic Nervous System, Heart Rate Variability, além de associações entre eles. Resultados: Foram encontrados 190 artigos, sendo excluídos 39 por serem repetidos, restando 151. Destes, 124 não se encaixaram nos critérios de inclusão e após leitura crítica e análise dos materiais foram selecionados 7 artigos. Grande parte dos estudos revelou que a manipulação da coluna, independente do segmento, demonstra alterações autonômicas, tanto em nível simpático quanto parassimpático. Conclusão: Existem diferentes metodologias para avaliação da modulação autonômica cardíaca, sendo a Variabilidade da Frequência cardíaca através do eletrocardiograma a mais utilizada. A manipulação vertebral exerceu influência, na maioria dos artigos, sobre a modulação autonômica cardíaca.Palavras-chave: manipulação da coluna, sistema nervoso autônomo, variabilidade da frequência cardíaca.
