AbstractParasites of the genusTrypanosomaare microorganisms that display wide morphological, biological and genetic variability. Here we present the first description of an isolate of the genusTrypanosomanaturally infecting the tickAmblyomma brasiliense. The ticks were collected from a specimen ofTayassu pecari(Queixada, white-lipped peccary) from the Itatiaia National Park, Itatiaia, Rio de Janeiro, Brazil. The isolate was characterized by molecular, morphometric and biological analyses. ATrypanosomaculture was isolated from crushed nymphal and adult ticks, propagated in the tick cell line IDE8 and maintained in L15B culture medium, incubated at 32 °C. The isolate grew well in L15B medium at 30, 32 and 34 °C but not at lower or higher temperatures. The culture remained stable in axenic L15B medium at 30 °C. Cryopreserved cultures retained viability after cryopreservation in liquid nitrogen. Growth in axenic medium and developmental forms of the trypanosomes were analysed. Analysis of the 18S rDNA region confirmed the authenticity of this new species and the nucleotide sequence was deposited in Genbank. The species was namedTrypanosoma amblyommisp. nov. strain C1RJ. Characteristics related to pathogenicity, involvement with vertebrate hosts, epidemiology, developmental cycle and transmission mechanisms are still unknown. Therefore, further studies are necessary to understand the aspects of the biological cycle ofT. amblyommisp. nov.

This study describes an isolate of the genusTrypanosomanaturally infectingRhipicephalus microplusticks, characterized through molecular, morphological and biological analysis. Trypanosome cultures, designated strain P1RJ, were obtained by isolation fromR. microplushaemolymph in cultures of the tick cell line IDE8. After isolation, strain P1RJ grew well axenically in L15B medium at temperatures of 30, 32 and 34 °C. The new trypanosome remained stable in axenic culture over 14 passages in L15B at 30 °C and was successfully cryopreserved and resuscitated. Morphometric analysis was performed on randomly selected developmental forms. 18S rRNA and 24Sα rDNA sequence analyses confirmed that strain P1RJ is a new species of the genusTrypanosoma. The nucleotide sequences described were submitted to Genbank. Pathogenicity, involvement in vertebrate hosts, epidemiology, developmental cycle and transmission mechanisms of strain P1RJ are still unknown. Therefore, more studies will be necessary to determine life cycle aspects of this trypanosome, for which we propose the nameTrypanosoma rhipicephalissp. nov.

Abstract Two lineages of Rhipicephalus sanguineus are known in Brazil: the temperate or southern and the tropical or northern populations. The distribution patterns of both lineages of R. sanguineus have epidemiological implications that can affect vectorial competence concerning Ehrlichia canis, the agent of canine monocytic ehrlichiosis. Intending to identify the microbiomes of both lineages and compare microorganisms in R. sanguineus, we used the 16S rRNA (V4-V5 region) gene-based metataxonomic approach, through NGS sequencing on the MiSeq Illumina platform. We selected specimens of females from the environment and samples of primary embryonic cell cultures, from both lineages, and this was the first study to investigate the prokaryotic microbiome in tick cell cultures. The results showed that many bacterial taxa detected in the samples were typical members of the host environment. A significant diversity of microorganisms in R. sanguineus females and in embryonic cell cultures from both lineages was found, with emphasis on the presence of Coxiella in all samples, albeit in different proportions. The Coxiella species present in the two lineages of ticks may be different and may have co-evolved with them, thus driving different patterns of interactions between ticks and the pathogens that they can harbor or transmit to vertebrate hosts.

A new species of the freshwater amphipod Hyalella Smith, 1874 is described for the plateau of the Itatiaia National Park, located between the states of Minas Gerais and Rio de Janeiro, southeastern Brazil. This is the first Brazilian species of Hyalella found at more than 2,200 meters of altitude. The specimens were found in a small stream, buried under rocks, in the higher area of the Park. The main morphological characteristics that differentiate the new species are the extreme reduction of the size of the uropod 3, absence of apical setae on telson, absence of comb-scales on gnatopods 1 and 2, absence of curved seta on inner ramus of uropod 1 and sternal gills tubular on pereonites 3 to 7. The new species presents similarities with some cave species of the genus, apparently being troglophile and that is the process of adaptation to the hypogean habitat. The importance of creating new protected areas for the conservation of Hyalella in Brazil is discussed.

Proisotoma ramosi Arlé is redescribed based on specimens collected for the first time at National Park of “Serra dos Órgãos”, Teresópolis, State of Rio de Janeiro. This species is easily identified by a set of characters: color white, 2+2 eyes, PAO long and narrow, labral setae pattern 4/554, external lobe of maxilla with bifurcate palp and 3 sublobal setae, sensillar pattern 53/22335 by half tergite, retinaculum with 4+4 teeth and two setae on corpus, manubrium with two anterior-distal setae, dens with 7 posterior and 11 anterior setae, mucro bidentate. Neotype for P. ramosi is designated.

Knowledge on Poduromorpha fauna from the littoral of Rio de Janeiro mainly come from studies conducted in “restinga” areas of Itaipuaçu, Maricá, and Marambaia. In this study two “restinga” areas were sampled for the first time to increase the taxonomic knowledge of Poduromorpha in littoral areas of Rio de Janeiro: Costa do Sol State Park and Restinga de Jurubatiba National Park, located respectively at Lake Region (“região dos lagos”) and at northern Rio de Janeiro State. The analysis of the material showed the presence of six families, 14 genera and 20 species. All records are new for both areas; Neanuridae had the highest diversity, with seven genera and 10 species. One of them, Paleonura Cassagnau, 1982, was registered for the first time in the littoral of Rio de Janeiro. A key was proposed for the Poduromorpha genera found in the littoral of Rio de Janeiro.

The northern coast of the state of Rio de Janeiro harbors the largest coastal sandy plains (restingas) of southeastern Brazil, severely threatened due to recent human occupation. In this context, mammal inventories becomes urgent to support biodiversity conservation and management policies. This paper aims to update the knowledge on the mammalian species richness of Restinga de Jurubatiba National Park by analyzing pellets of the Barn owl Tyto furcata. The analyses of 4745 mammalian bone fragments (1739 individuals) revealed the occurrence of 17 species belonging to the orders Rodentia (9 spp.), Didelphimorphia (2 spp.) and Chiroptera (6 spp.). The rodents Mus musculus and Cerradomys goytaca accounted for 77.2% of the total individuals preyed, suggesting that the Barn owl hunts in both disturbed and natural open shrub formations. A compilation of previous records, together with those provided in the present study, indicates the occurrence of 44 species of mammals in the Restinga de Jurubatiba National Park, of which six (13.6%) were exclusively detected in pellets. These new records include species adapted to open formations of the interior of Brazil that were not previously reported in restingas on the basis of voucher-specimens, such as the rodents Necromys lasiurus and Calomys tener, the latter being recorded for the first time in the littoral of Rio de Janeiro state. This study underscores the importance of owl pellet analysis as a complementary and indispensable method for inventorying mammals. The results highlight the Restinga de Jurubatiba National Park as one of the restingas in Southeastern Brazil with the highest number of mammal species.

We present new records of Passifloraceae sensu stricto from Itatiaia National Park, Brazil, contributing new data on the geographic distribution of four Passiflora L. species to the flora of Brazil: P. deidamioides Harms, P. miersii Mast., P. rhamnifolia Mast., and P. setulosa Killip. The last three species are reported from Itatiaia National Park and the last species from the state of Rio de Janeiro for the first time.
