Influence of the lactoperoxidase system on susceptibility of the udder toStreptococcus uberisinfection

SummaryLactoperoxidase (LP), thiocyanate (SCN-), pH and somatic cell counts (SCC) were measured in mammary secretions from 20 cows collected 14 d before drying-off, 7 and 21 d after drying-off, and 3–18 d postcalving. The inhibitory activity of the secretions onStreptococcus uberiswas determined and the susceptibility of the udder to infection by this organism was tested by intramammary infusion of 250 colony forming units at the above stages. LP, SCN-, pH and SCO increased during involution and fell postcalving. The secretions collected before drying-off, 7 d after drying-off and postcalving inhibited growth ofStr. uberis.; those collected 21 d after drying-off did not. Inhibitory activity in pre-drying-off secretions was destroyed by heating and restored by addition of LP, glucose and glucose oxidase, but addition of these substances to secretion 21 d after drying-off did not provide a full inhibitory system. The growth ofStr. uberisin the secretions was correlated with intramammary susceptibility, since challenges withStr. uberisat 14 d before drying-off, at 7 and 21 d after drying-off and postcalving led to 43·8, 25·0, 81·3 and 37·5% of quarters becoming infected. It is suggested that the LP/SCN-/H2O2system plays a role in protecting the lactating mammary gland from infection withStr. uberisbut becomes ineffective as involution progresses.

The use of somatic cell counts (SCCs) for the diagnosis of mastitis is not a well-established procedure for the caprine species, because nonleucocytic cell-like particles are normally observed as a result of the apocrine secretion process of the goat mammary gland. The infection levels of 124 goats were measured by the β-glucuronidase test, which was compared with the SCC method and the California mastitis test (CMT). Seventy-nine of 124 samples (63.7%) showed SCCs lower than 1.3 × 103 cells per ml. Of these samples, 93% showed low levels of β-glucuronidase activity (<15 U/ml). In the remaining 36.3% of the samples, SCCs were higher than 1.3 × 103 cells per ml. Of these samples, 88% showed high levels of β-glucuronidase activity (15 to 100 U/ml). The CMT gave similar results. In this study, the β-glucuronidase test was standardized for goat milk and shown to be reliable, enabling one to count only the somatic enzyme cells in milk and avoiding the interference encountered with the SCC method.

Mastitis is a complex disease causing inflammation of the udder, which has been estimated to cost the dairy farmer between £40-£117/cow per year (Stott et al., 2002). Economic loss occurs as a result of discarded milk, reduced milk yield and milk quality, increased vet costs and an increase in replacement costs. The objective of this study was to examine the effect of breed on the incidence of mastitis and somatic cell counts and milk production capabilities of Holstein Friesian, Brown Swiss and Brown Swiss crossbred cows.

Objectives were to investigate somatic cell counts (SCC) and total bacterial counts (TBC) in the raw bulk-tank milk of sheep flocks in Greece, to study factors potentially influencing increased SCC and TBC in the bulk-tank milk of sheep and to evaluate possible associations of SCC and TBC with milk content. Throughout Greece, 325 dairy sheep flocks were visited for collection of milk sampling for somatic cell counting, microbiological examination and composition measurement. Geometric mean SCC were 0.488 × 106 cells mL−1; geometric mean TBC were 398 × 103 cfu mL−1; 228 staphylococcal isolates were recovered form 206 flocks (63.4%). Multivariable analyses revealed annual incidence risk of clinical mastitis, age of the farmer and month into lactation period (among 53 variables) to be significant for SCC > 1.0 × 106 cells mL−1 and month into lactation period at sampling and availability of mechanical ventilators (among 58 variables) to be significant for TBC > 1500 × 103 cfu mL−1. Negative correlation of SCC with fat, total protein and lactose and positive correlation of SCC with added water were found. With SCC > 1.0 × 106 cells mL−1, significant reduction of protein content (2%) was observed, whilst in flocks with SCC > 1.5 × 106 cells mL−1, significantly lower annual milk production per ewe (42.9%) was recorded.

One hundred and seventy-seven (177) bulk tank milk samples were analyzed with a commercially available real-time polymerase chain reaction kit and 11 (6.21%), 41 (23.16%), and 58 (32.77%) tested positive for Mycoplasma bovis, Staphylococcus aureus, and Streptococcus agalactiae, respectively. Statistical analysis revealed a significant relationship between the presence of S. aureus and S. agalactiae. Enumeration of somatic cells was performed in the same samples by flow cytometry. The somatic cell counts were found higher in S. aureus and S. agalactiae positive samples. No association was found between M. bovis presence and somatic cells counts. Low internal assay control Ct values were found to be related with high somatic cell counts. Noticeably, this is the first report for the presence of M. bovis in Cyprus. Therefore, its presence was confirmed by bulk tank milk culture, conventional PCR, and next generation sequencing. Furthermore, M. bovis was typed with multilocus sequencing typing and was allocated to sequence type 29 (ST 29). Real-time PCR in bulk tank milk samples is a useful tool to detect mammary infections, especially for neglected pathogens such as M. bovis.
