It's always refreshing to read about two old techniques being combined in a novel way to yield new information. Osamu Ohtani and Yuko Ohtani recently accomplished this. Louis Ranvier probably introduced the use of silver nitrate as a histology stain over a century ago. Whereas Ranvier is best known for describing nodes along the nerve fiber, he also demonstrated that silver nitrate stained endothelial cells, allowing their borders to be clearly visualized. In the 1970's, Takuro Murakami first used the scanning electron microscope (SEM) to examine corrosion casts of the vasculature. Much of what we currently know about the microarchitecture of blood vessels has been discovered using this technique.

Silver nitrate and silver proteinate were both successfully used as electron stains for the demarcation of the aortic endothelial cell borders in the scanning electron microscope. By energy-dispersive X-ray analysis it was demonstrated that silver was present in the demarcation lines around the endothelial cells. The presence of silver closely coincided with the places where secondary electrons were produced. Judged by the quality of the aortic tissue in the transmission electron microscope, the method using silver proteinate was preferable to those using silver nitrate. The limited resolution of the method is demonstrated. The role of the dextran solution in such procedures was investigated, and it was shown that the presence of such a dextran solution, or its particle size or charge, does not contribute to the formation of these silver lines around the endothelial cells. In ultrathin sections of such material the cell surface-contrasting capacity of colloidal thorium dioxide solutions was compared to the cell border-contrasting capacity of silver proteinate. It was found that prior aldehyde fixation abolished the cell border demarcation by silver proteinate, but only reduced the cell surface demarcation by thorium dioxide particles.

Summary Three-dimensional observation of the microcirculation of the tendon was readily and clearly demonstrated by preparing methyl methacrylate casts and observing them under the scanning electron microscope. In the muscles blood vessels made a network like a ladder surrounding every muscle fibre. The fibrous digital sheath had blood vessels made in a fine meshed cylinder. By microdissection of the vessels of the sheath the blood vessels of the vinculae and tendons were observed stereoscopically in relation to the peritendinous tissues. The casting method contributes to better understanding of vascular architecture of tendons.

In recent years, endovascular treatment has often been used to treat cerebral aneurysms. Basic investigation to elucidate the process of endothelial cell growth within aneurysms is a crucial problem. We performed elecrtro-detachable coil embolization in aneurysms under administration of blood coagulation factor XIII, a wound-healing accelerator, and examined changes in endothelial cells on the surface of the inserted coil with a scanning electron microscope. Experimental aneurysms produced in animals were treated by coil embolization and histological changes in embolized coil after the treatment of factor XIII were investigated. Aneurysms were produced in four matured swines under general anesthesia and coil embolization was performed with a elecrtro-detachable coil. The aneurysms were resected immediately and 3 weeks after coil embolization, and investigated histologically with a scanning electron microscope. Seven animals were administrated with blood coagulation factor XIII on the day of embolized and the following 4 consecutive days. These aneurysms were also resected 3 weeks after embolization and investigated histologically. More marked fibroblast proliferation and growth of endothelial cells on the surface of luminal side of embolized coil were seen in the aneurysms in the group administrated with factor XIII than in aneurysms treated without factor XIII. More effective and sufficient coil embolization can be obtained by administration of factor XIII.

The distribution of the integumentary papillae of the cercaria ofHimasthla secundahas been studied by a variety of techniques. Structures stained by silver nitrate and visible under the light microscope correspond in their spatial distribution with papillae observed under the scanning electron microscope. The tegumentary papillae described with the light and scanning electron microscope are correlated with the specialized nerve endings in the tegument as seen in transmission electron microscopy. The ultrastructure of these papillae is examined by conventional transmission electron microscopy and the probability that these structures are sensory is discussed.
