<p>A boronic acid-based anthracene fluorescent probe was functionalised with an acrylamide unit to incorporate into a hydrogel system for monosaccharide detection<i>. </i>In solution, the fluorescent probe<b> </b>displayed a strong fluorescence turn-on response upon exposure to fructose, and an expected trend in apparent binding constants, as judged by a fluorescence response where D-fructose > D-galactose > D-mannose > D-glucose. The hydrogel incorporating the boronic acid monomer demonstrated the ability to detect monosaccharides by fluorescence with the same overall trend as the monomer in solution with the addition of fructose resulting in a 10-fold enhancement (≤ 0.25 M). <b><u></u></b></p>

It has been shown that glomerular ANG II receptors are downregulated and protein kinase C (PKC) activity is enhanced in diabetes mellitus. Therefore, we investigated glomerular and preglomerular vascular ANG II receptors and PKC isoform regulation in streptozotocin (STZ)-diabetic rats treated with insulin and/or captopril. Diabetic rats were prepared by injecting STZ (60 mg/kg). Those that developed diabetes after 48 h were treated with low or high doses of insulin, or with a low dose of insulin as well as captopril, and killed 14 days later. Their glomeruli and preglomerular vessels were purified, competitive binding studies were performed by using the ANG II antagonists losartan and PD-123319, and PKC analysis was carried out by Western blotting. Competitive binding studies showed that the AT1 receptor was the only ANG II receptor detected on both glomeruli and preglomerular vessels of all groups. Preglomerular vascular AT1 receptor density (Bmax) was significantly upregulated in low insulin-treated STZ rats, whereas glomerular AT1 Bmax was downregulated. Furthermore, both the captopril- and high insulin-treated groups had less glomerulosclerosis and vascular damage than the low insulin-treated group. PKCα, PKCδ, PKCε, and PKCμ isoforms found in preglomerular vessels were upregulated by captopril and high insulin doses, respectively, whereas no such regulation occurred in glomeruli. We conclude that in STZ-diabetic rats ANG II receptors and PKC isoforms on preglomerular vessels and glomeruli are differentially regulated by treatment with insulin and/or captopril.

A series of polystyrenes randomly labeled with 1-pyrenebutanol were prepared by copolymerizing styrene and 1-pyrenebutylacrylate yielding the CoBuE–PS series. Solutions of CoBuE–PS were prepared in nine organic solvents having viscosities ranging from 0.36 to 5.5 mPa·s and the fluorescence spectra and pyrene monomer and excimer fluorescence decays were acquired. Analysis of the fluorescence spectra yielded the IE/IM ratio, whereas analysis of the fluorescence decays with the fluorescence blob model (FBM) yielded the parameters N blobo , <kblob × Nblob> , and k blobo . These parameters were compared to those obtained with two other series of pyrene-labeled polystyrenes, which had been studied earlier, namely CoA–PS and CoE–PS where pyrene was attached to the polymer backbone via a methylamide and benzyl methylether linker, respectively. Although the parameters IE/IM, N blobo , <kblob × Nblob>, and k blobo took different values according to the specific nature of the linker connecting pyrene to the polystyrene backbone, they exhibited trends that were quite similar for all the pyrene-labeled polystyrene constructs. The excellent agreement between the parameters retrieved for the three different types of pyrene-labeled polystyrenes suggests that the FBM accounts satisfyingly for differences in the nature of the label used, while still retrieving information pertinent to the polymer of interest.
