Heat shock protein 27 (Hsp27) has an established role in tumor progression and chemo-resistance of castration-resistant prostate cancer (CRPC). Hsp27 protects eukaryotic translation initiation factor 4E (eIF4E) from degradation, thereby maintaining survival during treatment. Phenazine derivative compound #14 was demonstrated to specifically disrupt Hsp27/eIF4E interaction and significantly delay castration-resistant tumor progression in prostate cancer xenografts. In the present work, various strategies of encapsulation of phenazine #14 with either DOTAU (N-[5′-(2′,3′-dioleoyl)uridine]-N′,N′,N′-trimethylammonium tosylate) and DOU-PEG2000 (5′-PEG2000-2′,3′-dioleoyluridine) nucleolipids (NLs) were developed in order to improve its solubilization, biological activity, and bioavailability. We observed that NLs-encapsulated phenazine #14-driven Hsp27-eIF4E interaction disruption increased cytotoxic effects on castration-resistant prostate cancer cell line and inhibited tumor growth in castration-resistant prostate cancer cell xenografted mice compared to phenazine #14 and NLs alone. Phenazine #14 NL encapsulation might represent an interesting nanostrategy for CRPC therapy.

The admiration for colours by man has made dyes to have very useful applications on a great variety of materials. Pyocyanin (PCN) or 5-methyl-1-hydroxyphenazine, a phenazine derivative biosynthesized by Pseudomonas aeruginosa isolated from the stressed soil of a farmland at Awka was used to prepare three azo dyes namely, pyocyanin azophenol, pyocyanin azo-1-naphthol and pyocyanin azo-2-naphthol. The PCN was treated with SOCl2 to convert it to the chloro analogue. Treatment of chloro analogue with NaCN in ethanol and subsequent acid hydrolysis gave the carboxylic acid from which the amide was obtained after treatment with (NH4)2CO3 and heat. The amide was converted to the amine by Hoffman’s amide degradation. The amine was diazotized and coupled with phenol, 1-naphthol and 2-naphthol to give the three azo dyes respectively. UV-VIS and infrared spectra of the compounds agreed with the assigned structures. Acute Toxicity (LD50) tests showed the compounds to be reasonably non-toxic. Dyeing potentials of the Pyocyanin (PCN), PCN azophenol, PCN azo-1-naphthol and PCN azo-2-naphthol by mordant dyeing on cotton, silk and nylon textile materials showed them to have blue, dark green, purplish-brown and red colours respectively. The dyes were reasonably fast to light, washing, rubbing, alkali and acid. Pyocyanin, Pyocyanin azophenol, Pyocyanin azo-1-naphthol and pyocyanin azo-2-naphthol produced from Pseudomonas aeruginosa isolated from farmland soil can be used effectively to dye cotton, silk and nylon textile materials.

: Resistance to antibiotics has been widely existed in the health care and community setting, thus developing a novel aspect of new antibiotics is urgently necessary. Methicillin-resistance Staphylococcus aureus (MRSA) Pyruvate kinase (PK) is crucial to the survive of bacterial, making it a novel antimicrobial target. In the past decade, most reported PK inhibitors including indole, flavonoid, phenazine derivative from natural product small molecules or their analogues, or virtual screening from small molecule compound library. This review covers the PK inhibitors and their antimicrobial activities reported from the beginning of 2011 through the middle of 2020. The Structure Activity Relationships (SARs) was discussed briefly as well.
