The radiation-inactivation method allows the determination of the Mr of enzymes and receptors by monitoring the decay of biological activity as a function of absorbed dose. The presence of regulatory or effector proteins (inhibitors or activators) associated with an enzyme or receptor, or released in the preparation after tissue homogenization, may affect the decay of biological activity. How the activity is affected, however, will depend on the type of inhibition (competitive or non-competitive), the inhibitor or activator concentration, the dissociation constant of the enzyme-effector system, and the effector Mr relative to that of the enzyme. Since little is known on how effector proteins influence radiation inactivation of enzymes and receptors, we have considered a theoretical model in an effort to provide a framework for the interpretation of experimentally obtained data. Our model predicts that competitive and non-competitive inhibitors of enzymes could be distinguished by analysing irradiated samples with various substrate concentrations. Inhibitors will decrease whereas activators will increase the apparent target size of enzymes or receptors.

An amperometric biosensor based on tyrosinase, immobilized onto a carbon black paste electrode using glutaraldehyde and BSA was constructed to detect competitive inhibitors. Three inhibitors were used in this study: benzoic acid, sodium azide, and kojic acid, and the obtained values for fifty percent of inhibition (IC50) were 119 µM, 1480 µM, and 30 µM, respectively. The type of inhibition can also be determined from the curve of the degree of inhibition by considering the shift of the inhibition curves. Amperometric experiments were performed with a biosensor polarized at the potential −0.15 V vs. Ag/AgCl and using 0.1 M phosphate buffer (pH 6.8) as an electrolyte. Under optimized conditions, the proposed biosensor showed a linear amperometric response toward catechol detection from 0.5 µM to 38 µM with a detection limit of 0.35 µM (S/N = 3), and its sensitivity was 66.5 mA M−1 cm−2. Moreover, the biosensor exhibited a good storage stability. Conversely, a novel graphical plot for the determination of reversible competitive inhibition was represented for free tyrosinase. The graph consisted of plotting the half-time reaction (t1/2) as a function of the inhibitor concentration at various substrate concentrations. This innovative method relevance was demonstrated in the case of kojic acid using a colorimetric bioassay relying on tyrosinase inhibition. The results showed that the t1/2 provides an extended linear range of tyrosinase inhibitors.

The number of amalgam-covered surfaces and the occlusal area of the fillings, the concentrations of total mercury in plasma, erythrocytes and urine, the urinary excretion rate, and the absorbed daily doses estimated by two separate methods from intra-oral Hg emission were determined in 29 volunteers with a low amalgam load. The transfer ofHg from the fillings via the oral cavity and blood to urinary excretion was evaluated by multiple correla tions between these variables. In addition, the combina tion of variables most representative of the entire compartmental transfer of amalgam Hg was determined. Urinary excretion (1), Hg concentration in plasma (2) and absorbed dose (3) were most closely correlated to each other, followed by correlations with the variables of the fillings (4). Correlation coefficients were 0.75 for variables 1 vs 2 and 2 vs 3, and 0.49 for variables 3 vs 4. It was concluded that variables 1-3 best reflected the transfer of mercury from amalgam fillings throughout the organism and that they were relatively insensitive to dietary mercury. The determination of total mercury in plasma and of its urinary excretion rate appears, under practical aspects, most suitable for the investigation of Hg uptake from amalgam.

By using a standard graphical method values of Km and V may be found that are independent of the conditions and assumptions that the total substrate concentration approximates to its free concentration and that Km is much larger than the enzyme concentration. The procedure is also applicable to the determination of equilibrium binding parameters of a ligand to a macromolecule.

Accurately determining the height of the gas-guiding fracture zone in the overlying strata of the goaf is the key to find the height of the long horizontal borehole in the roof. In order to determine the height, in this study we chose the 6306 working face of Tangkou Coal Mine in China as a research example and used both the theoretical model and discrete element method (DEM) numerical simulation to find the height of the gas-guiding fracture zone and applied the height to drill a long horizontal borehole in the roof of the 6303 working face. Furthermore, the borehole was utilized to deep into the roof for coalbed methane drainage and the results were compared with conventional gas drainage measures from other aspects. The height of the gas-guiding fracture zone was found to be 48.57 m in theoretical model based on the bulk coefficient and the void ratio and to be 51.19 m in the DEM numerical simulation according to the temporal and spatial variation characteristics of porosity. Taking both the results of theoretical analysis and numerical simulation into consideration, we determined that gas-guiding fracture zone is 49.88 m high and applied it to drill a long horizontal borehole deep into the roof in the 6303 working face field. Compared with conventional gas drainage measures, we found that the long horizontal borehole has the high stability, high efficiency and strong adaptability for methane drainage.

Potassium-40 was determined in soil in an area with 40,000 m² situated in the western State of Pernambuco, Brazil. For radiometric measurements, the gamma spectrometry method with a high purity germanium (HPGe) detector was used. Sampling of 78 soil samples has been performed at intervals of 25 m. The specific activities of 40K were calculated based on the photopeak of 1.46 MeV. Values from 541 to 3,572 Bq kg-1 were obtained (mean of 1,827 Bq kg-1). These values allowed the determination of the elemental concentrations as well as the absorbed dose rates in air, 1 m above the ground. The values varied from 1.7 to 11.5% (mean of 6%) and from 23.4 to 154.3 nGy h-1 (mean of 79 nGy h-1), respectively.
