Effects of aqueous extract of neem seed kernel and azadirachtin on the fecundity, fertility and post-embryonic development of the melonfly, Bactrocera cucurbitae and the oriental fruit fly, Bactrocera dorsalis (Diptera: Tephritidae)

Since its invasion in Senegal (West Africa) in 2004, Bactrocera dorsalis Hendel, the oriental fruit fly, has caused loss of production of mango and other commercial fruits. The use of neem products (oil, cake, and seed powder) and kaolin may be an alternative to synthetic insecticides for controlling adult flies and larval-pupal stages and may contribute to integrated pest management tactics. In the laboratory, neem oil-treated fruit had the same number of landings and time spent on treated fruit as the control but showed reduced pupal development. Females selected kaolin-treated fruits for landing less often, with reduced pupal development. Soil treatment with neem cake and neem seed powder reduced pupal survival. In the field, neem treatment had a greater number of infested fruits and greater pupal development than the control, but fewer flies emerged. Kaolin was not different from the control in percent of infested fruits or pupal development, but fewer flies emerged. Neem cake and neem seed powder were found to lose efficacy in the field.

The objective of this study was to develop oral and topical toxicity data for fipronil in Solulys protein bait to wild melon fly, Bactrocera cucurbitae (Coquillett), and the oriental fruit fly, Bactrocera dorsalis (Hendel). For the oral study, both females and males were evaluated, whereas in the contact study only females were evaluated. The 24 h oral LC50 estimates for female B. cucurbitae and B. dorsalis were 113 and 108 mg ai/l, respectively. Female B. cucurbitae were more susceptible than males, but female and male B. dorsalis were equally susceptible to fipronil after the oral route of exposure. Female B. cucurbitae were significantly less susceptible to the fipronil-bait mixture after topical exposure compared with feeding exposure. However, female B. dorsalis were equally susceptible to either route of exposure. At the LC50, B. dorsalis was significantly more susceptible than B. cucurbitae by the topical route of exposure. At the LC90, B. dorsalis was significantly more susceptible than B. cucurbitae by both oral and topical routes of exposure. Results of this study indicate that there are differences In susceptibility between B. cucurbitae and B. dorsalis to fipronil, especially at the LC90. Bactrocera dorsalis was more susceptible to fipronil than B. cucurbitae by oral and topical routes of exposure. LC90 estimates were significantly lower than the 5,333 mg ai/l applied to Amulet Attract and Kill Stations for control of B. cucurbitae and B. dorsalis.

The oriental fruit fly, Bactrocera dorsalis (Hendel), is one of the most devastating and highly invasive agricultural pests world-wide, resulting in severe economic loss. Thus, it is of great interest to understand the transcriptional changes that occur during the activation of its zygotic genome at the early stages of embryonic development, especially the expression of genes involved in sex determination and the cellularization processes. In this study, we applied Illumina sequencing to identify B. dorsalis sex determination genes and early zygotic genes by analyzing transcripts from three early embryonic stages at 0–1, 2–4, and 5–8 h post-oviposition, which include the initiation of sex determination and cellularization. These tests generated 13,489 unigenes with an average length of 2185 bp. In total, 1683, 3201 and 3134 unigenes had significant changes in expression levels at times after oviposition including at 2–4 h versus 0–1 h, 5–8 h versus 0–1 h, and 5–8 h versus 2–4 h, respectively. Clusters of gene orthology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) annotations were performed throughout embryonic development to better understand the functions of differentially expressed unigenes. We observed that the RNA binding and spliceosome pathways were highly enriched and overrepresented during the early stage of embryogenesis. Additionally, transcripts for 21 sex-determination and three cellularization genes were identified, and expression pattern analysis revealed that the majority of these genes were highly expressed during embryogenesis. This study is the first assembly performed for B. dorsalis based on Illumina next-generation sequencing technology during embryogenesis. Our data should contribute significantly to the fundamental understanding of sex determination and early embryogenesis in tephritid fruit flies, and provide gene promoter and effector gene candidates for transgenic pest-management strategies for these economically important species.
