Lactic acid bacteria are groups of bacteria that have been commonly used as probiotics with functional targets controlling pathogens. In this study, BAL were isolated from penaeid shrimp intestinum was captured in natural waters and were tested of antibacterial activity against 3 types of pathogenic vibrio bacteria in shrimp. The shrimp samples were taken from the North Coast of Java (Situbondo and Semarang) and the South coast of Java (Cilacap). BAL isolation was done by pour plate method using MRS medium enriched with CaCO3. The antibacterial activity test was performed by using paper diffusion method using V. harveyi, V. anguilarum and V. parahaemolyticus as bacteria indicator. The isolation result obtained 224 isolates of BAL intestinal of penaeid shrimp. The active BAL isolates against V. Harveyi, V. anguilarum and V. parahaemolyticus were 59, 83 and 66 isolates, respectively. Isolates are active against 3 types, 2 types and 1 type of Vibrio were 51, 25 and 11 isolates respectively.Keyword : lactic acid bacteria, antibacteria, vibrio, penaeid shrimp Bakteri asam laktat merupakan kelompok bakteri yang telah banyak digunakan sebagai probiotik dengan target fungsional mengendalikan pathogen. Pada penelitian ini telah dilakukan isolasi BAL dari intestinum udang penaeid yang ditangkap di perairan alam dan pengujian aktivitas antibakteri terhadap 3 jenis bakteri vibrio pathogenik pada udang. Sampel udang diambil dari Pantai Utara Jawa (Situbondo dan Semarang) dan pantai Selatan Jawa (Cilacap). Isolasi BAL dilakukan dengan metode taburan menggunakan medium MRS yang diperkaya dengan CaCO3. Uji aktivitas antibakteri dilakukan dengan paper diffusion method menggunakan bakteri uji V. harveyi, V. anguilarum dan V. parahaemolyticus. Hasil isolasi diperoleh 224 isolat BAL intestinal udang penaeid. Isolat BAL yang aktif terhadap V. Harveyi, V. anguilarum dan V. parahaemolyticus berturut-turut 59, 83 dan 66 isolat. Isolat yang aktif terhadap 3 jenis, 2 jenis dan 1 jenis Vibrio uji berurut-turut 51, 25 dan 11 isolat. Kata Kunci : bakteri asam laktat, antibakteri, vibrio, udang penaeid

Abstract Alginate polysaccharide/oligosaccharide (APS/AOS) has been proven as a good immunostimulant. FNCC–0041 & FNCC–0040 are lactic acid bacteria (LAB) producing exopolysaccharides. Vibrio bacteria are pathogenic for aquatic cultivans. This study determines the performance of APS/AOS in combination with LAB as immunostimulants. These were conducted in Artemia challenge test assay against three species of Vibrio spp., namely Vibrio parahaemolyticus (Vp), V. harveyi (Vh), and V. vulnificus (Vv). The treatments were prepared by Factorial Design with two factors (APS/AOS doses and Vibrio spp.) and replicated three times. The APS/AOS concentration was 0, 300, 600, 900 ppm. There were 8 levels of vibrio challenges, namely non-Vibrio, Vp, Vh, Vv, Vp-Vh, Vh-Vp, Vv-Vp, and Vp-Vh-Vv. LAB and APS/AOS were fermented with seawater encapsulated by newly hatched Artemia’s nauplii for one hour. Ten nauplii were taken out and challenged with 108 cells/mL Vibrio. Its survival rate (SR) was counted every six hours until reached 100% mortality. Results show that SR of all nauplii Artemia bio encapsulated treatments was higher than control (p<0.05). The best survival rate was reached from 400 ppm AOS. It has appeared that there is a synergically positive effect among the bio encapsulated AOS and LAB to accelerate the Artemia’s immune system.

ABSTRACT In the present work, the effects of several lactic acid bacteria on the immune response of turbot (Scophthalmus maximus) macrophages have been studied both in vitro and in vivo. Out of six lactic acid bacterial strains tested, only heat-killed Lactococcus lactis significantly increased the turbot head kidney macrophage chemiluminescent (CL) response after 24 h of incubation. Nitric oxide (NO) was also significantly enhanced by this bacterium after 72 h of incubation with either viable (103 and 106 cells/ml) or heat-killed (106 cells/ml) bacteria. Viable Leuconostoc mesenteroides (106 cells/ml) was also capable of significantly increasing NO production. Since L. lactis proved to be the strain with more effects on the host immune function, further in vivo and in vitro experiments were conducted with this bacterium. The in vitro capacity of L. lactis to adhere to turbot intestinal mucus was positively confirmed. When orally administered, L. lactis significantly increased the macrophage CL response and the serum NO concentration after 7 days of daily administration. The antibacterial effect of the extracellular products from the six LAB strains against the fish-pathogenic bacterium Vibrio anguillarum was also demonstrated in vitro.
